        Background : The root of Platycodin grandiflorum has been widely used as a crude drug or a dependent on the cultivation area and cultivation method. Therefore, this study was conducted to know the effect of organic fertilizer on the changes of antioxidents and antioxidant activity of Platycodin grandiflorum Ridix. Methods and Results : As basal fertilizer were treated before transplanting Platycodin grandiflorum Ridix with chemical fertilizer(Super21, 21-17-17, Namhae Chemical), mixed organic fertilizer, bacterial culture, fermented oil cake and manure in each trial of nitrogen based on 3㎏/10a criteria. The total polyphenol content was highest values were 15.5mg/g appear in the mixed organic fertilizers, the lowest were 9.1mg/g appeared in the manure and control. Flavonoid content showed the highest 15.3 mg/g in the mixed organic fertilizer, and investigated at the lowest 5.0 mg/g in the fermented oil cake. ABTS radical scavenging activity was the highest in was 65.7 mgTE/g in the chemical fertilizer, showed the lowest value in the control to 13.0 mgTE/g. Therefore, both flavonoid and polyphenol content showed a high value on organic fertilizers. Conclusion : Therefore, both flavonoid and polyphenol content showed a high value on organic fertilizers. It is determined that to have researches for a variety of antioxidants by the organic fertilizers made in the future.

        Background : Ginsenosides, the main ingredient of ginseng roots can be confirmed various physiological activity such as anticancer, antioxidant, a natural ginsenosides is there a structure to be absorbed into the body does not work well absorbed through this process biologically active thus a high conversion ginsenosides. β-glucosidase enzyme is observed in several of the microorganism with an enzyme that serves to convert a ginsenoside prosper that is absorbed into the body. Methods and Results : To view a primary β-glucosidase activity, the bacteria were innoculated in esculin agar medium and the color change of the media were measured by the time and degree of changing color. In the other method, 5 mM of p-nitrophenyl-β-D-glucopyranoside (pNPG) containing 25 mM phosphate buffer solution (pH 7.0) was added to 50 ul enzyme solution. Then the solution was added to 50 ul reaction for 5 min at 30°C. The amount of p-nitrophenol liberated measured at 405 nm absorbance. The experimental results showed higher β-glucosidase activity in Pediococcus pentosaceus, Leuconostoc mesenteroide, Leuconostoc mesenteroides subsp. cremoris, and Paenibacillus polymyxa by using esculin agar medium method. Similarly in second method, β-glucosidase activity was higher in P. pentosaceus 402.32±11.43 unit/l, L. mesenteroide 353.73±14.64 unit/l, Lactobacillus sakei 198.4±15.47 unit/l Lactobacillus plantarum subsp. plantarum 164.1±8.12 unit/l. Conclusion : The result that the β-glucosidase activity was higher in P. pentosaceus, L. mesenteroide, and L. plantarum subsp. plantarum as compared to tested microbes. Therefore selected bacteria can be used in the industry of functioned foods and beverage to improve human healths.

        최근 인공지반녹화가 주변 생활공간의 쾌적화, 에너지 절약효과, 도시 미관의 환경개선 등의 장점을 갖고 있어 콘크리트 위에 녹화 공간 조성이 증가하고 있다. 이에 따라 건축공사 표준 시방서, 건축물 녹화 설계기준 등에서는 방수층의 수밀안정성 확보를 위해 일정 품질 이 상의 방근층 설치를 규정하고 있으나, 실제 우리나라의 많은 현장은 방수층 위에 누름콘크리트 타설 후 인공녹화를 시행하고 있는 실정이다. 이에 본 연구에서는 방근층을 시공하지 않은 현장을 대상으로 지표를 설정하였으며, 누름콘크리트의 균열에 따른 식물의 뿌리가 균열부로 침 입 및 관통하여 누름콘크리트 하단부의 아스팔트 방수시트에 미치는 영향을 평가하였다.

        Small-sized tuber of sweet potato (Ipomoea batatas Lam.) is more preferred by Korean consumers, because it is convenient to eat with hands, and can easily be steamed or roasted in small pan as a healthy snack for a small family. The present study was aimed to investigate the effect of highdensity plantings and harvesting times on production of smallsized tuber (50~200 g). Four varieties (‘Annobeny’, ‘Daeyumi’, ‘Shinzami’, and ‘Yeonhwangmi’) were planted in 2-row zigzag high-density planting (75×30 cm, A; 75×25 cm, B) and 1-row planting (75×25 cm, control) covered with black vinyl film on May 23th, 2014. Marketable yields and small-sized tuber yields were compared between 120- and 150-day harvesting. Vine length and fresh weight per plant were significantly decreased in 2-row planting A and B plots compared to the control at 120 days after planting. Weight of tuber per plant, mean weight of tuber and number of tuber per plant were significantly decreased in 120-day harvesting of A and B plots than in the control. Marketable yields per 10a were increased by 17% (2.4 ton‧10a -1 ) and 8% (2.6 ton‧10a -1 ) in 120-day harvesting of A and B plots compared to the control (2.4 ton‧10a -1 ), and those were largely increased by 29% (4.0 ton‧10a -1 ) and 26% (3.9 ton‧10a -1 ) in 150-day harvesting than in the control (3.1 ton‧10a -1 ). Proportions of small size tuber (50~200 g) were increased by about 65% in 150-day harvesting of A and B plots compared to the control (55.3%). Small-size tuber yield and marketable yield were highly increased in 150-day harvesting of 2-row zigzag high-density planting A (75×30 cm).

        The roots of Platycodon grandiflorum species either dried or fresh, are used as an ingredient in salads and traditional cuisine in Korea. To interpret the root proteins, a systematical and targeting analysis were carried out from diploid and tetraploid roots. Two dimensional gels stained with CBB, a total of 39 differential expressed proteins were identified from the diploid root under in vivo condition using image analysis by Progenesis Same Spot software. Out of total differential expressed spots, 39 differential expressed protein spots (≥ 1.5-fold) were analyzed using LTQ-FTICR mass spectrometry. Except two proteins, the rest of the identified proteins were confirmed as down-regulated such as Isocitrate dehydrogenase, Proteasome subunit alpha type-2-B. However, the most of the identified proteins from the explants were mainly associated with the oxidoreductase activity, nucleic acid binding, transferase activity and catalytic activity. The exclusive protein profile may provide insight clues for better understanding the characteristics of proteins and metabolic activity in various explants of the economically important medicinal plant Platycodon grandiflorum.

        Panax ginseng C.A. meyer (family: Araliaceae) is a perennial crop that has been widely used as a traditional medicine in Korea. Various P. ginseng cultivars exhibit a range of morphological and physiological traits as well as genetic diversity. To elucidate the differences of primary metabolism underlying such genetic diverstiy, we performed primary metabolite profiles in adventitious roots from five Panax ginseng cultivars using gas chromatography-mass spectrometry (GC-MS). The GC-MS analysis revealed eight primary metabolites as biomarkers and allowed us to classify the five cultivars into three groups. We selected three cultivars to represent each group and analyzed their transcriptomes by Illumina sequencing. We inspected 100 unigenes involved in seven primary metabolite biosynthesis pathways and found that 21 unigenes encoding 15 enzymes were differentially expressed among the three cultivars. Integrated analysis of transcriptomes and metabolomes revealed that the ginseng cultivars differ in primary metabolites as well as in the putative genes involved in the complex process of primary metabolic pathways. Our data derived from this integrated analysis provide insights into the underlying complexity of genes and metabolites that co-regulate flux through these pathways in ginseng.

        The roots of Platycodon grandiflorum are known as traditional medicine, has been extensively used since ancient times as a therapeutic to treat cold, cough and asthma in Korean traditional medications. This study was conducted in order to profile proteins from the hormone induced diploid and tetraploid roots using high throughput proteome approach. Two dimensional gels stained with CBB, a total of 64 differential expressed proteins were identified from the diploid root using image analysis by Progenesis SameSpot software. Out of total differential expressed spots, 20 differential expressed protein spots (≥ 2-fold) were analyzed using MALDI-TOF-TOF mass spectrometry whereas a total of 13 protein spots were up regulated and 7 protein spots were down-regulated. However, in the case of tetraploid root, a total of 78 differential expressed proteins were identified from tetraploid root of which a total of 28 differential expressed protein spots (≥ 2-fold) were analyzed by mass spectrometry whereas a total of 16 protein spots were up regulated and a total of 12 protein spots were down-regulated. However, proteins identified using iProClass databases revealed that the identified proteins from the explants were mainly associated with the nucleic acid binding, oxidoreductase activity, transporter activity and isomers activity. The exclusive protein profile may provide insight clues for better understanding the characteristics of proteins and metabolic activity in various explants of the economically important medicinal plant Platycodon grandiflorum.

        Panax Ginseng is a perennial medicinal plant originated from North-east asia. Because of its well-known tonic effects mainly from ginsenosides, various types of processed ginseng products have been distributed around the world. Here, we analyzed secondary metabolite profiling of adventitious roots of 5 korean ginseng cultivars, Chunpoong (CP), Sunhyang (SH), Gopoong (GO), Sunun (SU), and Cheongsun (CS). At the same time, the profiles of relative gene expressions related to ginsenoside biosynthesis pathway were compared among ginseng cultivars. Secondary metabolite profiles were revealed by UPLC/Q-TOF-MS from extracts of bioreactor derived adventitious roots of five ginseng cultivars. Using principal component analysis, secondary metabolite profiles of ginseng cultivars were categorized into three groups. Metabolites with high VIP values were annotated using known database and standards compounds. Relative gene expression of ginsenoside related gene were analyzed using realtime PCR. The three groups had distinct metabolite contents. Furthermore, gene expression profiles related to ginsenoside were also different, which might contribute diverse secondary metabolite composition of ginseng cultivars. Further integrated analysis would provide a relationship between genetic background of ginseng cultivars and secondary metabolite profiles.

        This study was performed to identify the effect of mixed bed soil on growth of aerial parts and root zone of daughter plants for nursery field strawberry seedling raising with expanded chaff. The plant height and leaf area of daughter plants were highest or largest in the mixed soil of ERH +RH (100:0, v/v), followed by ERH+RH (75:25). The higher the mixing ratio of RH, the shorter the plant height or the smaller the leaf area. A similar tendency was observed in fresh weight. Within a root diameter of 0-0.4 mm and a root height range of 0.4-0.8 mm, root surface area and volume were statistically significantly better with treatment of ERH+RH (100:0, v/v) compared to those of roots treated with ERH+RH (75:25), ERH+RH (50:50) and ERH+RH (25:75). The growth rate of aerial parts and root zone of daughter plants were noticeably lower in two mixing ratios of 50:50 and 25:75. According to the mixing ratios of ERH+CD surface treatment, the number of roots was greatest in plants treated with ERH+CD (80:20, v/v) and ERH+CD (85:15) on August 1. However, the number of roots was highest in plants treated with ERH+CD (85:15, v/v) on August 15. Root length was longest in the plant with no treatment, and drastically shortened from ERH+CD (90:10, v/v) in both surface and mixed treatment. Although root weight showed a significant difference in ERH+CD (90:10, v/v) treatment, its increase was gradual. The rate of root growth was highest in ERH+CD (85:15). These study findings suggest that the content ratios of mixed soil ERH+RH (75:25, v/v) or below and ERH+CD (85:15) are thought to be desirable for the production of high quality seedlings.

        Rice is a staple food for over one-half of the world population, especially in Asian countries. Recently, the growth and yield of crop plants was affected by various abiotic stresses, such as salt, drought, and high temperature due to change of climate environment. To study molecular functions of Oryza sativa nuclear-targeted RING Finger Proteins (OsNRFPs) in response to abiotic stresses, we selected 44 OsNRFPs genes, whose subcelluar localizations are predicted to the nucluear, on the basis of expression patterns of a microarray dataset. A total of 44 OsNRFPs were grouped into two types such as RING-HC and RING-H2 via phylogeneitc analysis of their RING domains structures. Subsequently, we surveyed the expression patterns of 44 genes in response to salt stress via qRT-PCR in roots. We found 10 salt stress-induced OsNRFPs and then examined their subcellular localizations. These genes were clearly localized to the nucleus (OsNRFPHC-10), cytoplasm (OsNRFPHC-17 and OsNRFPH2-16) and microtuble (OsNRFPHC-23, OsNRFPH2-17 and OsNRFPH2-05), respectively. These results might provide a key clue for understanding moleuclar functions of the OsNRFP genes associated with salt stress-related signaling pathway

        Platycodon grandiflorum is a perennial flowering plant, known as Chinese bell flower, widespread in northeast Asia. The roots of this species are used for centuries to treat diseases, and have extensive pharmacological effects such as reducing adiposity, hyperlipidemia as well as anti-atherosclerotic disorder. In this study, systematical and targeting proteome analysis were executed from the 3, 4 and 5 months aged diploid and tetraploid roots of Platycodon grandiflorum and the proteins were separated by 2-DE and stained by CBB. In diploid roots, a total of 30 protein spots (≥ 2-fold) were analyzed using MASCOT search engine according to the similarity of sequences with previously characterized proteins along with the UniProt database. Among the 30 differentially expressed proteins, 21 proteins sopts were identified as up-regulated and 9 proteins were identified as down-regulated. In contrary, a total of 40 differentially expressed proteins were confirmed from tetraploid roots whereas 28 protein spots were confirmed as up-regulated and 12 proteins were identified as down-regulated. However, the differentially expressed proteins from diploid and tetraploid roots were classified into 12 and 14 possible functional categories respectively using Protein Information Resources. The results revealed that the identified proteins from diploid and tetraploid roots were mainly involved in oxidoreductase activity, nucleotide binding, transferase activity and catalytic activity in bellflower roots. In conclusion, the exclusive proteins from diploid and tetraploid roots may provide insight clues for better understanding the characteristics and functions of proteins and metabolic activity of Platycodon grandiflorum.

        Platycodon grandiflorum, known as Doraji in Korea, is used in various medications and traditional cuisine in Korea. This study was conducted to characterize the hormonal effects of diploid and tetraploid roots of P. grandiflorum using proteomics technique. Prior to proteome analysis, different kinds of growth hormones; IBA (1mg/L), NAA (1mg/L) and IAA (1mg/L) were applied in the adventitious (Diploid and tetraploid) roots for investigation. Solid (1/4MS) and liquid (1/2MS) medium were performed in the present study to investigate the hormonal effects. In diploid roots, two dimensional gels stained with CBB, a total of 1154 protein spots were identified using image analysis by Ludesi REDFIN 3 programme (Ludesi AB, Lund, Sweden: www.ludesi.com). Out of 1154 differential expressed protein spots, a total of 33 protein spots (≥ 2-fold) were selected for mass spectrometry. Among the 33 protein spots, 7 protein spots were up-regulated in IBA, 12 proteins in NAA and 14 proteins in IAA. In the case of tetraploid roots that performed under solid medium, a total of 842 differentially expressed protein spots were identified of which 34 proteins spots (≥ 1.5-fold) were selected for mass spectrometry. Out of 34 protein spots, 11 proteins were up-regulated in IBA, 10 proteins in NAA and 13proteins in IAA. However, a total of 659 differentially expressed proteins were confirmed from the liquid medium of tetraploid roots from which 32 proteins spots (≥ 1.5-fold) were sorted for MS analysis. Out of these 32 proteins, a total of 3 proteins were up-regulated in IBA, 7 proteins in NAA and 22 proteins in IAA. The identified proteins may provide insight clues for better understanding of the characteristics of proteins and biological activity from adventitious roots of Platycodon grandiflorum.
