        Xylitol is a sugar alcohol with a variety of functions including bactericidal and anticariogenic effects. However, the cellular mechanisms underlying the role of xylitol in bone metabolism are not yet clarified. In our present study, we exploited the physiological role of xylitol on osteoclast dif-ferentiation in a co-culture system of osteoblastic and RAW 264.7 cells. Xylitol treatment of these co-cultures reduced the number of tartrate-resistant acid phosphatase (TRAP)- positive multinucleated cells induced by 10 nM 1α,25(OH)2 D3 in a dose‐dependent manner. A cell viability test revea-led no marked cellular damage by up to 100 mM of xylitol. Exposure of osteoblastic cells to xylitol decreased RANKL, but not OPG, mRNA expression in the presence of 10-8 M 1α,25(OH)2D3 in a dose‐dependent manner. Furthermore, bone resorption activity, assessed on bone slices in the co- culture system, was found to be dramatically decreased with increasing xylitol concentrations. RANKL and OPG proteins were assayed by ELISA and the soluble RANKL (sRANKL) concentration was decreased with an increased xylitol con-centration. In contrast, OPG was unaltered by any xylitol con-centration in this assay. These results indicate that xylitol inhibits 1α,25(OH)2D3-induced osteoclastogenesis by reducing the sRANKL/OPG expression ratio in osteoblastic cells.

        The purpose of this study was to estimate the consumption prevalence of vitamin and mineral supplements as health functional foods (VM-HFF) and to examine the factors associated with VM-HFF consumption behaviors in adolescents. A total of 1,407 adolescents attending middle or high schools from various cities and rural communities in Korea participated in this study. The prevalence of VM-HFF consumption was 41.7%, with boys showing a higher consumption than girls (p<0.01). VM-HFF consumption was higher in families with higher socioeconomic status and for families with parents that exhibited higher concerns about their child's health, growth, and nutritional intake (p<0.001). Most consumers of VM-HFF consumed HFFs 'when healthy' (50.1%), and acquired nutritional information from 'their families & relatives' (50.9%). Most consumers responded that VM-HFF was 'a little effective' (54.3%), followed by 'no obvious effects' (37.1%), and 'very effective' (7.4%). The effectiveness of consuming VM-HFF was mainly for 'fatigue recovery' (39.0%) and 'health improvement' (28.2%). Most consumers purchased HFFs at 'pharmacies & oriental medicine clinics' (53.8%) and at 'health functional food stores' (18.8%). Most consumers 'occasionally' (51.1%) or 'seldom' (27.3%) checked nutrition facts when purchasing, with 58.9% of consumers understanding the nutritional label for 'the most part', but only 8.7% of them understanding it 'very well'. Among the VM-HFF, consumers preferred calcium- and vitamin C-supplements. Consumers' mini-dietary assessment scores were higher than those of non-consumers. The results above showed that VM-HFF consumption was widely spread among adolescents, but few consumers checked and understood the nutrition label when they purchased VM-HFF, and were highly dependent on the advice and information from non-professional nutritionists, such as families & relatives. Therefore, it is necessary to educate adolescents to help them read nutrition labels and select the proper VM-HFF.

        경상북도, 성주 지역에서 2009년 6월에서 8월 사이 매달 참외를 수확하여 수확시기의 기상환경이 참외의 수용성 비타민 함량 및 품질에 미치는 영향을 평가하고자 하였다. 과실 품질평가를 조사하기 위하여 무게, 경도 그리고 당도 등의 특성을 조사하였고 수용성 비타민인 엽산과 비타민 C의 함량을 측정하였다. 수확시기에 따른 과실특성을 조사한 결과 강우량이 적고 일사량이 많은 6월에 수확한 과실이 무게, 경도 및 당도가 가장 높게 나타났고, 강우량이 많은 7월에 급격히 감소하는 경향을 보여주었다. 8월에 일사량이 증가하고 강우량이 감소하면서 다시 증가하는 것을 확인할 수 있었다. 참외의 대표적인 비타민인 엽산과 비타민 C의 함량을 측정한 결과, 6월에 수확한 참외에서 가장 높은 함량을 보여주었고, 7월에 현저히 감소하였다. 과실의 특성 조사 결과와 마찬가지로 엽산과 비타민 C의 함량은 강우량이 감소하는 8월에 다시 증가하는 경향을 보여주었다. 부위별로 분석해 본 결과 태좌 부위에 다량의 엽산과 비타민 C가 함유되어 있음을 알 수 있었다. 이상의 연구결과는 강우량 증가에 의한 수분장애 및 일사량 부족은 참외 과실의 품질뿐만 아니라 수용성 비타민 함량에도 영향을 미치고 있고, 특히 엽산 함량을 크게 감소시키는 것으로 나타났다.

        Effects of roasting on vitamin E content, color, microstructure and moisture of peanuts, and vitamin E content in peanut oils prepared from the roasted peanuts were investigated. Runner-type peanuts were roasted at 140, 150, and 160ºC for 10-20 min. As roasting temperature and time increased, the CIELAB L* value of peanuts decreased while a* and b* values increased, resulting in formation of the golden brown color of roasted peanuts. Moisture ratio (M/Mo) and color b* value of peanuts roasted at 140 to 160ºC showed a correlation of b* = 21.61 (M/Mo)2 – 40.62 (M/Mo) + 34.12 (R2 = 0.9123). Overall changes in the tocopherol contents of peanuts and peanut oils were significantly affected by roasting temperature and time (p<0.05). Roasting at 140ºC caused a slight increase in the levels of tocopherols of peanuts over roasting time up to 20 min (p<0.05). There was no significant change in the tocopherol levels of peanuts during roasting at 150 ºC for 20 min (p>0.05). At 160ºC, the levels of tocopherols significantly decreased during the initial 10 min of roasting (p<0.05) while there was no extended loss after 10 min, resulting in about 5, 12, 20, and 10% losses of α-, β-, γ- and δ-T, respectively. After 20 min, total tocopherols decreased by 18%. However, tocopherol contents of pressed peanut oils significantly decreased at all roasting temperatures (p<0.05). After roasting peanuts at 160ºC for 20 min, about 84% of initial α-T in peanut oils was retained. α-T was the most stable to roasting while γ-T was the least. Swollen epidermal cells on the inner surface and broken cell walls of parenchyma tissue of peanut cotyledon were observed in peanuts after roasting at 160ºC for 15 min. Severe changes in microstructure of peanut by roasting would contribute to vitamin E stability because of exposure of oil droplets in peanuts to oxygen.

        This study was carried out to assess the effect of vitamin E against the reactive oxygen species (ROS) on chemical activation of in vitro matured oocytes. Bovine oocytes were aspirated from slaughtered ovaries and transferred to maturation medium with or without vitamin E (). After 22 hours of culture, oocytes with polar bodies were selected and submitted to activation treatments with or without vitamin E. After activation, oocytes were cultured in mSOF medium and rate of development was monitored. For ROS () detection, in vitro matured and activated oocytes were selected and stained with DCFDA and observed under fluorescence microscope. The ROS contents were not significant differences in IVM rate, activation process and embryonic development to blastocysts with or without vitamin E. The cell number of blastocyst showed significant difference (p<0.05) in embryos matured and activated with vitamin E. The results of the present study demonstrated that the exposure of vitamin E in IVM and activation process improved the quality of embryos evaluated by the cell number of blastocysts.

        Taking vitamin and mineral supplements is increasingly common with the rapid economic growth. The aim of this study was to determine the prevalence of vitamin and mineral supplement use among adults aged 20 or older from the third Korean National Health and Nutrition Survey data and to explore the effect of sociodemographic and lifestyle factors as well as nutrient intake on taking supplements. People who had participated in both a health questionnaire and a nutritional survey were selected, and 2,871 men and 3,555 women were finally included in this analysis. Both men and women with a higher level of education, those residing in a metropolitan area, and those with higher income were more likely to take supplements. Health behaviors were not significantly associated with taking supplements. Mean nutrient intake of all nutrients except energy intake was not significantly different in men or women taking supplements after adjusting for age, education, marital status, resident area, smoking, and energy intake. Compared to Dietary Reference Intake (DRI) for Koreans, most vitamins and minerals, except vitamin B2 and calcium, were consumed at higher than the Recommended Intake (RI) without supplements. In conclusion, taking supplements such as vitamin B2 and calcium may promote health and prevent disease. However, the type and frequency of other vitamin and mineral supplements consumed should be considered with caution.

        We surveyed 821 children (ages 12-15 y) in Daejeon city and Cunngcheong province, Korea for their use of vitamin·mineral supplements (VMS), either over-the-counter drugs and/or health functional foods (HFF). We also considered the significant variables of VMS use, including demographic characteristics, health-related variables, eating habits and nutritional beliefs of VMS by subjects. The frequency of VMS use was 24.8% and, in comparison to counterparts, it was higher in subjects attending middle school (p<0.001), in those living in a large city (p<0.001), in those with a high monthly family income (p<0.05), and in those with high concerns about school performance by parents (p<0.01). Selfhealth concerns by subjects (p<0.05) and concerns about health of offspring by parents (p<0.01) were higher in users than in nonusers. Eating habits of subjects did not affect the use of VMS. Users had a more positive view concerning the potential health benefits of VMS than did nonusers (p<0.05). Most users took VMS when they were healthy (78.9%) and expected ‘disease prevention and health maintenance’ (44.6%). However, most users did not feel any special effects from VMS use (58.8%), and they got their information on VMS through family and relatives (53.9%). Users of VMS preferred vitamin C supplements (44.2%) and Ca-supplements (56.9%). Therefore, the results show that VMS use is widely spread among the subjects, and that use of VMS is influenced by multiple variables and subjective criteria. Therefore, practical guidelines on VMS use for children’s health and growth should be given through nutritional education.

        This study was conducted to determine the effects of vitamin C on the activity of liver function enzymes and electromicrographic changes in white rats treated with aflatoxin B1(AFB1) or X-ray and AFB1. Six week-old male Sprague-Dawley rats were randomly divided into five groups: a control group, AFB1 treated group, AFB1 treated group with vitamin C, X-ray and AFB1 co-treated group, X-ray and AFB1 co-treated group with vitamin C. On the first day of the experiment, only one dose of X-rays was exposed to the entire liver at 1, 500 cGy. Next, vitamin C was injected at 10㎎/㎏body weight by intraperitoneal injection, followed 1 hr later by the administration of 0.4㎎/㎏of AFB1 by intraperitoneal injection. These treatments were then administered every three days over a period of 15 days. On the 16th day of treatments, the animals were sacrificed. Analysis of the activity of the liver function enzymes, GOT, ALK phatase and LDH, in the sera of rats revealed that they were somewhat increased by AFB1 treatment, X-ray and AFB1 co-treatment when compared to the control group. Furthermore, the activity of these enzymes decreased in response to administration of vitamin C. Especially, the levels of GOT were remarkably decreased in the AFB1 treated group treated with vitamin C when compared to the group treated with AFB1 alone(p<0.001). Electromicrographic analysis revealed cloudy swelling, necrosis, vesicular degeneration and fat accumulation of hepatocytes in response to treatment with AFB1 or co-treatment with X-ray and AFB1. However, the destruction of hepatic cells was considerably lower in the vitamin C-treated group. These results indicate that vitamin C had ameliorating effects on the hepatic cell damage.

        The increase in the total number of cows on farms, which breed Korean Native Cattle (KNC), is associated with many problems. In particular, the services per conception and calving interval have increased. In this study, we examined the effect of dietary supplementation with vitamin and mineral complex on the estrus rate, pregnancy rate, gestation length, and birth weight of KNC calves. Multiparous or primiparous KNC were divided into 3 groups with 40 heads per group. Experimental group 1 was administered a consisting of 35,000 IU vitamin A, 100 IU vitamin E, 200 mg -carotene, 200 mg Zn methionine, and 1.5 mg Se. Experimental group 2 was administered a vitamin complex (100 g/day) consisting of 100 IU vitamin E and 200 mg -carotene. In the case of multiparous KNC, the estrus detection rate in the control group was 90.0% and those in the experimental groups were 75.0% to 95.0%. However, the first-service pregnancy rate after parturition in the control group was 41.2%, which was significantly lower than that in the experimental groups (71.0% to 76.7%; p<0.05). The average duration of pregnancy in the group supplemented with the vitamin complex was days, which was similar to that in the case of the control group. The birth weight of calves from cows fed with vitamin complex was 25.3 to 27.0 kg, which was similar to that in the case of the control group (25.2 to 26.0 kg). In the case of primiparous KNC, no differences in the estrus rate, pregnancy rate, gestation length, or the birth weight of calves were noted between the groups. Thus, dietary supplementation vitamin and mineral complex have no effect on the reproductive efficiency in primiparous cows, but the pregnancy rate was observed to have increased in multiparous KNC with these supplements.

        돌나물 수집종간 생육특성과 비타민 C함량 및 항산화활성을 비교하기 위하여, 13 지역종을 포장에 삽목재배하였다. 금산, 부천, 용인, 포천 지역종은 줄기가 가늘고 초장이 길며, 잎은 좁고 긴 타원형으로 다른 지역종들과 형태적 차이를 보였으며, 완주와 군산 지역 종은 초장과 절간장이 짧고, 개화기에도 꽃이 피지 않았다. 비타민 C 함량은 407.8-434.4mg·kg-1의 범위로 광양, 부안, 완주, 군산 지역종에서 높았으나, 수집종간 유의한 차이는 보이지 않았다. 항산화 활성을 나타내는 전자공여능(EDA)은 생체추출물(45.0~66.5%)에서보다 동결건조 추출물(61.3~66.5%)에서 높았으며, 광양, 순천, 완도 지역종들이 다른 지역종에 비하여 높은 항산화 활성을 나타내었다. 비타민 C함량 및 항산화 활성은 광양 지역종에서 가장 높았다.

        본 연구는 저장전 비타민 C 처리가 엔디브, 청로레인, 청경채, 적로메인, 롤롤로사, 쌈추의 베이비채소의 저장성에 미치는 영향을 알아보고자 수행하였다. 비타민 C 처리는 수확 전 3차례 실시한 엽면살포와 수확한 베이비 채소의 세척과정에서 5분간 침지하는 두 가지 방법으로 실시하였다. 비타민 세척과 재배중 엽면살포 처리 후 6가지 베이비채소의 비타민 C 함량은 저장전에는 모두 비타민 C 처리구에서 대조구보다 높았으나 저장후 일정한 경향이 없어 모두 저장 전에 비해 감소하였다. 생체중 변화의 경우 6가지 베이비 채소 모두에서 처리간 큰 차이는 없었으나 쌈추와 롤롤로사의 경우 대조구에 비해 비타민 처리구의 생체중 감소가 다소 적었고, 외관상 품질의 경우 비타민 C 침지 처리구에서 일부 효과가 보였으나 대조구와 통계적 유의성은 없었다. 포장내 이산화탄소와 산소농도는 비타민 처리로 인한 큰 변화는 없었으며, 에틸렌 농도 역시 처리간 차이를 나타내지 않았다. 따라서 베이비 채소 재배 중 혹은 세척 중 실시한 비타민 C 처리는 베이비 채소의 기능성의 향상의 효과는 있었으나 저장성 향상에는 큰 영향을 주지 못하였다.

        The purpose of this study was to examine the effects of vi tamin D3 and 1'etinoic acid(RA) on the human mesenchymal stem ce!ls(MSC) g1'owth and osteogenic differentiations. Cell proliferation, mineralization, cell cycle, expression of cell cycle regu l atOJγ proteins and markers fo1' osteogenic differenatiaiton were determined by MTI assay, mineralization assay, flow cytomet1'Y‘ and Western blot analysis, respectively. Cell viability was dec1'ease by each vitamin D3 and RA added to MSC. it was more decrease by vitamin D3 and RA. Mineralized nodule formation revealed similar expression pattern with positive cont rol group at vitamin D3 and RA mixed add to MSC. At vitamin D3 and RA mixed add to MSC after 7 days of incubation was increase G1 s tage. after 21 days of incubation was inhibit cell cycle prog1'ess by inc1'ease of sub-G1 Treatment vitamin D3 to MSC inhibits p53 and p21, but inc1'ease pRb. RA inhibit p53, but increase p21 and pRb, vitamin D3 plus RA group was same as added RA group. so two vitamin was effect to inhibited cell growth each different mechanism. Expression of BMP-2 protein was prominent in osteogonic supplement treated g1'oup of MSC at 2 weeks cultivation days, but vi tamin D3 treatment decreased BMP-2 expression rather than in (+) control group. BSP protein was notably increased in the OS compa red to positive controls at 2 weeks cultivation, but similar to that of vitamin D3 group t1'eatment group and was least expressed in plus RA mixed group, at 3 weeks, BSP expression was similar to 1'esult of 2 weeks Collectively, these results shows that vitamin D3 and RA have diffe1'ential effects on the MSCs g1'owth and differ entia tion 211

        본 연구에서는 수컷 랫드에 DEHP를 투여하여 실험적으로 생식 독성을 유발하고, vitamin E와 catechin을 단독 및 병용 투여하여 수컷 랫드에서 정소의 조직학적 변화, 정액 특성의 변화 및 정자의 운동성 변화 등을 조사하여 그 예방 효과를 알아보고자 하였다. DEHP를 투여한 군은 대조군에 비해 정자농도, 정자 생존율, 정상 정자율이 감소하였다. DEHP에 의한 생식 독성을 예방하기 위해 vitamin E와 catechin을 단독 또는 병행

        Various aspects of antioxidant activity in vitamin C were evaluated in this study. Relatively high level of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was detected in vitamin C, but not in non-antioxidative vitamin, vitamin B1. Vitamin C also reduced the production of lipid peroxidation in Chinese hamster lung fibroblast (V79-4) cells with IC50 value of 4μg/mℓ. Vitamin B1 showed comparable reduction in lipid peroxidation products (IC50 value was about 10μg/mℓ). It was shown that vitamin C also dose-dependently enhanced the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) in V79-4 cells, and these effects were not observed in vitamin Bl-treated cells. Our data suggest that well-known antioxidant vitamin C involved in direct activation of SOD, CAT and GPX.
