        병결점 부근의 저장온도에 따른 수삼의 내적 품질 변화 연구로 수삼을 각각 , 및 에서 16주간 저장하면서 저장온도에 따른 호흡률, 경도, 가용성 고형물함량, 전분함량, pH, 사포닌함량과 관능적 품질의 변화를 조사하였다. 수삼의 호흡률은 저장온도가 낮을수록 낮았으나 그 차이는 미미하였다. 수삼 표피층의 정도는 저장기간이 경과됨에 따라 증가하였고, 내부 조직의 경도는 감소하는 경향을 보였으나 저장온도에 따라 경도 변화의 차이는 뚜렷하지 않았다.

        The principal objective of this study was to develop a discrimination method using SSR markers in Korean ginseng cultivars. Five cultivars--Chunpoong, Yunpoong, Gopoong, Sunpoong, and Kumpoong--were evaluated by nine markers out of 22 SSR markers. A total of 23 alleles were detected, ranging from 1 to 4, with an average of 2.6 alleles per locus, and an averages of gene diversity (GD) of 0.480. Nine markers were tested in order to distinguish among five Korean ginseng cultivars. Two markers out of nine SSR markers, GB-PG-065 and GB-PG-142, were selected as key markers for discrimination among Korean ginseng cultivars. Two genotypes were detected in GB-PG-065. Chunpoong and Kumpoong shared the same allele type, and Yunpoong, Gopoong, and Sunpoong shared another identical allele type. In the case of GB-PG-142, a specific allele type differentiated from those of other four cultivars was observed only in Sunpoong cultivar. Consequently, the SSR markers developed in this study may prove useful for the identification of Korean ginseng cultivars and the development of ginseng seed management systems, as well as tests to guarantee the purity of ginseng seeds.

        Growth characteristics, root yield and ginsenoside contents of 3-year-old ginseng in greenhouse shaded by 30˚ sloped-curtain made of aluminum were compared to traditional shade facility in order to develop cultural practice for organic ginseng. Light transmittance ratio in greenhouse with 30˚ sloped-curtain shade was distinctly lower than that of traditional shade from sunrise to 9 a.m., while its ratio in greenhouse was higher than traditional shade since 9 a.m. due to the reflection of light. Air temperature of greenhouse was 1.3℃ higher than that of traditional shade on the first ten days of August due to more reflected light. Root yield of greenhouse was 44% higher than that of traditional cultivation because of the inflow of reflected light and the decrease of disease of Alternaria and Anthracnose by blocking rainfall. Dry matter partitioning ratio of rhizome and lateral root were increased in ginseng cultivated at greenhouse due to longer survival time in leaf than traditional cultivation. Total ginsenoside contents cultivated at greenhouse was decreased in the part of taproot, while it was increased in the part of lateral and fine root compare to traditional cultivation. Individual ginsenoside contents between greenhouse and traditional cultivation showed significant difference more frequent in fine root than taproot and lateral root. Total ginsenoside contents including Rb1, Rb2, Rc, Rd, Re, Rf, Rg1, and Rg2 in whole root of 3-year-old ginseng did not showed significant difference by greenhouse and traditional cultivation.

        This study was conducted to examine major growth responses, the production and partitioning of dry matter on different growth stages, and yields and to select the optimal shading material in both quality and productivity of ginseng. Two cultivars of ginseng, Cheonpoong and Geumpoong, were cultivated in the paddy soil with three different shading materials; three-layered blue and one-layered black polyethylene (TBP), blue polyethylene sheet (BPS), and aluminium-coated polyethylene sheet (APS). Plant heights were linearly increased until June 24 and then maintained with showing higher height in Cheonpoong than that in Geumpoong cultivar. Root lengths were gradually increased until October 16. They were longer in Cheonpoong than that in Geumpoong cultivar, showing slightly longer with APS compared to TBP and BPS. The ability of producing dry matter of leaves was much higher from April to June compared to those of other growth periods, whereas its ability of root was concentrated from the end of June to the end of August. Among the shading materials, the ability of producing dry matter of shoot was higher with TBP than those with BPS and APS, while its ability of root was not appeared certain tendency unlike the shoot. The yield of ginseng roots was the highest with TBP among three shading materials and it was higher in Cheonpoong than that of Geumpoong cultivar. The shading materials which affect the light intensity and the temperature would be considered as an important factor to get better quality and productivity of Korean ginseng.

        국내에서 수집된 인삼 자원 중에서 작물학적 특성이 우수하다고 조사된 54점(6~7년 근)을 대상으로 UPOV 조사 기준에 따라 주요 형태적인 특성들을 조사한 결과 경수는 1~4개 범위로 1개인 것이 42.6%이고, 2개인 것이 38.6%로 경수가 1~2개인 것이 전체 81.2%로 나타났으며, 경수가 5개인 것은 없었다. 장엽수는 3~6엽 범위로 5엽이 55.6%였고 4엽인 것은 25.9%였다. 경색은 자색계열이 전체 81.5%로 자색이 46.3%, 연자

        국내 수집 재래종 인삼과 기존육성품종과의 유전적 연관을 통해 풍기지역 고유의 재래인삼 특성을 구명하고 지리적 표시제 및 신품종 육성을 위한 자료로 활용하기 위하여 수행한 결과는 다음과 같다. 유전적 연관 분석에서 가장 먼 것으로 분류된 풍기지역 수집종 331002 등 5점과 금산지역 수집종 332009 등 5점간의 형태적 특성을 비교한바 풍기 재래종 인삼의 경수는 1~2개로 금산지역 수집종과 차이가 없었으나, 장엽수, 경색, 엽병색, 소엽 모양에서는 금산지역 수집종과 차이를 보였다. 즉 풍기 재래종은 장엽수가 5개이고 소엽 모양이 타원형이며, 경색과 엽병색이 자색, 연자색, 진자색 등 다양한 것이 특징적이었다. 특히 유전적 연관에서 풍기지역 수집종 중 331002, 331005, 331007 및 331026은 함께 grouping 되었지만 331002는 대비 품종인 천풍과 99.5%에서 함께 group지어져 있었다. 특히 331002 등 이 3개의 풍기 수집종은 기존 품종과 유전적 차이를 나타내보였다. 이를 통해 풍기 수집종 331002, 331005, 331007은 국내 재래종에서 순계분리법으로 선발된 기존육성품종과는 유전적 차이가 있음을 알 수 있었다. 특히 331007은 농가 조사에서 나타난 주요 특성을 모두 갖추고 있어 풍기 재래인삼의 고유 특성을 가진 것으로 추측 되었다. 본 연구를 통해 풍기지역 재래 인삼의 주요 형태적 특성을 확인하였고, 유전적인 면에서도 타 지역 수집종과 기존육성품종과도 구분되어짐을 확인하였다. 더불어 선발된 OPD05 등 9개의 prime는 경수 1개, 장엽수 5개, 소엽 모양은 타원형, 경색과 엽병색이 자색계열인 특성을 지닌 풍기 재래인삼을 구분하는데 유용하게 활용될 수 있어서 인삼 유전자원의 특성 구분 및 품종개량에 유익하게 활용될 수 있을 것으로 사료된다.

        It's crucial to control Alternaria blight and Anthracnose emerging mostly on ginseng leaves during the rainy season to increase the organic ginseng products. The purpose of this study is to investigate the efficay of lime-brodeaux spray on the ginseng leaves and evaluate the growth and yield of the ginseng, and the contents of ginsenoside and 70% ethanol extracts from 3-year-old ginseng variety, Cheonpoong. Lime-bordeaux sprayings were conducted in the ratio of 6-6 in June, 8-8 from July to September every 15 days. After June 10, the spraying have no effects on the growth leaf and stem, and there was no significant increase in chlorophyll contents. The ratio of intact leaf and root were distinctly increased because Alternaria blight and Anthracnose were decreased by spraying lime-bordeaux mixture. Root weight per plant and root yield were increased by 15%, and 62% in 3-year old ginseng, respectively, because the ratio of intact leaf and root were higher by using lime-bordeaux mixture. Furthermore, spraying of lime-bordeaux mixture is prone to increase the ratio of rusty root in ginseng. Spraying of lime-bordeaux mixture decreased both of the contents of ginsenoside and 70% ethanol extract by 13.7%, and 15.2% in 3-year-old ginseng, respectively.

        This experiment was carried out to select suitable organic matter in To-jik nursery (self soil nursery) for complement To-jik nursery's defects that are deterioration of raw material by poor quality of seed ginseng and reduction of the quantity in seed ginseng production. Organic matter used were Yacto, rice bean, defatted rice bran, soybean cake and their mixture. As follows, bulk density in soil physical property by treating organic composts was the greatest in soybean cake and the next was followed by mix, Yacto, defatted rice bran, and rice bran treatment in order. Soil pore space ratio was totally the opposite; that was rice bran the first and followed by defatted rice bran, Yacto, mix and soybean cake treatment. The incidence rate of damping off by treating organic composts was 1.5% in both soybean cake and mix while the others was 1.0%. Emergence time was the same among treatment on April 16 and Emergence rate was the highest at 73% in Yacto. There was no significant differences among treatment in the growth of aboveground part but it was a little better in defatted rice bran treatment. In Yacto treatment, the growth of underground part, total root number per kan, rate of first grade ginseng seedling, and rate of usable ginseng seedling etc. were entirely higher but there was little differences. Using defatted rice bran was slightly lower in productivity compared to Yacto, but the possibility was high as a alternative for Yacto in a view of managing cost down.

        As cool-season plant, Panax ginseng C. A. Meyer is planted under shade-installation with tall front and low rear. However, at different planting positions, distinct differences come out because ginseng grows at the same position within 3~5 years and the growth circumstance changes a lot by the shade-installation. So, in this study, changes of temperature, photosynthesis and chlorophyll fluorescence with varieties of shading material and planting position were investigated. Light transmittances by polyethylene shade net and silver-coated shading plate as planting materials were measured according to different planting positions. Photosynthetic rate and chlorophyll fluorescence were measured by LI-6400-40 (Li-Cor). According to different planting positions, light intensity was higher in silver-coated shading plate than in polyethylene shade net, and higher at front than rear. Also, photosynthetic rate showed the same tendency, which had a positive correlation to light intensity. But this treatment caused a lower Fo compared with polyethylene shade net because of the stress by light and temperature. Also, Fv/Fm and ETR were higher in silver-coated shading plate. Fo was similar at front and rear according to silver-coated shading plate and ETR was higher at front.

        유연관계를 분석하여 지역 고유의 재래종 인삼의 특성을 구명함과 동시에 신품종 육성을 위한 자료로 활용하기 위해 국내 주요 인삼 재배 지역에서 수집한 재래종 중 작물학적 특성이 우수한 것으로 조사된 풍기 및 경북 지역 수집종 21점과 금산지역 수집종 29점 등 총 58점을 시험재료로 사용하였다. 수집 재래종의 형태적 특성은 UPOV 조사 기준에 따라 경수, 엽병수, 경색, 엽병색, 엽형, 주름새 그리고 거치형 등을 조사하 였으며, RAPD marker를 이용 DNA 수준에서 인삼 수집 개체들의 유전적인 유연관계를 분석하였다. 경수는 1개에서 4개 범위로 경수가 1~2개인 것이 전체의 81.2%로 나타났으며, 경수가 5개인 것은 없었다. 장엽수는 3엽에서 6엽 범위로 5엽인 것이 55.6%였고, 4엽인 것은 25.9%로 나타났으며, 6엽인 것은 6.7%로 나타났다. 경색은 자색계열이 전체의 81.5% 였다. 71개의 polymorphic band를 이용 UPGMA방법으로 분석한 결과 크게 금산지역 수집종과 풍기지역 수집종으로 구분되었고, 4개의 소그룹으로 분류되었는데 풍기 수집종인 01-9, 01-35, 01-44는 금산지역으로 grouping되어진 반면, 금산지역 수집종 332001, 332002, 332003은 풍기지역으로 grouping되었다. 금산지역 수집종은 65~86%로 변이가 심한 반면, 풍기지역 수집종은 73~95%사이로 금산 수집 종의 혼계 정도가 더 심하였다. 특히 형태적 특성 면에서 두 지역간에 경수는 1~2개로 차이가 없었으나 장엽 수, 경색, 엽병색, 소엽의 모양 등에서는 다소 차이가 있었다.

        This study was conducted to investigate the effect of planting position on the growth characteristics, yield and ginsenoside content in Panax ginseng C.A. Meyer at different growth stages. Referring to shoot growth characteristics, stem length, stem diameter and leave area were higher at front than rear, increasing as the proceeding of growth stages. But a lower chlorophyll contents was caused at front compared to rear and decreased as the proceeding of growth stages contrarily. According to root characteristics, root length and main body length were higher at front, with a positive correlation to growth stages, which was also shown on fresh root weight and dry root weight with the maximum in August. Meanwhile, the effect of planting position on ginsenoside content could also be definite by the highest content at front showing high light intensity, increasing as the proceeding of growth stages as well.

        인삼의 해가림구조에서 차광판 및 차광망의 차광재료 차이에 따른 인삼 재배포의 광량 및 미기상 반응과 광수용체로 작용하는 엽록소 함량의 변화를 조사하여 인삼생육에 적합한 해가림 차광재료의 표준화를 위한 기초 자료를 제공하고자 한 본 연구의 결과를 요약하면 아래와 같다. 1. 차광재료에 따라 맑은 날의 광량은 차광판이 차광망에 비해 70 ~ 76%, 흐린 날은 77 ~ 82% 수준으로 감소되는 양상을 나타내었다. 2. 두둑위치 중 전주의 경우는 하루 총 누적광량에 대비해 맑은 날의 경우 39% 수준, 흐린 날의 경우 51% 누적광량이 높은 양상을 나타내며, 중앙 및 후주행은 맑은 날 및 흐린 날 구분 없이 모두 하루 총 누적광량에 비해 낮은 양상을 나타내므로, 두둑위치에 따른 누적광량의 차이가 분명하였으며, 차광재료에 관계없이 두둑위치에 따라서는 전주 > 중앙 > 후주행의 순으로 광량이 낮아지는 양상을 나타내었다. 3. 차광재료에 따라 차광망이 차광판에 비하여 높은 온도를 유지하였고, 두둑위치에 따라 차광망이 차광판 보다 1.9~3.1℃ 높은 양상을 나타내었다. 4. 차광재료별 맑은 날의 평균 누적온도는 외부온도 대비차광판의 경우 7.3% 감소하는 양상을 나타내었으나, 차광망의 경우 0.7% 증가되는 양상을 나타내었고, 흐린 날의 평균 누적온도는 차광판의 경우 0.7% 감소하는 양상을, 차광망의 경우 1.6% 증가하는 양상을 보였다. 5. 차광재료에 따른 두둑위치별 일일 누적온도의 차이를 비교해 보면 차광망이 차광판에 비해 맑은 날의 경우 전주, 중간 및 후주행이 각각 7.0, 7.8 및 8.8% 높은 양상을 나타내고, 흐린 날의 경우 각각 1.7, 1.6 및 3.5% 높은 양상을 나타내어 두둑위치에 관계없이 차광판이 차광망에 비해 온도가 낮은 양상을 나타내었다. 6. 엽록소 SPAD 값은 전주와 중앙의 두둑위치에서는 차광판이 차광망에 비하여 높게 나타났고 후주에서는 차이가 없었다.

        인삼 논재배 안전생산기술을 개발하기 위해 배수불량지와 배수약간불량지 논토양에서 청3+흑1중직 차광망, 청색차광지 및 은박차광판 해가림을 처리하여 3년생 천풍의 생육특성 및 진세노사이드 함량을 조사한 결과는 다음과 같다. 해가림 유형별 투광량과 기온은 차광지 > 은박차광판 > 차광망 순이었다. 배수불량지의 토양수분장력은 평균 64mbar (절대토양수분함량 25%)로 과습한 조건이었으며, 배수약간불량지에서는 평균 123 mbar (절대수분함량 17%)이었다. 배수불량지의 토양수분장력은 계절과 해가림 유형별 뚜렷한 차이를 보이지 않았지만 배수약간불량지에서는 60~200 mbar의 범위에서 계절적 변화를 보였으며, 해가림유형에 따라 약간의 차이를 보였다. 배수불량지의 수량성은 황증과 적변 발생율의 증가로 배수약간불량지보다 현저히 감소되었다. 해가림 유형별 수량성은 배수불량지에서 차광지가, 배수약간불량지에서 차광망이 가장 높았다. 적변 발생율은 배수등급과 해가림 유형에 따라 일정한 경향이 없었다. 배수불량지는 Panaxatriol 계열 (Rg1, Re 및 Rf)의 진세노사이드 함량이 감소되고 Panaxadiol 계열 (Rb1, Rc 및 Rd)의 함량이 증가되었다. 총 진세노사이드 함량은 배수불량지보다 배수약간불량지에서 높았으며, 해가림 유형별 총 진세노사이드 함량은 배수등급에 관계없이 차광지에서 가장 높았다.

        This study was carried out to investigate the correlation between root diameter and ginsenoside compositionof Panax ginseng C. A. Meyer cultivar Yunpoong. Dry matter ratio of main root was a little higher than that of lateral rootand fine root, and that was higher by the increase of root diameter in the same root parts. Total ginsenosides composition ofmain and lateral roots increased by the decrease of root diameter, especially in lateral root. Similar resulted in fine root, butthere was no significant difference where root diameter was below 2.5㎜. Except for ginsenoside-Rg1, other ginsenosidescomponent, PDs, PTs and total ginsenosides had highly negative correlation with the root diameter within whole root, mainroot+lateral root and lateral root+fine root, while Rg1 had positive correlation with the root diameter.

        To study the optimal cultivation for paddy soil, growth characteristics and ginsenoside content was investigated by both of poor drainage class (PDC) and imperfect drainage class (IDC) in three-year-old ginseng of varieties, Cheonpoong (CP), Yeonpoong (YP), Hwangsookjong (HS), and Jakyeongjong (JK). Root yield of IDC was higher than that of PDC by 3.6 times because stem length, leaf area, and chlorophyll contents were increased, while discolored leaf ratio was decreased. Root yield of HS in PDC was highest among four varieties because chlorophyll contents, leaf area, and survived plant ratio were relatively high. Root yield of CP in IDC was highest among four varieties because of high leaf area and survived plant ratio, and low discolored leaf ratio. Ratio of rusty-colored root showed significant difference by varieties, which was the highest in HS and the lowest in CP among four varieties irrespective of drainage classes. Total ginsenoside contents showed significant difference by drainage classes, which were high in IDC of good growth and low in PDC of poor growth. Total ginsenoside contents were high in JK and CP, while low in HS and YP both of drainage classes.

        Cinnamyl alcohol dehydrogenase (CAD, EC 1.1.1.95), catalyzes the reduction of hydroxycinnamaldehydes to give hydroxycinnamyl alcohols, or "monolignols," the monomeric precursors of lignin. Lignins are important components of cell walls and lignified secondary cell walls play crucial roles in long distance transport of water and nutrients during plant growth and development and in plant defense against biotic and abiotic stresses. Here a cDNA clone containing a CAD gene, named as PgCAD, was isolated from a commercial medicinal plant Panax ginseng. PgCAD is predicted to encode a precursor protein of 177 amino acid residues, and its sequence shares high homology with a number of other plant CADS. The expression of PgCAD in adventitious roots and hairy roots of P. ginseng was analyzed using reverse transcriptase (RT)-PCR under various abiotic stresses such as salt, salicylic acid, wounding and chilling treatment that triggered a significant induction of PgCAD at different time points within 2-48 h post-treatment. This study revealed that PgCAD may help the plants to survive against various abiotic stresses.

        The parts of leaves, flowers and stems in ginseng were obtained for analyzing the component of saponin on 15th April, 25th April, 5th May, 25th May, which were considered as ginseng foliation stage. The total saponin content of the leaves were 97.29, 66.42, 67.61, 36.24 mg/g, respectively, in which the content of Re, Rb1 and Rd were more than 2/3 amount of total saponin. Especially, the saponin content of leaves decreased according to the sequential collection days, in which the similar results were observed from the flowers and stems of ginseng. The total saponin content of the flowers and stems were 141.09,143.84,139.25,133.47 and 13.32, 9.85, 8.00, 4.65 mg/g, respectively. Among them, the content of Re, Rd and Rb2 in flowers were more than 2/3 while the content of Re, Rg1 and Rd in stems showed more than 9/10 amount of total saponin. The total saponin content of individual leaf were 19.46, 28.56, 58.82 and 169.24 mg/plant, 2.53, 2.76, 5.20 and 12.32 mg/plant in stems, and 14.11, 30.21, 37.60 and 73.41 mg/plant in flowers. Therefore, the total saponin content of aboveground parts in ginseng were leaves 〉 flowers 〉 stems.

        Somatic embryos on growth regulator-free medium can be produced directly from cotyledon explants of Panax ginseng C. A. Meyer. When the embryo developmental stage was torpedo and cotyledon, the germination rate of embryos was quite high on MS medium supplemented with gibberellic acid (GA3). However, the percentage of plantlet formation at the cotyledon stage was higher than that at the torpedo stage. This result demonstrates that the embryo at the cotyledon stage was the most appropriate for increasing germination by GA3. Embryos cultured on medium including four levels of GA3 concentrations (3, 5, 10, or 20 mg/l) showed all quite high germination rates (87-91%). When the well-developed embryos were continuously cultured on media including high concentrations of GA3 from 10 to 20 mg/l, the percentage of formation of normal plantlets was lower than that seen under low concentrations from 3 to 5 mg/l. This treatment of high concentrations resulted in shoots with abnormal shape. The optimal GA3 treatment provides a basis for the efficient method obtaining healthy plantlets derived from ginseng somatic embryos.

        This study was conducted to assess the response of LED (Light-emitting diode) irradiation on the growth characteristics and saponin contents of Panax ginseng C. A. Meyer. LED irradiation showed a positive effect for most of the parameters studied. The content of chlorophyll a in leaves was increased by 4.9~36.5%, under LED and fluorescent light conditions compared to the control. The content of chlorophyll b was also increased by 44.4~55.6% under blue and red LED compared to the control except under the red plus blue LED condition. The shoot and root weight were increased by 20~60% and 14.8~59.3%, respectively under LED and fluorescent light conditions compared to the control. The total saponin content was increased by 1.8% under blue LED compared to the control, while total saponin content was decreased by 8.8~11.5% under red LED, red plus blue LED and fluorescent light conditions.

        본 연구는 5 년근의 인삼에서 차광시설의 재배위치에 따른 사포닌의 함량 차이를 검정하였다. 재식 위치별 전행 (1~2행) 에서 연풍과 재래종 주근의 총 사포닌 단위함량은 각각 15.01 mg/g, 21.79 mg/g, 지근은 각각 35.93 mg/g, 43.32 mg/g, 세근은 각각 87.85 mg/g, 105.51 mg/g이었다. 중행 (3~5행) 에서 연풍과 재래종 주근의 총 사포닌 단위함량은 각각 18.73 mg/g, 23.19 mg/g, 지근은 각각 44.92 mg/g, 43.50 mg/g, 세근은 각각 92.97 mg/g, 110.70 mg/g이었다. 후행 (6~7행) 에서 연풍과 재래종 주근의 총 사포닌 단위함량은 각각 21.88 mg/g, 26.68 mg/g, 지근은 각각 38.41 mg/g, 44.89 mg/g, 세근은 각각 101.03 mg/g, 107.06 mg/g이었다. 재배위치에 따른 연풍과 재래종의 주근, 지근 및 세근의 총 사포닌과 주요 사포닌 함량의 차이는 연풍의 주근을 제외하고 모든 부위에서 크지 않았다. PD/PT 비율은 전행 인삼은 중행과 후행에 비하여 낮았으며 뿌리의 부위별로도 차이가 컸다.
