본 연구는 6개 핵돈군생산농장(Great grand parents, GGP)에서 2015년 이후 태어난 Landrace종과 Yorkshire종의 두 품종에 대하여 생시체중 (Birth weight; BW), 복당평균생시체중(Mean of litter birth weight; MBW0)과 변이계수(Coefficient of variation in MBW0; CVMBW0) 및 산자수(Litter size) 등에 대한 유전모수를 추정하였다. Landrace종에서 총산자수(Total number of piglets born; TNB), 생존산자수(Number of piglets born alive: NBA), 복당평균생시체중 및 복당평균생시체중 변이계수의 유전력은 각각 0.14, 0.09, 0.33 및 0.16. Yorkshire종에서는 각각 0.11, 0.99, 0.28 및 0.07로 추정되었다. 산자수와 복당평균생시체중 간에는 부(-)의 상관관계를 나타내었으며, 복당평균생시체중과 변이계수사이의 표현형상관과 유전상관이 두 품종에서 모두 –0.40에서 –0.45로 높게 나타났다. 자돈 생시체중에 대한 균일도를 개량하기 위해서는 복당평균생시 체중(MBW0)과 복당평균생시체중변이계수(CVMBW0)를 선발지수식에 포함시키는 것이 바람직하다고 생각된다.
Some behaviors of pigs that are not expressed in the wild state or are observed in a small minority of individuals after groups of pigs are mixed have been reported to indicate poor welfare. A GWAS analysis was performed by measuring the frequency and duration of the four ethological traits and using the mlma command provided by the genome-wide complex trait analysis (GCTA). The positional candidate genes on significantly identified single nucleotide polymorphism (SNP) markers were identified by using the dbSNP provided by the National Center for Biotechnology Information (NCBI). When the GWAS analysis was applied the 43,565 (of the purebred Landrace population) and 41,700 (of the purebred Yorkshire population) SNP markers, 1, 2, and 1 significant SNP markers were identified for the traits of feeding frequency (LOC110262254), locomotion time (LOC110260361), and locomotion frequency (LOC110260361) of the purebred Landrace population, respectively. Meanwhile, 1 and 7 significant SNP markers were identified for the traits of drinking time (LOC 110260090) and feeding frequency (MAP3K19; LOC110257013; ACMSD; TMEM163; RAB3GAP1) of the purebred Yorkshire population, respectively. The results of this study may suggest that the GWAS analysis of the ethological traits of purebred Landrace and Yorkshire populations could be used to perform a GWAS analysis on non-economic traits, and the results can thus be provided as basic data for GWAS analyses of other non-economic traits in the future.
According to Livestock Inspection Standards, the piglets enter the feedlot at approximately 30 kg, and the inspection starts after the preliminary feeding period. The reason for applying the preliminary feeding period is to select inspection piglets with no diseases after the complete growth of the internal organs until 10 weeks of age. Furthermore, the age of 10 week is the time when the muscle fibers grow to their maximum size and the piglets are prepared for fat deposition at the later fattening period. In the study, a genome-wide association study (GWAS) was performed through the mlma command of the genome-wide complex trait analysis (GCTA) program with 703 purebred Landrace population, and the candidate genes associated with the weight of 10 week were searched. The GWAS identified 3 single nucleotide polymorphism (SNP) markers, which have a significant genome-wide suggestive level, on chromosome 6 (DIAS0002615; p-value=1.62×10-6, MARC0083933; p-value=4.94×10-6, ASGA0028717; p-value=5.40×10-6). The 2 genes (Ubiquitin protein ligase E3 component n-recognin 4; UBR4, WD and tetratricopeptide repeats 1; WDTC1) in which these 3 SNP markers are located are positional candidate genes of the weight of 10 week of the purebred Landrace population. 2 candidate genes have been reported to be associated with fattening. Therefore, the positional candidate genes in this study, UBR4 and WDTC1, are expected to be usable as genes for traits associated with the weight of 10 week weight and fattening through additional experimental research with other population.
This study was conducted to find out that how much does it effects as it considered not only animal additive genetic effect but also maternal genetic effect for improving litter traits of pigs. The data of 10,836 records on 2,636 sows in Landrace and 14,463 records on sows in Yorkshire were analyzed which had been measured from 1998 to July 2017 in a nucleus herd of pig population. The traits used on this analysis were total number of born with (TNB2) and without mummy (TNB1) and number of born alive (NBA). Two different multivariate animal mixed models were considered and compared of variance components estimated from these models. The one (Model 1) was set up with assumed to parity, return events and batch effects as fixed and service sire, permanent environment and animal additive genetic effects as random. The other (Model 2) was same with Model 1 except considering maternal additive genetic effects as random. (Co)variance for random effects and genetic parameters were estimated using restricted maximum likelihood method and breeding values as best linear unbiased prediction were estimated using preconditioned conjugate gradient algorithm on each model and breed. From these models, heritability estimates for NBA were about 0.10 and 0.11 on both models in Landrace and Yorkshire, respectively. Forthermore, it was estimated that there were little variations in the maternal genetic effects with roughly 1~2% of total variation. Result from comparing estimated breeding values for each trait between each model, ranking of genetic capability through total breeding values on model 1 and on model 2 showed highly correlated with more than 0.92. Consequently, for improving litter traits, selection based on breeding values by direct genetic effects without considering maternal genetic effects were reccommendable.
돼지의 생시체중은 생존율과 폐사율에 밀접한 관련이 있어 양돈산업에서 자돈 관리와 직결된 중요한 경제형질이다. 본 연구는 Genome-Wide Association Study(GWAS) 분석을 통해 순종 랜드레이스의 생 시체중과 관련된 위치상 후보유전자 탐색을 실시하였다. 생시체중의 유의적 관련 분석 결과, genomewide suggestive level에서 유의성 있는 single nucleotide polymorphism(SNP) marker는 3번 염색체 (ASGA0098921, P=2.41×10-5)와 4번 염색체(H3GA0013451, P=2.47×10-5)에서 각각 1개씩 동정되었다. 이들 SNP marker가 위치한 3개의 유전자(LOC110260055, LOC100156472, LOC100157689)들은 순종 랜드레이스 생시체중의 위치상 후보 유전자이며, 이들 유전자 정보를 이용해서 순종 랜드레이스 생시 체중을 선발할 수 있는 기초 연구 자료가 될 것으로 사료된다.
Alterations affecting the status of robustness and health can bring about physiological changes including hematological parameters in pigs. To identify quantitative trait loci (QTL) associated with 8 hematological phenotypes (one leukocyte trait, six erythrocyte traits, and one platelet trait), we performed a genome-wide association study using the Porcine SNP 60K BeadChip in an intercross population between Landrace and Korean native pigs. A total of 36,740 SNPs from 816 F2 offspring were analysed for each blood related traits after filtering by quality control. Data were analysed genome-wide rapid association using the mixed model and regression (GRAMMAR) approach. A total of 257 significant SNPs (P<1.36x10-6) on SSC3, 6, 8, 13, and 17 were detected for blood related traits in this study. Interestingly, the genomic region between 17.9 and 130 Mb on SSC8 was found to be significantly associated with RBC, MCV, and MCH. Our results include 5 significant SNPs within five candidate genes (KIT, IL15, TXK, ARAP2, and ERG) for hematopoiesis. Further validation of these identified SNPs could give valuable information for understanding the variation of hematological traits in swine.
Previously, we reported a quantitative trait locus (QTL) that affect total teat number (TTN) on pig chromosome 7 (SSC7) in a large F2 intercross population between Landrace and Korean native pigs. The aim of this study was to refine the QTL associated with TTN and to identify positional candidate gene(s) within the refined genomic region. TTN was recorded in 1,105 F2 progeny. All experimental animals were genotyped using 998 informative single nucleotide polymorphism (SNP) markers located on SSC7. A haplotype-based linkage and association analysis using the PHASEBOOK programme was applied to perform high-resolution QTL analysis. Additionally, linear mixed-effect models were used to assess the effect of a positional candidate gene on TTN and other economically important traits [i.e., thoracic vertebrae number (THO), carcass body length (CBL) and weight (CW), back fat thickness (BFT) and intramuscular fat content (IMF) in loin muscles]. Joint linkage and association analysis refined the critical region to a 1.07 Mb region that included a novel positional candidate gene, BRMS1L, that encodes the breast cancer metastasis-suppressor 1-like protein, which could possibly be implicated in normal mammary gland development. Significant association of an SNP marker (g.-1087 G>A) in the 5’-flanking region of BRMS1L with TTN (P=1.10x10-8), THO (P=5.80x10-4), and CBL (P=0.038) was observed. Based on these data, we propose BRMS1L as a positional candidate gene for TTN in pigs. After validation of the association in other independent populations and further functional studies, these results could be useful in optimizing breeding programmes that improve TTN and other economically important traits in swine
The aim of this study was to identify quantitative trait loci (QTLs) influencing teat number traits in an F2 intercross between Landrace and Korean native pigs (KNP). Three teat number traits (left, right, and total) were measured in 1105 F2 progeny. All experimental animals were genotyped with 173 informative microsatellite markers located throughout the pig genome. We detect that seven chromosomes harbored QTLs for teat number traits: genome regions on SSC1, 3, 7, 8, 10, 11, and 13. Six of fourteen identified QTL reached genome-wide significance. In SSC7, we identified a major QTL affecting total teat number that accounted for 5.6% of the phenotypic variance, which was the highest test statistic (F-ratio = 61.1 under the additive model, nominal P = 1.3×10-14) observed in this study. In this region, QTL for left and right teat number were also detected with genome-wide significance. With exception of the QTL in SSC10, the allele from KNP in all 6 identified QTLs was associated with decreased phenotypic values. In conclusion, our study identified both previously reported and novel QTL affecting teat number traits. These results can play an important role in determining the genetic structure underlying the variation of teat number in pigs.
Growth traits, such as body weight, directly influence productivity and economic efficiency in the swine industry. In this study, we estimate heritability for body weight traits usinginformation from pedigree and genome-wide single nucleotide polymorphism (SNP) chip data. Four body weight phenotypes were measured in 1,105 F2 progeny from an intercross between Landrace and Jeju native black pigs. All experimental animals were subjected to genotypic analysis using PorcineSNP60K BeadChip platform, and 39,992 autosomal SNP markers filtered by quality control criteria were used to construct genomic relationship matrix for heritability estimation. Restricted maximum likelihood estimates of heritability were obtained using both genomic- and pedigree- relationship matrix in a linear mixed model. The heritability estimates using SNP information were smaller (0.36-0.55) than those which were estimated using pedigree information (0.62-0.97). To investigate effect of common environment, such as maternal effect, on heritability estimation, we included maternal effect as an additional random effect term in the linear mixed model analysis. We detected substantial proportions of phenotypic variance components were explained by maternal effect. And the heritability estimates using both pedigree and SNP information were decreased. Therefore, heritability estimates must be interpreted cautiously when there are obvious common environmental variance components.
The aim of the present study was to investigate the ovulation rate and its relationship to fertilization ability in Landrace, Durock and Crossbred pigs. Gilts were natural mated at a body weight of at least 120 kg under the same hormone treatment. Embryos were surgically collected 1 day after natural mating (Day 0). Embryos derived from in vivo-fertilized oocytes were cultured in medium PZM-3. The ovaries were examined and the pathological findings were recorded. The number of corpus hemorrhagicum was counted, and was assumed to equal the ovulation rate. There was no difference in the number of corpus hemorrhagicum (20.4, 28.8 and 23.2) and ovulation (13.5, 26.8 and 17.2) in the Landrace, Durock and Crossbred pigs. The two pronucleus formation was 76.0, 80.0 and 86.9%. The Day-7 embryos had blastocyst rates of 68.0, 75.0 and 73.9%. There was no difference in the number of total cells and apoptotic cells. In the future, more studies require determining relationships between ovulation and fertilization rate in different species of pigs.
앙골라에 잘 적응하는 다수성 옥수수 품종 육성을 위한 기초지식을 얻기 위해 앙골라 옥수수 재래종 자원 89점을 대상으로 24개의 SSR 마커를 이용하여 유전적 다양성 분석을 실시한 결과는 다음과 같다.
총 254개의 allele가 탐지되었고 마커당 평균 allele 수는 10.6이었다.
마커의 PIC 값은 0.23 ~ 0.92 범위에 있었으며 평균값이 0.64이었다.
앙골라 옥수수 재래종 유전자원들은 매우 높은 유전적 다양성을 보여 계통간 비유사성(dissimilarity) 평균값이 0.643 이었고, 뚜렷하게 소수의 그룹들로 구분되지 않았다.
이러한 고도의 다양성을 가진 재래종 옥수수 유전자원을 이용하여 앙골라에 잘 적응하는 다양한 다수성 품종을 육성할 수 있을 것으로 기대된다.
The aim of this study was to develop the production of crossbred pigs suitable to the Korean consumers’demand. Crossbred (Jeju native pig × Landrace) pigs (n=175) at the age of 200 days, approximately classified into three slaughter weight groups (59~95 kg, 96~110 kg and 111~148 kg) were used to investigate the carcass traits and retail cuts characteristics. We have chosen longissimus dorsi, triceps brachii, biceps femoris, semimembranosus muscles, shoulder butt, thoracic vertebra (5-11th) and last thoracic vertebra for investigations of aforementioned parameters. The results showed that the 111~148 kg group had larger loin area, higher scores for meat color, quality and carcass grade than the other remaining groups (p<0.05). Weights of shoulder butt, rib, picnic shoulder, loin, belly, tenderloin, ham retail cuts, lean meat, fat and bone increased with increasing carcass weight (p<0.05). Ratios of shoulder butt, picnic shoulder, tenderloin, ham, lean meat, skin and bone in the 59~95 kg group were higher than that of the other groups (p<0.05). Ratios of belly in the 96~110 kg group were higher than that in the 59~95 kg group (p<0.05) but similar to the ratios of the 111~148 kg group. Total length, longest length and shortest length of belly increased with increasing carcass weight (p<0.05). Additionally, the longest width and shortest width of belly in 59~95 kg group were lower (p<0.05) than that in the other remaining groups. Length of shoulder butt increased with increasing carcass weight (p<0.05) whereas, the width of shoulder butt showed no differences between the groups. Weights of triceps brachii of picnic shoulder, biceps femoris and semimembranosus muscles of ham increased with increasing carcass weight (p<0.05). Overall, the crossbred pigs of 111~148 kg group possessed better quality and carcass grade however, the yield of belly in 96~110 kg group was higher than the other groups. Based on the consumption preference of Korean consumers therefore, the slaughter weight of crossbred pigs at ≥96 kg could produce higher economic benefit.
This study was conducted to investigate the changes of hormone levels of serum progesterone (P4) and estradiol-17 β (E2) in sows of Landrace (L), Yorkshire (Y) and F1 (L × Y) (respectively n=3) with excellent ability, and to provide a baseline data for improving reproductive performance. In this experiment, the sows at the age of 12 months or more were used. The sows were fed by two way methods, one is conventional methods and the other is 3 days-flushing feed before estrus. Each pig’s blood was collected in 3, 6, 9, 12, 15 and 18 days after the estrus for the analyses of P4 and E2. Serum was separated by centrifugation for 15 min. with 3,000 rpm. Progesterone and estradiol-17β were measured by immunochemical assay (ELIZA test). In conventional feeding, serum progesterone levels were significantly (p<0.01) higher in F1 than in L and Y. No significant differences in P4 concentrations were seen between the L and Y of sows. Serum E2 levels were similar the serum progesterone levels. In the case of flushing feed, the tendency of hormonal changes were similar to conventional methods. But almost of hormonal levels were a little higher than that of conventional methods. P4 level of L and Y in flushing feed were significantly different (p<0.01). Serum E2 level of Y in flushing feed was significantly different among the breeds (p<0.01). These results were similar to the tendency of hormonal changes in general sows and moreover, flushing feed is known to develop the swine production, these results proved the fact of the methods. And these results suggested that more studies about hormonal changes in sows according to seasonal and nutritional factors should be needed.
돼지의 체장, 체폭, 체중 등의 연속적인 변이를 가지는 양적형질에 속하는 형질인 성장형질은 양돈업에 있어서 생산성과 수익성에 직결된 중요한 경제 형질이다. 성장형질로는 일당증체량, 등지방두께, 등심단면적 및 도체품질 등이 있으며, 그중에서 등심단면적과 관련이 있는 후보 유전자를 Genome-wide 수준에서 선발하기 위하여 Illumina 사의 Porcine SNP 60K chip을 이용하여 순종 Landrace 집단(수컷 : 50두, 암컷 : 440두)을 분석하였다. Plink program을 이용하여 filtering 과정을 거쳐 최종적으로 31,960개의 SNP marker를 선별하였으며, Genome-wide 임계수준을 bonferroni correction (i.e.1/31957=3.12×10-5)을 이용해서 결정했을 때, SSC16에서 유의성 있는 4개의 SNP marker들을 발견하였다. 이 중 가장 유의성 있는 SNP marker인 DRGA0016148(P=1.38×10-5)와 근접한 후보유전자로 PDE4D를 선정하였고, PDE4D에서 2개(g.9870 T>C, g3976 C>T)의 SNP를 탐색하였다. 본 연구에서는 Genome-wide association study (GWAS)를 이용하여 등심단면적과 관련성이 있는 SNP를 선발하여, 등심단면적과 후보유전자 PDE4D와의 관련성을 규명하고자 연구를 실시하였으나, PDE4D g.9870 T>C 에서는 Hardy-weinberg equilibrium P-value 0.7581로 음의 상관관계를 나타내었으며, PDE4D g.3976 C>T에서는 모두 호모타입이 확인되었다.
국내 토종 자원으로 알려진 앉은뱅이 밀의 밀가루 특성 밀 과자 적성을 평가하기 위해서 경남 진주시에서 구입한 앉은뱅이 밀의 통밀가루 및 백밀가루와 중형제분기로 제분한 밀가루를 평가하였다. 앉은뱅이 밀 구입처에서 제분된 제분된 통밀가루와 백밀가루는 중형제분기에서 제분된 밀가루에 비하여 밀기울 혼입으로 인하여, 회분과 단백질 함량이 높고 밀가루 색깔이 어두웠고 침전가와 건부율 및 아밀로스 함량은 낮았다. 반죽 시간이나 안정도는 밀가루간에는 차이가 없었지만, 반죽 가수량은 밀기울을 포함한 통밀가루가 높았고, 쿠키 직경이 적게 나타났다. 중형제분기로 제분된 앉은뱅이 밀가루의 밀가루 입도와 손상 전분 함량이 각각 54.44μm와 2.46%로 연질밀 특성을 나타내었으며, 총아라비노자일란 함량과 수용성 아라비노자일란 함량이 각각 28.52%와 9.49% 로 국내 밀 품종보다 높았다. 쿠키 직경은 86.88mm로 수입산 박력분과 유사하였으며, 쿠키 표면의 균열도 쿠키 적성에 적합한 것으로 나타났다.
본 연구는 농촌진흥청 국립축산과학원에서 사육중인 제주재래돼지와 랜드레이스의 상호교차교배를 통해 생산된 417두의 F2집단을 대상으로 성장형질(생시체중, 3주령 체중, 10주령 체중, 20주령 체중)을 측정하고, 개체별 혈액으로부터 Illumina Porcine SNP 60k BeadChip을 이용하여 유전자형 분석을 실시하여 성장형질과 유전체 전장의 단일염기다형의 유전자형 간에 연관성을 알아보았다. 단일연기다형의 유전자형 분석은 돼지의 성염색체를 제외한 18개의 상염색체 내에 나타난 52,574개의 SNP표지인자 중다형성이 나타나지 않은 7,564개의 표지인자, 모든 개체에서 이형으로 나타난 47개 표지인자 및 결측률이 10 % 이상인 1,830개의 표지인자가 나타나 분석에 앞서 사전 제거를 실시하였으며, 남은 44,133개의 표지인자를 체중형질과 표지인자간 연관성 분석하는데 이용하였다. 체중에 영향하는 고정효과에 대해 일반선형모형을 설정하여 사전 보정을 실시하였으며, 여기서 얻어진 잔차값을 이용하여 표지인자와 월령별 체중간의 연관성 분석을 실시하였다. 분석결과 전장의 유전체 정보 중 통계적 판단의 오류를 고려하여 연관성이 강한(p <10-6)표지인자가 생시(BWB), 3주령(BW3), 10주령(BW10) 및 20주령(BW20) 체중에서 각각 657개, 846개, 49개, 122개로 추정되었다. 생시와 3주령 체중에 공통으로 유의적 영향을 하는 표지인자가 286개로 나타났으며, BWB와 BW10에서 5개, BWB와 BW20에서 8개, BW3와 BW10에서 13개, BW3과 BW20에서 11개, BW10과 BW20에서 1개로 나타났다. 또한 염색체별 성장형질에 영향하는 표지인자의 분포를 조사한 결과, 주령별 체중에 영향하는 표지인자는 전장의 유전체에 고르게 분포하는 것으로 조사되었으며, 특히 생시 및 3주령 체중에 영향하는 표지인자는 특정 염색체(SSC9)에서 고도의 통계적 유의차(p <10-15)를 나타내는 유전자 좌위가 있는 것으로 추정되었다.
The objective of this study was to find out candidate genes associated with litter size trait in pigs of inbred Large Yorkshire and Landrace populations. 86 sows were screened for candidate genotypes along with litter size data recordings. Association of litter size with genotypes of candidate genes were investigated to verify the usefulness of each gene's genotypes as markers for the trait. For the lines of Large Yorkshire, PRLR3 and RBP4 genes were genotyped. Frequency distribution of PRLR3 with genotypes AA, AB and BB were each 0.14, 0.44 and 0.42. And the average litter size by PRLR3 genotypes were 8.83, 10.81 and 10.70 piglets per litter, the average estimated breeding values of which were 0.243, 0.332, 0.365, respectively for AA, AB and BB genotypes. Genotypic frequencies of RBP4 by AA, AB and BB genotypes were 0.10, 0.44 and 0.46. The average litter size by genotypes of RBP4 were 10.40, 10.57 and 10.35 piglets per litter and their corresponding average estimated breeding values were 0.451, 0.353 and 0.261, respectively for genotypes AA, AB and BB. Significance in differences among genotypes were not observed, but B allele of RBP4 seems to be associated with litter size. In Landrace lines, frequencies of RBP4 genotypes, AA, AB and BB were 0.29, 0.55 and 0.16. And the average litter size of these genotypes were 10.50, 11.08 and 11.00 piglets per litter. The corresponding averages of estimated breeding values of each genotypes were 0.172, 0.135 and 0.104. In Landrace lines, allele A was more likely to be associated with litter size, even if differences among average litter size were not significant. We conclude that genotyping of two candidate genes is a helpful tool to identify genetic potentials of litter size in pigs.
This study investigated the developmental ability and gene expression of somatic cell nuclear transfer embryos using ear skin fibroblast cells derived from miniature pig. When miniature pig (m) and landrace pig (p) were used as donor cells, there were no differences in cleavage (79.2 vs. 78.2%) and blastocyst rates (27.4 vs. 29.7%). However, mNT blastocysts showed significantly higher apoptosis rate than that of pNT blastocysts (6.1 vs. 1.7%) (p<0.05). The number of nuclei in pNT blastosysts was significantly higher than that of mNT (35.8 vs. 29.3) (p<0.05). Blastocysts were analyzed using Realtime RT-PCR to determine the expression of Bax-, Bcl-xl, H19, IGF2, IGF2r and Xist. Bax- was higher in mNT blastocyst than pNT blastocyst (p<0.05). There was no difference in Bcl-xl between two NT groups. Bax-/Bcl-xl was, however, significantly higher in mNT blastocyst compared to pNT. The expression of imprinting genes were aberrant in blastocysts derived from NT compared to in vivo blastocysts. H19 and IGF2r were significantly lower in mNT blastocysts (p<0.05). The expression of IGF2 and Xist was similar in two NT groups. However, imprinting genes were expressed aberrantly in mNT compared to pNT blastocysts. The present results suggest that the NT between donor cells derived from miniature pig and recipient oocytes derived from crossbred pig might affect reprogramming of donor cell, resulting in high apoptosis and aberrant expression patterns of imprinting genes.
