본 연구에서는 감국 꽃 추출물의 항산화 및 항염활성을 검색하고 유효성분을 분리하여 화학구조를 규명하였다. 감국 꽃 에탄올 추출물의 DPPH 및 ABTS+ 라디칼 소거 활성을 측정한 결과 우수한 라디칼 소거 활성이 있음을 확인하였다. 또한 LPS로 유도된 RAW264.7 대식세포에서 세포독성 없이 우수한 nitric oxide (NO) 저해 활성을 나타내었다. 감국 꽃 에탄올 추출물의 유효성분을 찾고자 medium pressure liquid chromatography (MPLC)를 실시하여 2개의 화합물을 분리하였으며 1H 및 13C NMR 데이터 분석 및 문헌 비교를 통하여 화학구조를 동정하였다; Cynaroside (1), Apigetrin (2). 분리된 화합물을 HPLC를 통하여 정량분석을 수행하였으며 감국 꽃 추출물의 주요 성분으로 확인되었다. 화합물 1, 2의 항염활성을 확인한 결과 모두 농도의존적으로 우수한 NO 저해활성을 나타내었다. 이러한 연구 결과를 바탕으로 감국 꽃 추출물은 천연 항산화 및 항염 소재로써 활용 가능할 것으로 사료된다.
감국(Dendranthema indicum)의 환경조건에 따른 개화기의 초장, 개화소요일수, 카로티노이드 함량 및 peroxidase 활성을 고랭지(대관령, 표고 800m)와 평난 지(강릉, 표고 20m)의 노지와 온실에서 각각 비교하여 원예화 개발 가능성을 검토하였다. 환경조건에 따른 광 량은 고랭지 노지 > 평난지 노지 > 고랭지 온실 > 평난지 온실 순으로 감소하였다. 고랭지의 노지에서는 UV A와 UV B의 세기가 22.4, 3.02W·m-2로 평난지 노지의 19.9, 2.24W·m-2보다 각각 12.6, 34.8% 더 강했다. 감국의 초장은 평난지의 온실조건에서 가장 컸 으나 개화는 고랭지의 노지에서 가장 빨랐다. 감국 꽃 의 주요 색소인 카로티노이드는 고랭지의 온실에서 60.9OD·g-1으로 가장 많았다. 그러나 peroxidase 활성 은 평난지의 온실에서 자란 식물체에서 5.72unit·mg-1 protein 가장 높았다.
감국에 존재하는 항충치활성 물질 및 glucosyltransferase 저해 효과에 대한 정보를 얻고자 본 연구를 시도하였다. 감국 ethanol 추출물의 항충치활성을 S. mutans 및 구강세균에 대해 검색한 결과, S. mutans와 2종의 구강세균에 대하여 활성이 높게 나타났다. 감국 ethanol 추출물의 농도에 따라 GTase 저해효과는 78.4~92.3%의 범위로 나타나 농도가 증가할 수록 활성도 함께 증가하는 경향을 나타냈다. 감국을 ethanol로 추출, 용매분획하여 얻어진 활성성분을 silica gel adsorption chromatography, Sephadex LH-20 chromatography, TLC 그리고 HPLC 등의 방법으로 분석한 결과, 감국에 존재하는 항충치활성 물질은 약간의 극성을 띠는 중성물질로 분자량이 200-400정도 되는 저분자 물질인 것으로 확인되었다.
Background : Chrysanthemi Indici Flos (甘菊) is listed in 「The Korea Herbal Pharmacopoeia (KHP)」as the original plant of Chrysanthemum indicum L. C. indicum was one of the most representative medicinal plants in Asteraceae, Dried flowers of this plant have been valid chemical composition such as flavonoids, phenylpropanoids, terpenoids, and polysaccharides, possessing broad spectrum antibacterial, antiviral, antihypertensive and anti-oxidation functions. Meanwhile, C. indicum was a polymorphic species, its morphological characteristics showed great diversity due to the different geographical and environmental factors. For this reason, there was conducted to develop molecular markers to distinguishing these C. indicum with C. morifolium, C. zawadskii var. latilobum and Aster spathulifolius by using conventional polymerase chain reaction (PCR).
Methods and Results : In this study, In order to clearly identify origin of Chrysanthemi Indici Flos, these samples (C. indicum, C. morifolium, C. zawadskii var. latilobum and A. spathulifolius) were analyzed from five barcoding regions of chloroplast DNA (rbcL, matK, rpoB, atpF-atpH) and nuclear ribosomal DNA (ITS2) to evaluate the ability of discrimination for each barcoding region. Based on genetic distance, the percent of variable sites were provided the highest ITS2 value (56.9%), followed by atpF-atpH (48.18%), matK (27.2%), psbK (8.2%), and rbcL (2.9%). Comparative analysis based on the complete genome sequence of the petL-petG region INDEL (insertion/deletion) that the gene annotations were registered to the GenBank (accession number: JN-867592.1, NC-020092.1, MF-034027.1, NF-279514.1).
Conclusion : From the above results, we may suggest that the petL-petG region INDEL analysis were conducted for molecular authentication of four plants (C. indicum, C. morifolium, C. zawadskii var. latilobum and A. spathulifolius). The findings of results indicated that petL-petG region might be established INDEL analysis systems and hence were proved to be an effective tools for molecular evaluation and comparison of “Chrysanthemi Indici Flos” with other plants.
Background : Matricaria recutita and Dendranthema indicum are known to have effects such as antioxidant and antihypertensive effects, and they are used as processed food materials. Therefore, it is necessary to examine the possibilities as a natural material for cosmetics. This study was carried out to analyze the volatile flavor components of flowers during cultivation in order to examine the possibility of cosmetics using M. recutita and D. indicum.
Methods and Results : This experiment was carried out from April to October, 2017 at Unbong-eup (500 m above sea level) in Namwon, Jeollabuk-do. M. recutita and D. indicum were planted in late May. Then, flowers were collected in October and volatile flavor components were analyzed by Solid Phase Micro Extraction (SPME) method. Samples of flowers for component analysis were weighed in 0.4 g each and placed in a 20 ㎖ vial. GC/MS was used with Agilent Technologies 7890A/5975C (Agilent, USA). Column and carrier gas were DB-5MS (30 m × 0.25 ㎜ × 0.25 ㎛) and helium gas, respectively. As a result of the analysis of M. recutita flowers, we identified a total of 24 components. The major flavor components were mono terpenes and ketones. The main compounds were β-ocimene (24.08%), artemisia ketone (22.82%), γ-terpinene (16.65%), cis-3-hexenyl isovalerate (3.92%), artemisiatriene (3.72%) and O-cymene (3.44%). As a result of analyzing the composition of D. indicum flower, we identified 33 kinds of ingredients. The major flavor components were monoterpenes and sesquiterpenes. The main compounds were sabinene (19.67%), trans-oiperitol (13.22%), α-phellandrene (10.05%), myrcene (7.53%), cineole (4.36%), α-terpinene (3.60%) and trans-ocimene (3.57%).
Conclusion : We identified 24 flavor components and 33 flavor components in flower of M. recutita and D. indicum, respectively. Eight of the identified or estimated compounds were common to both samples. The reason why the flavor components kinds of M. recutita flowers were few was thought to be due to the influence of collection time. The main flowering period of M. recutita and D. indicum were May and October, respectively, but the use of the samples collected in October seemed to have influenced.
Background : Chrysanthemum indicum L. is used in medicine, for bee, ornamental as multivoltinism plant resources. Since ancient times, Chrysanthemum indicum L. had been widely used as drug and tea. To diversity and increase the demand of Chrysanthemum, it is breeding for medicinal herb, tea and ornamental flower. Therefore size and shape of plant, color of flower have been used for cross breeding. CI2013 lines collected from Jeju have been used for landscape and ornament. and CI2010 lines collected from Sancheong have been used for medicinal herb tea. As a result, many new lines were developed. Therefore it was carried out to use as a date for new varieties through the characterization of good lines. Methods and Results : 7 lines were investigated and these plants were forstered and cultivated from Gyeongsangnam-do Agricultural Research & Extension Services (Hamyang). Breeding Lines were CI201001, CI201002, CI201005, CI201006, CI201303, CI201307, CI201401. The shape of CI201303 and CI201307 were round, plant height is low and width is wide, it may be used for landscape and ornament. As the size of flower in CI201005 was big, it may be used for medicinal herb tea. Conclusion : CI201303 and CI201307 lines were good for landscape and ornament because of round shape. CI201005 line was good for medicinal herb tea because of big flower.
Background: Planting vigorous cuttings that quickly develop shoots and roots is essential to the biological and economic success of producing medicinal flowers. The present study aimed to evaluate the effect of storage temperature and duration on seedling capacity in the propagation of Chrysanthemum indicum L. and to investigate the effect of rooting media on the growth of C. indicum L. after cutting.Methods and Results:Returning cuttings to supplemental cold storage (2.0 ± 1.0°C) may extend duration of cutting viability 6 weeks, returning cuttings to supplemental warm storage (25.0 ± 1.0°C) is not recommended. The treatment of the growing media experiments, which were conducted in the 2014 planting seasons, included sawdust, river sand, topsoil + sawdust, topsoil + poultry manure, sawdust + river sand, river sand + poultry manure, topsoil + river sand + poultry manure, topsoil + poultry manure + river sand + sawdust. Result indicated that the topsoil + poultry manure media performed best and supported the highest number of branches (3.47), branch length (26.39), and number of leaves (88.63).Conclusions:The results of the present study suggest that cold storage and the topsoil + poultry manure growth media was superior in supporting the early establishment of C. indicum cutting, this result will have a tremendous influence on propagation of this species.
Background : Chrysanthemum indicum L. is used in medicine, for bee, ornamental as multivoltinism plant resources. The purpose of this study was to evaluate methods of rooting promotion to make high quality medicinal plants, thereby increasing farm income. This study investigated the effects of different cuttage period, extraction site, shading degree, nursery tray for promoting cutting slips rooting in Chrysanthemum indicum L. Methods and Results : It is used New variety "Gamkuk 1" that were forstered in Gyeongsangnam-do Agricultural Research & Extension Services. Method of application with cuttage period were conducted under five condition; Around the middle of April, Toward the end of April, Early in May, Around the middle of May, Toward the end of May. Early in May plant length was longer than other cuttage period as 41.2%, 83.7% and are significant at significance level 0.05. Method of application with extraction site were conducted under three condition; 1 - 3 node, 4 - 6 node, 7 - 9 node. Leaf number in 1 - 3 node was more than other extration site as 19.4%, 33.6%. Also root length in 1 - 3 was more than other extration site as 10.5%, 23.2%. Method of application with shading degree were conducted under three condition; 50%, 70%, 90%. Plant length in 50 - 70% was longer than 90% shading degree as 23.8%, 24.2%. Also shading degree 50 - 70% had many root length and root number. Method of application with nursery tray were conducted under five condition; 72, 105, 128, 162, 200 tray. Plant length in 105, 128 tray was longer as 8.5 - 35.3% than other nursery tray and are significant at significance level 0.05. Conclusion : According to the results, Early in May, extraction site 1 - 3 node, shading degree 50 - 70%, nusery tray 105, 128 showed the highest growth on cutting slips root promotion in Chrysanthemum indicum L.
Background: Chrysanthemum indicum L. is used in medicine, for bee, ornamental as multivoltinism plant resources. Since ancient times, Chrysanthemum indicum L. had been widely used as drug and tea. From 2000, breeding began and the three varieties "Manhyang", "Geumhyang", Gamro" were developed so far. As demand has increased, growth traits of "Wonhyang" has been tested and fostered to report growth characteristics of "Wonhyang". Methods and Results: It is used New variety "Wonhyang" that were forstered in Gyeongsangnam-do Agricultural Research & Extension Services. After going through selection, proliferation and characteristics of CI0901, production performance test carried out in 2012~2013. As a result, it had named as "Gamkuk 4" in high quality, quantity and of strong in the disease. Local adaptability trial test carried out in 2014~2015. Conclusion: "Wonhyang" was a good growth in plant height, lengths of branches. Leaf color is light green, flower color is light yellow. Disease and pest resistance of "Wonhyang" is relatively better than "Manhyang". Since plant height is small and plant width is big, "Wonhyang" is available for medicine, food and landscaping. As a result of local adaptability trial test, Yield(131.1㎏/10a) of three region Hamyang,Hapcheon,Changnyeongis 10% higher than "Manhyang". Based on the results of this study, "Wonhyang" was to be registered for a new breed through the breed fostering council.
