        Golden apple snails(GAS) are native to warm regions such as Central and South America and Southeast Asia, and were first introduced as a high-protein food. GAS are omnivorous and have a habit of eating plants submerged in water, so they have been used for eco-friendly weed control in rice fields since 1992. When the GAS was first introduced, it was thought that it would be impossible to overwinter in Korea. However after 2000, overwintering individuals were founded and damage to rice occurred and the development of means to control GAS has been required. In this study, we tested the effectiveness of an eco-friendly pest control agent using Styrax japonicus that grow naturally in Korea. As a result of exposing GAS to S. japonicus fruit powder, a 100% molluscicidal effect was confirmed at 66.7ppm. To investigate the duration of effect, treatment was performed at the same concentration and molluscicidal effect of more than 90% was up to 3 days after treatment. The killing effect of each part of the S. japonicus was compared, and the seed extracts showed no killing effect at all concentrations, while the sarcocarp extracts showed a 100% killing effect up to 33.3ppm, and the fruit extracts showed a 100% killing effect up to 200ppm.

        Aster koraiensis Nakai (A. koraiensis) which has been used as a food and medicinal plant in the past, is valuable as functional food material. Therefore, the aim of this study was to determine the antioxidant properties and major phenolics of A. koraiensis extracts with different ethanol concentrations (0, 50, 70, and 100% aqueous ethanol solution). When ethanol concentration in the extraction solvent was increased, extraction yield decreased; 34.2, 23.2, 21.0, and 5.5% in 0, 50, 70, and 100% ethanolic extracts, respectively. Total phenolics content and antioxidant activities of extracts were increased in an ethanol concentration-dependant manner. The major phenolics in the extracts were chlorogenic acid (21.264~58.666 mg/g), isochlorogenic acid A (10.432~145.353 mg/g), and isochlorogenic acid C(0.239~13.148 mg/g), and these phenolic contents were higher in 70 and 100% ethanolic extracts than other extracts. Significant correlations were observed between ethanol concentration of extraction solvent, antioxidant properties, and major phenolics. These results indicated that the optimal ethanol concentration for extraction was 70%.

        본 연구는 황기 에탄올 추출물과 황기 생물전환 추출물의 기능성 화장품 소재로서 가능성 여부를 규명하고자 하였다. 항산화 효과를 확인하기 위해 폴리페놀과 플라보노이드 함량을 측정하였고, 세포실험을 통해 피부 세포에 대한 독성과 피부 세포 노화에 대한 연구를 수행하였다. 항산화 실험 결과, 폴리페놀, 플라보노이드 효과에서는 폴리페놀과 플라보노이드의 함량이 농도 의존적으로 증가하였고, 생물전환 추출물에 비해 에탄올 추출물에서 높은 것으로 확인되었다. 황기 추출물과 생물전환된 황기추출물이 HDF 세포에서 농도 의존적으로 MMP1 발현 억제 효과가 증가되는 것을 확인하였다. 따라서 본 연구는 황기 추출물과 황기 생물전환 추출물의 기능성 화장품 소재로 사용 시 항산화와 피부 노화 예방 관점에서의 기능성 화장품 소재로 매우 유용하게 활용될 수 있을 것으로 사료된다.

        Barley’s nutritional value as a health food is increasing due to its excellent nutritional functionality. In this study, the levels of β-glucan, total polyphenols, and total flavonoids were analyzed in the ethanol extracts of different barley cultivars (Hinchalssal, Heuksoojeongchal, Betaone, Ganghochung, and Saechalssal). Also, the free radical scavenging abilities of 2,2-diphenyl-1-picrylhydrazil (DPPH) and 2,2’-azino-bis-3-ethylbenzo-thiaxoline-6-sulfonic acid (ABTS) were measured to determine their antioxidant activity. The results confirmed that Betaone extract contained highly activefunctional components and exhibitedantioxidant activity. Next, we evaluated the hepatoprotective and inhibitory effects of reactive oxygen species (ROS) generated by barley ethanol extracts after inducing oxidative stress with tert-butyl hydroperoxide (tBHP) in HepG2 cells. Hinchalssal and Saechalssal extracts showed the most significant cytoprotective effect and also reduced ROS production significantly. These results suggest that Hinchalssal, Saechalssal, and Betaone represent potential natural antioxidant and hepatoprotective agents.

        This study investigated the preventive effect of Dendropanax morbifera leaf extracts (DMLEs) drew out with water, 30, 50 and 70% ethanol against alcohol-induced hepatotoxicity in vitro and vivo. The 3-(4,5-dimethylthiazol-2-yl)-2,5-dipenyltetrazolium bromide (MTT) assay was performed to assess cytotoxic activity of DMLEs, and the half-maximal inhibitory concentration (IC50) ranged between 1.11 and 2.08 mg/mL on Hep3B cells. In preventing ethanol-induced-hepatotoxicity on Hep3B cells, 30 and 50% ethanol DMLEs were significantly effective compared to the 5% ethanol treatment control. In addition, the 30% ethanol DMLE was orally provided to rats 30 min prior to the administration of ethanol (3 mL/kg body weight). At 1, 3 and 5 h after ethanol treatment, the plasma levels of ethanol and acetaldehyde were determined. The 30% ethanol DMLE effectively decreased the plasma ethanol levels during 5 h but increased the plasma acetaldehyde levels until 3 h and then significantly decreased at 5 h, as compared to the control. These results indicate that the 30% ethanol DMLE possesses a potent preventive effect against ethanol-induced liver toxicity in Sprague-Dawley rats.

        본 연구는 박하 추출물을 통해 독성검사 및 항산화, 항염증과 같은 생리활성을 평가하고자 하였다. 박하 추출은 열수 및 80% 에탄올로 추출하였으며, RAW 264.7 세포를 통해 MTT를 통해 세포 생존율을 평가하였다. 또한, LPS로 유도한 RAW 264.7 세포를 통해 활성산소(ROS), 산화질소(NO), 류코트리엔 B4(LTB4), 항염 또는 염증성 사이토카인(IL-1β, IL-6, TNF-α, IL-10) 등을 ELISA 및 Luminex로 측정하였다. 그 결과, 박하 추출물은 열수 및 80% 에탄올 추출물에서 세포독성이 없었으며, 80% 에탄올 추출물의 100 ㎍/㎖ 농도에서 활성산소의 생성을 억제하였다. 또한, NO, LTB4, 염증 또는 항염증성 사이토카인의 생성을 모든 추출물에서 농도 의존적 효능을 나타냈다. 즉, 박하 추출물은 세포 독성 없이 항산화 및 항염증 활성을 나타내는 생리학적 효능이 나타났다. 이와 같은 결과는 본 연구와 관련된 증상을 개선하기 위한 새로운 건강식품 및 치료제의 원료로 개발될 수 있다.

        In this study, 23 ethanolic extracts from 20 medicinal plants were evaluated for biological activities. Results revealed that of 23 samples, seven samples have demonstrated good antimicrobial activity. Minimum inhibitory concentrations were 0.4-2.0 mg/mL, while minimum bactericidal concentrations were mostly high 0.8- 2.0 mg/mL for selected seven samples. Five samples revealed > 70 mg gallic acid equivalent (GAE)/g of total phenolic contents. Among test samples, six samples exhibited > 80% inhibition of the 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical and only two samples exhibited > 80% inhibition of 2,2’-Azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) radicals. A total of five test samples revealed Trolox equivalent antioxidant capacity more than 1000 μm/ g. The MTT assay indicated that eight test samples exhibited > 90% viability of murine macrophage cells (RAW 264.7) at 250 μg/mL and suppressed iNOS mRNA expression at transcriptional level when stimulated by lipopolysaccharide (LPS). Some medicinal plants revealed promising results, and so they have prospective for further more inclusive studies.

        장어의 과산화 억제를 위해 월계수잎, 총백, 헛개열매, 녹차를 시판 소주(30°)를 용매로 하여 추출한 것을 시료로 사용하였다. 장어 250 g에 5 g의 주정 추출물을 장어 전체에 고루 바른 후 35℃ 15시간 열풍건조한 후 실험에 사용하였다. 주정 추출물의 항산화 활성은 DPPH 소거 활성 실험, 반건조 장어의 산가, 과산화물가, 리놀레산 및 장어유 과산화 지연반응을 통해 확인하였다. DPPH 소거 활성이 가장 높았던 것은 녹차 추출물이었으며, 다음으로 활성이 높았던 것은 월계수잎, 헛개열매 추출물이었다(p<0.05). 21일 동안 냉장보관한 장어의 산가 측정에서 헛개열매 추출물로 처리한 경우, 산가가 가장 낮았으며, 다음으로 녹차 추출물로 처리한 경우의 산가가 낮았다. 유리지방산 생성 억제 효과와 유사하게 헛개열매, 녹차 추출물이 장어의 열풍건조 동안 초기 과산화물 생성을 억제하고, 냉장보관 21일 동안 반건조 장어의 과산화물의 생성을 억제하는 것으로 나타났다. 4종의 주정 추출물들은 반응 20일 동안 리놀레산의 과산화를 억제하는 효과가 있는 것으로 나타났다. 반응 20일 동안 장어유의 과산화를 지연시킨 효과가 높았던 것은 헛개열매와 녹차 추출물이었으며, 다른 추출물에 비해 상대적으로 장기간 안정적으로 장어유의 과산화를 억제하는 것으로 판단되었다. 본 연구의 결과를 종합해 볼 때, 장어의 건조 전 전처리에 헛개열매와 녹차 주정(30° 소주)추출물을 적용하는 것은 장어의 건조 및 반건조 장어의 냉장보관 중의 과산화를 지연시키는데 효과가 있는 것으로 판단되었다.

        스피룰리나는 지구상의 가장 오래된 조류중 하나이며 피코시아닌, 토코페롤, 베타카로틴을 함유하고 있어 노화를 방지한다고 보고된 바 있다. 본 연구에서는 재배방법이 다른 스피룰리나 에탄올추출물의 UVB로 유도된 활성산소종(reactive oxygen species, ROS)의 저해와 항산화효과를 확인하였다. 실내배양 스피룰리나 에탄올추출물(ICAE)과 야외배양 스피룰리나 에탄올추출물(OCAE) 0.1, 0.5, 1 ㎎/㎖의 DPPH 라디칼 소거능과 SOD 유사 활성을 측정하여 항산화효과를 확인하였다. UVB로 유도된 ROS의 저해 효과는 제브라피쉬 배아와 HaCaT 세포를 이용하여 확인하였다. ICAE와 OCAE 0, 0.01, 0.05, 0.1, 0.5, 1 ㎎/㎖을 제브라피쉬 배아와 HaCaT 세포에 처리하고 UVB로 ROS를 유도하였고 DCFH-DA로 염색하여 세포내 ROS의 양을 검출하였다. 항산화효과를 측정한 결과 양성대조군인 ascorbic acid의 DPPH 라디칼 소거능은 73%, SOD 유사 활성은 86%로 나타났다. ICAE와 OCAE 1 ㎎/㎖의 농도에서 각각 43, 57%의 DPPH 라디칼 소거능을 나타내었고, 20, 19%의 SOD 유사 활성을 나타내어 양성대조군인 ascorbic acid보다 낮았지만 유의적인 항산화효과가 있었다. UVB로 유도된 제브라피쉬 배아와 HaCaT 세포의 ROS가 음성대조군보다 증가하였고, ICAE와 OCAE 처리된 군은 농도 의존적으로 UVB만 조사된 양성대조군 보다 ROS가 감소하였다. 본 연구의 결과는 스피룰리나 에탄올추출물이 피부보호용 화장품 소재로 사용가치가 있음을 시사한다.

        To evaluate the anti-oxidant and anti-tumor potential of the green pepper (Capsicum annuum L. cv. DangZo), total polyphenol content, radical scavenging activities and anti-tumor properties were measured. The total polyphenol content of the 70% ethanol extracts from green pepper (Capsicum annuum L. cv. DangZo) was 30.29 mg gallic acid equivalent/g extract. The DPPH radical and hydroxyl radical scavenging activities of 70% ethanol extracts of green pepper (Capsicum annuum L. cv. DangZo) were documented at 2.87 and 10.55, respectively. For α-glucosidase and α-amylase inhibitory activity, 70% ethanol extracts of green pepper (Capsicum annuum L. cv. DangZo) were documented at 35.67% and 58.41% respectively. The green pepper (Capsicum annuum L. cv. DangZo) demonstrated greater capability in terms of anti-neoplastic activity vis-a-vis colon cancer cell lines when compared to other cancer cell lines.s. er (Capsicum annuum L. cv. DangZo) higher activities of anticancer activities on colon cancer cell lines compared to other cancer cell lines.

        This study was conducted to certify the effect of aqueous extracts from fifty medicinal plants on the activities of alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) in vitro. Each aqueous extract was prepared by combining one-part medicinal plants with twenty-parts distilled water at 80°C for 8 h. Among the fifty medicinal plants, Allium sativum L. and Cinnamomum cassia Presl were regarded as an effective anti-hangover substance. Allium sativum L. extract increased ALDH activity more than 2 times compared with ADH activity, enhancing the acetaldehyde degradation. Cinnamomum cassia Presl extract dramatically inhibited ADH activity compared with ALDH activity, thus potently decreasing the acetaldehyde formation. ADH and ALDH activities were proportionally inhibited according to the increased concentration of Cinnamomum cassia Presl extract. The aqueous extract of Cinnamomum cassia Presl at a concentration of 45.33 μg/mL inhibited ADH activity by 52.8% and ALDH activity by 11.0%.

        Caesalpinia sappan L. is an oriental medicinal plant distributed in the Asia Pacific region including India, Malaysia, and China. The dried heartwood of Caesalpinia sappan has been traditionally used as an analgesic and anti-inflammatory drug. In this study, the effects of extract methods of C. sappan on contents of total polyphenols and flavonoids, antioxidant activity, and cytotoxic activity were evaluated. As a result, hot water extract from C. sappan (CSWE) significantly exhibited contents of total polyphenols and flavonoids (22.6 mg GAE/g and 14.5 mg QE/g) higher than 70% ethanol extract (CSEE) (17.6 mg GAE/g and 13.2 mg QE/g). However, CSEE showed greater antioxidant activity than CSWE in both DPPH and ABTS. Also, the cytotoxicity of C. sappan against three kinds of cancer cell lines was higher in CSEE than in CSWE. These results show that ethanol extract is a better extract method than hot water method to maintain antioxidant and anti-cancer activities.

        Fludarabine, a chain terminating anti-cancer drug, is a purine analogue that causes DNA strand breaks in normal cells. In this study, we determined if A. melanocarpa and Korean red ginseng extract mixture reduce cytotoxicity of fludarabine. Treatment of HaCaT cells with 10 μM of fludarabine for 24 hours decreased cell viability and increased DNA strand breaks. Treatment of A. melanocarpa and Korean red ginseng extract mixture for 24 hours increased cell viability as compared with single extract treatment. The protective effect of these extracts on cell activity increased in a concentration-dependent manner. DNA strand breaks induced by fludarabine decreased as concentration of extract mixture increased. p-H2AX level, a marker of DNA strand breakage, decreased depending on the concentration of extract mixture. The effect of mixed extract of A. melanocarpa and Korean red ginseng on DNA damage is due to the anti-oxidative effect of A. melanocarpa and signal transmission through glucocorticoid receptor upon binding of saponin of Korean red ginseng.

        Although Semisulcospira libertina is generally regarded as a supplement for the alleviation of alcohol hangover, little is known about its effects on cell metabolism. Therefore, this study was conducted to analyze the constituents of the extracts prepared using different extraction methods and to compare their biochemical properties. The amino acid contents were found to be much higher in acidic and enzymatic hydrolysates than hot water extracts from S. libertina. DPPH radical scavenging activities in acidic and enzymatic hydrolysates were higher than those of hot water extracts. Three types of S. libertina hydrolysate was added to HepG2 cells damaged by acetaminophen (AAP), after which the survival rate of HepG2 cell were measured. In addition, lactate dehydrogenase (LDH) activities in the culture media were evaluated. The survival rates of HepG2 cells were 77.0±4.3% and 81.5±1.3% at 3 h and 5h enzymatic hydrolysates, respectively. These cell survival rates were higher compared to those of the negative control group (67.8±4.3%) treated only with acetaminophen. Cellular toxicities induced by treatment with AAP were also significantly alleviated in response to treatment with the extracts of S. libertina. In addition, the activities of 2 key enzymes that metabolize ethanol, alcohol dehydrogenase and aldehyde dehydrogenase, were upregulated by 4.7- and 2.7-fold respectively in response to treatment with a 3 h enzymatic hydrolysate of S. libertina. Taken together, these results provide biochemical evidence of the method by which S. libertina exerts its biological functions, including the alleviation of alcohol hangover and the protection of liver cells against toxic insults.

        본 연구에서는 뱀딸기풀의 열수추출물과 에탄올추출물간의 항산화 활성 비교를 위해 25~500μg/mL의 뱀딸기풀의 농도를 이용하여 in vitro 실험으로 관찰하였다. DPPH radical 소거능은 열수추출물 (16.23~89.54%)과 에탄올추출물(6.85~89.41%)에서 농도 의존적으로 높은 활성을 나타내었으며, ABTS radical 소거능을 비교한 결과는 열수추출물에서 15.48~93.49%, 에탄올추출물에서는 7.42~92.81%로 농도 의존적으로 높은 소거 활성을 나타내었다. 이 두 결과를 미루어 열수추출물은 에탄올추출물에 비해 유의적 으로 더 높은 radical 소거능을 나타내는 것을 확인할 수 있었다. 환원력은 열수추출물에서 0.167~1.023, 에탄올 추출물에서 0.161~0.783의 활성을 나타내었다. NO radical 소거능은 열수추출물에서 16.24~ 67.93%, 에탄올추출물에서 12.46~70.78%의 활성을 나타내었으며 특히 75~200μg/mL 농도에서는 열수추 출물이 에탄올추출물에 비해 유의적으로 더 높은 활성을 나타내었다. 뱀딸기풀 추출물의 IC50값을 비교 한 결과 DPPH, ABTS radical 및 NO radical에서 각각 열수추출물에서 90.6μg/mL, 101.3μg/mL 및 147.0μg/mL으로 나타났고, 에탄올추출물의 경우에는 164.7μg/mL, 151.9μg/mL 및 235.6μg/mL으로 나타 났다. DPPH, ABTS radical 및 NO radical 모두에서 에탄올추출물이 더 높은 IC50값을 보였다. 총 페놀 함량은 500μg/mL, 1000μg/mL의 농도에서 열수추출물에서 각각 42.3mg/g, 91.6mg/g, 에탄올추출물에 서 39.6mg/g, 82.3mg/g의 함량을 나타내었다. 플라보노이드 함량에서는 500μg/mL, 1000μg/mL의 농 도에서 열수추출물에서 각각 26.3mg/g, 31.9mg/g, 에탄올추출물에서 22.3mg/g, 27.7mg/g의 함량으로 열수추출물이 유의적으로 더 높은 함량을 나타내었다. 이러한 결과로 미루어 뱀딸기풀의 용매에 따른 추 출물은 항산화의 효과를 나타내는 실험 모두에서 높은 활성을 나타내었으므로 뱀딸기풀은 효과적인 항산 화 효능을 가진 천연식품으로 이용되어 질 수 있으리라 기대되어진다.

        본 연구는 식용으로 사용하는 곰취의 국내 산지별(태백, 정선, 양구, 평창, 인제) 항산화 효과를 평가하고, 곰취의 유효성분 분리를 통해 항산화 효능평가를 수행함으로써 산 지별 곰취의 항산화 효능에 대한 탐색 및 소재 개발 가능 성을 검토하고자 하였다. 그 결과, 인제산 곰취 50% EtOH 추출물의 농도에 따라 DPPH 라디칼 소거능과 ABTS 라디칼 소거능이 각각 71.77%, 98.95%로 높은 항 산화 활성이 관찰되었다. 또한, ORAC activity assay kit를 이용한 항산화 활성 측정 결과에서도 인제산 곰취 50% EtOH 추출물의 농도에 따라 높은 항산화 활성이 관찰되었 고, 100 μg/mL의 농도에서 290.19±5.79 μM TE/g으로 높 은 항산화 활성을 나타내었다. 산지별 가운데 가장 높은 항산화 활성을 보인 인제산 곰취 50% EtOH 추출물로부 터 유효물질 5개(chlorogenic acid (1), neochlorogenic acid (2), isochlorogenic acid A (3), isochlorogenic acid B (4), isochlorogenic acid C (5))를 분리하여 곰취 compound의 항산화 활성을 분석한 결과, 인제산 곰취 50% EtOH 추출 물과 유사하게 compound 4, 5 (isochlorogenic acid B, isochlorogenic acid C)의 농도에 따라 높은 DPPH 라디칼 소거능(89.76%, 92.72%)과 ABTS 라디칼 소거능(99.09%,98.95%)이 관찰되었고, ORAC assay에서도 compound 4, 5에서 각각 362.56±5.91 μM TE/g, 362.35±1.92 μM TE/g 으로 높은 항산화 활성을 나타내었다. 이러한 결과들을 통해 산지별 곰취 50% EtOH 추출물 가운데 인제산 추출물이 높 은 항산화 활성을 보유하는 것을 확인할 수 있었으며, 이러 한 인제산 곰취 추출물에는 높은 항산화 활성 성분(compound 4: isochlorogenic acid B, compound 5: isochlorogenic acid C)이 함유되어 있음을 확인할 수 있었다. 본 결과는 식용으로 사용되고 있는 곰취의 항산화 활성을 보유한 기 능성 소재로서의 활용을 위한 기초자료로 이용될 수 있을 것으로 판단된다.

        This study was conducted to investigate the antioxidant and anti-obesity activity of ethanol extracts from fermented Arctium lappa L. Arctium lappa fermented twice with Phellinus linteus (2nd FA) showed more DPPH radical scavenging activity than non-fermented Arctium lappa (NFA), and Arctium lappa fermented once (1st FA) at a concentration of 62.5 ppm. The 2nd FA showed the highest level of ABTS radical scavenging activity at concentration range of 31.5~125 ppm. The ABTS free radical scavenging activities of 1st FA and 2nd FA were similar to that of BHA, a synthetic antioxidant, at a concentration of 250 ppm. The total polyphenol content of 2nd FA was higher than those of NFA and 1st FA. The flavonoid content was significantly increased in 1st FA and 2nd FA than NFA. During adipocyte differentiation, lipid accumulation in 3T3-L1 cells was significantly decreased in the order of NFA, 1st FA, and 2nd FA at all concentrations. In conclusion, the antioxidant and anti-obesity activities of A. lappa were increased depending on the degree of fermentation. It is suggested that fermented Arctium lappa, especially 2nd FA, could be used as a natural ingredient for functional foods and medicine.
