세계적으로 친환경 해충 방제를 위해 식물 추출물을 사용하는 연구가 화학농약에 대한 의존도를 줄이기 위해 진행되고 있다. 제충국 식물은 살충제용 피레트린의 원료로 널리 알려져 있지만, 한국에서 재배된 식물의 추출물과 효능에 대한 연구는 제한적이다. 본 연구는 한국에서 재배된 본 연구는 한국에서 재배된 두 가지 제충국 품종에서 피레트린을 추출하는 방법을 비교하였다. 또한, 분리된 피레트린 함유 추출물의 주요 식물 해충 에 대한 효능을 조사하였다. 피레트린 함량은 프랑스에서 수입한 '계통' 품종에 비해 '달마시안' 품종에서 통계적으로 유의하게 높았다. 따라서, 달마 시안 품종의 피레트린 추출물을 이용하여 본 연구에 사용하였다. 추출된 피레트린의 양은 추출에 사용된 에탄올 농도가 증가함에 따라 증가하였으 며, 95% 에탄올을 사용했을 때 가장 높은 수준(6,064 ppm/100 g 꽃 건물중)이었다. 총 피레트린 함량과 피레트린 I/II 비율은 만개기에 수확한 꽃 에서 가장 높았다. 추출물의 in vitro 생물검정에서 복숭아혹진딧물의 LC50 값은 34 ppm, 배추좀나방 유충은 69 ppm, 뿌리혹선충 유충은 0.1 ppm이었다. 이러한 결과는 한국에서 재배된 달마시안 꽃의 에탄올 추출물이 식물 해충 방제를 위한 천연 살충제 개발에 유용할 수 있음을 나타냈 다. 본 연구는 한국에서 재배된 T. cinerariifolium var. 달마시안 꽃 추출물이 국내에서 식물 해충을 효과적이고 친환경적으로 방제하는 데 잠재력이 있음을 보였다.
This study evaluated the changes in the quality characteristics of extracts (Quantum Energy) from Rehmannia glutinosa, ginseng, balloonflower, Poria cocos, and Adenophora stricta. The extracts were prepared through a six-day aging process, during which the pH decreased with a longer aging period while the viscosity and oBrix values were increased. The color difference (E) of the extracts ranged from 0.27 to 0.49 for all samples, indicating minimal color change with aging. The free sugar content was highest for fructose and glucose, while lactose was not detected. Furthermore, the total free sugar content ranged from 24.518 to 27.646 g/100 g, generally decreasing with the aging time. Benzo[a]pyrene in the extracts ranged from 0.040 to 0.074 g/kg, which is below the limit of quantification. In addition, all extracts were free of total aerobic bacteria, Escherichia coli, and coliforms, confirming that the aging method used in this study was microbiologically safe.
Helicobacter pylori are known as a causative agent of gastritis, gastric duodenum and peptic ulcer, and gastric cancer, and multiple drug use is associated with various side effects in patients. The discovery of antibacterial substances against H. pylori from Korean resource plants is an important substitute for antibiotics. 52 species of Korean resource plants were collected and extracted with 50% ethanol, and antibacterial activity against H. pylori was measured using the disk diffusion method. The toxicity of plant extracts to human gastric adenocarcinoma(AGS) cells was measured by MTT assay, and the level of IL-8 secreted when gastric epithelial cells were inoculated with H. pylori was measured. As a result of measuring the antibacterial activity of H. pylori, antibacterial activity was confirmed in 38 plant extracts. The plant species with the strongest antibacterial activity were Chrysanthemum indicum, Rheum rhabarbarum, Patrinia scabiosaefolia and Petasites japonicus. C. indicum was not cytotoxic to H. pyroli-infected AGS cells and showed anti-inflammatory effects. This study's results can be used to develop healthy, functional foods and medical materials.
Calystegia pubescens Lindl. (C. pubesens)는 전통적으로 이뇨, 피로 및 혈당 강하 효과로 사용되어 온 한국 토종 식물이다. C. pubesens는 이전 연구에서 항산화 및 미백 효과를 보였지만 항균 및 항염증 특성에 대한 연구가 제한적이고 기능성 소재로서의 입증이 부족하다. 본 연구에서는 70% 에 탄올로 추출한 C. pubesens의 잎(CPL)과 줄기(CPS) 추출물의 항균, 항산화 및 항염증 효과를 조사했다. CPL은 CPS보다 여드름균, 황색포도상구균, 대장균과 녹농균에 대해 우수한 항균 효과를 보였다. 또한 CPL은 CPS보다 DPPH 및 ABTS+ 라디칼 소거 분석에서 더 높은 항산화 활성을 나타냈다. 따라서 CPL 은 CPS에 비해 우수한 항균 및 항산화 효과를 보였다. 이러한 결과를 바탕으로 RAW264.7 대식세포를 이용한 CPL의 항염증 효과를 확인하기 위한 추가 연구를 진행하였다. 지질다당류(LPS) 자극에 의해 생 성된 산화질소(NO)는 CPL에 의해 감소되었다. 또한, LPS에 의해 증가된 iNOS의 단백질 및 mRNA 발 현은 CPL에 의해 감소되었으며, 이는 NO 생산과 동일한 경향을 나타내었다. 결론적으로 본 연구는 CPL이 CPS에 비해 우수한 항균 및 항산화 효과를 가지고 있음을 확인하였으며, LPS에 의해 유도된 iNOS 발현 및 NO 생성 억제를 통해 CPL의 항염증 효과를 입증했다. 본 연구 결과를 바탕으로 CPL의 기능성 소재로서의 가능성을 제시한다.
본 연구는 국내산 왕벚나무 수피를 건강기능식품 소재 로서 활용 가능성을 확인하기 위하여 용매별 추출물에 대 한 특성 분석과 항산화 활성을 평가하였다. 용매별 소재 의 아미노산과 지방산 분석을 통하여 성분함량 및 조성 차이를 확인하였고, 항산화 활성 비교를 위해 총 폴리페 놀 함량, 총 플라보노이드 함량, DPPH 라디칼 소거능, ABTS 라디칼 소거능, FRAP 활성을 측정 및 비교 후 상 관관계를 분석하였다. 아미노산 분석 결과, 열수 추출물, 30% 에탄올 추출물, 70% 에탄올 추출물에서 13종의 아미 노산이 검출되었고, 주요 아미노산으로 aspartic acid, arginine, proline으로 분석되었으며, 총 아미노산 함량은 각 각 0.17%, 0.16%, 0.09%로 확인되었다. 지방산 분석 결과, 포화지방산(SFA)이 열수 추출물, 30% 에탄올 추출물, 70% 에탄올 추출물에서 62.7-66.7%로 높은 조성비를 보였고, 확인된 지방산은 열수 추출물에서 behenic acid (C22:0)을 제외한 13종의 지방산이 검출되었으며, 30% 에탄올 추출 물과 70% 에탄올 추출물에서 14종 지방산이 검출되었다. 주요 지방산은 열수 추출물, 30% 에탄올 추출물, 70% 에 탄올 추출물에서 palmitic acid (C16:0)로 확인되었고, 총 지방산 함량은 각각 0.66 mg/g, 2.46 mg/g, 10.04 mg/g으 로 70% 에탄올 추출물에서 가장 높았다. 총 플라보노이 드와 총 폴리페놀 함량은 열수 추출물, 30% 에탄올 추출 물, 70% 에탄올 추출물에서 각각 727.70-769.87 mg QE/ g, 309.24-348.09 mg GAE/g이었다. 항산화 활성 측정 결 과 열수 추출물, 30% 에탄올 추출물, 70% 에탄올 추출물 에서 DPPH 라디칼 소거능은 0.05-1 mg/mL 농도에서 각 각 14.42-81.77%, 16.85-83.38%, 19.33-81.12%의 결과를 보였고, ABTS 라디칼 소거능은 0.05-1 mg/mL 농도에서 각각 6.32-89.55%, 16.14-70.61%, 8.99-92.08%의 결과를 보 였으며, FARP 활성은 0.05-0.5 mg/mL 농도에서 각각 0.32- 2.91, 0.07-0.61, 0.39-3.43으로 나타나 농도 의존적으로 항 산화 효과를 증가시키는 것을 확인하였다. 용매별 추출물의 총 플라보노이드 함량, 총 폴리페놀 함량과 항산화 활 성 간의 상관관계를 분석한 결과, 높은 양(+)의 상관성을 보였다(P<0.01). 본 연구결과를 통해 왕벚나무 수피 추출 물을 이용한 건강기능식품 소재 개발의 기초 자료로 활용 될 수 있을 것으로 사료된다.
Buckwheat leaves have the best antioxidant properties, including flavonoids, rutin, and 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging ability compared to common buckwheat and tartary buckwheat. The total dietary fiber content of the powder extracted by mixing buckwheat leaves and sea tangle was 30.5%. To manufacture bread containing mixed buckwheat flour, 10, 20, and 30% buckwheat flour was mixed in to produce buckwheat bread. The bread’s characteristics underwent significant changes with varying levels of buckwheat flour. As the buckwheat content increased, the size of the bread decreased, but its hardness, gumminess, and chewiness tended to increase. The sensory characteristics of the bread were not improved due to the addition of gluten in the case of bread mixed with 20% buckwheat flour. When the buckwheat flour content was mixed at 20%, and the mixed extract of buckwheat leaves and sea tangle were added at 0.4% and 1.0%, there was no significant difference in the appearance of the buckwheat bread. Still, regarding sensory properties, the preference was higher in the sample with 1.0% added buckwheat flour.
Green pesticides, derived from natural sources, have gained wider attention as an alternative approach to synthetic pesticides in managing polyphagous pests like Spodoptera litura. In this study, the methanolic flower extract of Nyctanthes arbor-tristis (Mx-Na-t) was subjected to chemical screening, and major peak area derivatives 3-Hydroxy-1,2-dimethyl-4(1H)-pyridone (3H-dp) and Tyrosol (Ty-ol) were identified. The toxicity against S. litura larvae of Mx-Na-t (at 500 ppm) was highest in third instars (96.4%), and for 3H-dp and Ty-ol (at 5 ppm) in II instars (76.5% and 81.4%, respectively). The growth and development of S. litura larvae and pupae were significantly reduced by all three extract and phytochemical treatments. Fecundity rates also declined with the treatments, from 1,020 eggs (control) to 540 eggs (Mx-Na-t), 741 eggs (3H-dp), and 721 eggs (Ty-ol). The extract and its active constituents decreased adult emergence and slowed total larval development in a dosedependent manner. The major gut enzymes of S. litura decreased in young larvae (II instar) exposed to Mx-Na-t, 3H-dp, and Ty-ol. Fourth instar midgut tissues were severely damaged by Mx-Na-t (250 ppm), 3H-dp, and Ty-ol (2.5 ppm) treatments, which induced structural damage to the epithelial cells and gut lumen. The earthworm Eisenia foetida was used to test for non-target toxicity. Crude Mx-Na-t at 500 ppm (13% and 3%) and 3H-dp (9.3% and 2.1%) and Ty-ol (10.2% and 1.5%) at 5 ppm produced lower mortality than the synthetic chemical cypermethrin at 1.0 ppm (27% and 18%) in filter paper and artificial soil assays, respectively. In addition, there was no significant change in earthworm weights under all three phytochemical treatments compared to controls. Additionally, the in-silico predictions of BeeTox and ProTox II indicated little or no toxicity toward honey bees and other nontargets associated with 3H-dp and Ty-ol. Overall these phyto-chemicals offer an effective pest management strategy.
Bangladeshi medicinal plants (BMP) have a history of traditional use in treating chronic inflammatory diseases, but a BMP bark’s antioxidant and anti-inflammatory effects remain largely unexplored. This study assessed methanolic extracts’ antioxidant and anti-inflammatory properties from the bark of 15 medicinal plant species native to Bangladesh. The methanol extracts of BMP bark were evaluated for their total antioxidant activity and ability to counteract inflammation induced by lipopolysaccharide (LPS) in RAW 264.7 macrophages. Among the 15 bark extracts from BMP, Albizia odoratissima (A. odoratissima), Engelhardia spicata (E. spicata), and Shorea robusta (S. robusta) showed the highest total phenolic contents and total antioxidant capacity by effectively scavenging free radicals. In particular, these three bark extracts significantly reduced the mRNA expression of LPS-induced inflammatory cytokines and enzymes inducible by inflammation in macrophages. Also, the mRNA expression of NADPH oxidase 2 was significantly suppressed by the three bark extracts in LPS-induced RAW 264.7 macrophages. These results suggest that out of the 15 bark extracts obtained from medicinal plants in Bangladesh, the extracts from A. odoratissima, E. spicata, and S. robusta exhibit substantial total antioxidant capacity by efficiently scavenging free radicals and also inhibit LPS-induced inflammation in macrophages.
제충국(Tanacetum cineariaiaefolium), 데리스(Derris elliptica), 고삼(Sophora flavescens) 추출물은 다양한 해충을 방제하는데 사용되고 있다. 하지만, 국내에서 판매되고 있는 식물추춞물 자재는 유효성분의 표기가 없고, 살충농도와 살충시간에 대한 자료가 전무한 상황이다. 본 연구에서는 상 용화된 주요 식물추출물의 살충유효성분의 농도를 결정하고 복숭아혹진딧물에 대해 살충농도와 살충시간을 측정하였다. 식물추출물의 살충활성성분 인 pyrethrins, rotenone, matrine과 oxymatrine의 농도는 액체 크로마토그래피에서 표준물질을 활용하여 질량분석을 통해 측정하였다. 식물추출 물을 농도별로 희석하여 복숭아혹진딧물에 살포하여 살충력을 측정하였다. 표준화합물과 비교한 후 질량분석 및 결정했습니다. Myzus persicae에 대 한 lethal concentation과 lethal time을 조사했다. 살포 후 48시간 후 치사 농도(LC50)는 pyrethrins (20.4 ppm), roteone (34.1 ppm), matrine (29.6 ppm)였고, 100 ppm 살포한 LT50은 pyrethrins (13.4시간), rotenone (15.1시간), matrine (14.4시간)로 측정되었다. Kaplan-Meier 생존분 석 결과, 100 ppm에서 세 가지 식물 추출물의 LT50은 대조구인 화학 살충제인 Sulfoxaflor를 살포 처리구보다 유의하게 빨랐습니다. 본 결과는 복숭 아혹진딧물 방제를 위해 식물추출물의 제형화에 단일 또는 혼합 제제를 개발하는데 기준 살충농도와 살충시간을 제고하는데 의미가 있다.
We investigated the anti-inflammatory effects of shiitake mushroom and kelp (SMK) mixture extracts in lipopolysaccharide (LPS)-stimulated murine RAW 264.7 cells. Treatment of RAW 264.7 cells with LPS significantly increased NO (nitric oxide) production, pro-inflammatory cytokines (tumor necrosis factor (TNF)-α, interferon (IFN)-γ, interleukin (IL)-6, and IL-1β), and inflammation-related genes (COX-2 and inducible nitric oxide synthase (iNOS)). In cytotoxicity testing using RAW 264.7 cells, SMK mixture extracts in the range of 1-16 μg/mL did not inhibit cell proliferation. However, SMK mixture extracts significantly inhibited NO production in a dose-dependent manner (p<0.05). SMK treatment significantly decreased TNF-α, IL-6, IFN-γ, and IL-1β levels compared to the LPS group, and similarly, pro-inflammatory cytokine mRNA levels also decreased. SMK mixture extracts reduced the mRNA expression of COX-2 and iNOS in RAW 264.7 cells compared to LPS (p<0.05). The above results show that SMK mixture extracts suppressed the inflammatory response induced by LPS. In particular, the extracts were shown to regulate the inflammatory response by suppressing the expression of inflammatory cytokines and inflammation-related enzymes.
To improve usability of super sweet corn, extracts were prepared with hydrolytic enzyme and changes in physicochemical and antioxidant properties were analyzed. Soluble solids and reducing sugars contents were higher in all enzyme treatment groups than in the control. When enzyme treatment time increased, contents of soluble solids and reducing sugars were also increased. There was no significant difference in lightness between treatment groups, with redness showing the highest value in the control and yellowness showing the highest value in the invertase treatment group. Free sugar content in the control was the lowest. However free sugar content in the enzyme combination treatment group was increased by more than four times compared to that in the control. Contents of ascorbic acid, flavonoids and polyphenols were higher in the enzyme treatment group than in the control. In particular, the enzyme combination treatment group showed the highest content. DPPH and ABTS radical scavenging abilities were significantly higher in all enzyme treatment groups than in the control. Radical scavenging abilities of cellulase treatment group and enzyme combination treatment group showed high activity. The activity increased when enzyme treatment time increased. The combined enzyme treatment method for super sweet corn was suitable for food processing.
To produce an intestinal immunomodulatory beverage containing Centella asiatica extract (CAE), three types of CAE-added beverage prototypes were prepared, and their immunomodulatory activities and marker compounds were analyzed. As a result of the cytotoxicity assessment, all the beverages did not show significant toxicity compared to the control group. Next, the immunomodulatory activities of the beverage prototype were evaluated using the inflammatory model of IL-1β-induced intestinal epithelial cell line. All the samples significantly reduced the production of IL-6, IL-8, and MCP-1 in a CAE concentration-dependent manner. In addition, CAE-added beverages inhibited NO, IL-6, and IL-12 production in LPS-induced RAW 264.7 cells. When the major triterpenoids, as marker compounds for the production of CAE-added beverages, were analyzed by HPLC-DAD, only asiaticoside was detected beyond the limit of quantification, while madecassoside, madecassic acid, and asiatic acid were not detected. The amounts of asiaticoside in CAE-added beverage prototypes were confirmed in No. 1 (19.39 μg/mL), 2 (19.25 μg/mL), and 3 (19.98 μg/mL). In conclusion, the results of this study suggested that CAE-added beverage prototypes induced immunomodulatory effects in the intestinal inflammatory cell line models and asiaticoside could be used as a marker compound for CAE-added beverage production.
The fungus Alternaria alternata, responsible for causing brown to black spotting on numerous fruits and vegetables globally, was identified in 2022 as the causative pathogen of brown spot disease in potatoes in Korea. In pursuing potential inhibitors against A. alternata growth, we evaluated 15 mushroom culture filtrates: eight from Trametes spp. and seven from Polyporus spp., known for their antibacterial and anticancer properties. Antifungal activity was assessed by exposing each filtrate to A. alternata on a paper disc. Four filtrates displayed inhibitory action against the fungus, albeit with mild effects. Our findings highlight the potential of Trametes and Polyporus fungi as emerging antifungal candidates, offering promise in preventing potato brown spots.
This study investigated the surface tension and foaming properties of the hot-water extracts of pumpkin leaf and chickpea, as well as the effects of the plant hot-water extracts on white pan bread baking. Propylene glycol alginate (PGA), a synthetic emulsifier widely used in bakery, was used as a control. Pumpkin leaf water extract showed lower surface tension and comparable foaming capacity, compared with chickpea water extract and PGA solution when total solid 0.15% (w/w). Chickpea water extract showed the highest foam stability when total solid 0.15% (w/w). The dough was found to have a weak gel structure, and its viscoelastic properties were not significantly influenced by adding 0.05% or 0.15% (w/w) (based on total solid content) plant water extracts or PGA. The specific volume of the bread increased, and the baking loss was reduced by adding the two plant water extracts of total solid 0.15% (w/w). The hardness and chewiness of the bread crumb were reduced to a level comparable to the crumb containing 0.05% (w/w) PGA. The results showed that the pumpkin leaf water extract could be an effective natural emulsifier with a high phenolic content for bakery products.
This study aimed to examine the possibility of upcycling extracts of Angelica keiskei and Oenanthe javanica juice by-products through comparing enzyme extraction (EE) and complex extraction (CE) methods to increase the extraction yield and flavor of materials. A higher extraction yield was obtained for free amino acid content with EE and CE for A. keiskei and O. javanica juice by-products, respectively, and a higher extraction efficiency was achieved with juice by-products than with extracts prepared from raw materials before juice production. The content of major amino acids varied depending on the extraction method used. When used according to the characteristics of the extract, their use as a functional material was confirmed along with improvement in the flavor of the food. Consistently high extraction yields for organic acid and sugar levels were obtained with CE in A. keiskei and O. javanica juice by-products. The DPPH radical scavenging ability and TPC were consistently high with CE in A. keiskei and O. javanica juice by-products; the increase in extracted content was likely because of the reaction between the ethanol used for CE and the phenolic compounds. However, because the antioxidant capacity of the juice by-product extracts was somewhat lower than that of the extracts from raw materials before juice production, the amount used should be reviewed. The TFC was found to be higher in extracts obtained with EE than with CE for A. keiskei juice by-products; however, no significant difference was observed between EE and CE in the O. javanica juice by-products. Through this study, the taste compounds and antioxidant properties of extracts obtained from juice by-products produced after the production of A. keiskei and O. javanica green juice were analyzed, and the availability of high value-added materials was confirmed. Based on these research results, expanding specific R&D for practical use should be explored.
본 연구는 브로콜리 추출물의 DPPH radical 소거활성을 통한 항산화 활성과 저온(5℃이하), 고 온(50℃이상)의 보관 조건을 통해 화장품 제조 시 나타날 수 있는 온도 변화에 따른 항산화력 변화를 비교 해보고, 브로콜리 추출물 5%가 함유된 크림을 제조하여 화장품의 제형 안정성, 변색 및 변취, pH, 1차 첩 포 테스트를 통해 물리적, 화학적 안정성에 대해 확인하고 하였다. 실험 결과, 브로콜리 추출물 1%의 농도 에서 95.5%의 높은 DPPH radical 소거 활성이 확인되었으며, 온도 변화에 따른 보관 조건에도 항산화력 은 높에 유지되는 것을 확인되었다. 브로콜리 추출물 5% 함유된 크림에서도 제형의 안정성, 변색 및 변취, pH, 1차 첩포 테스트 모두 안전성과 안정성이 유지되는 것이 확인되었다. 본 연구들을 통해 브로콜리 추출 물의 높은 항산화 활성과 온도 변화에 따른 항산화력 유지력을 통해 피부에 안전하고 효과적으로 사용가능 한 화장품 소재로써의 가능성이 확인되었다.
Ipomoea aquatic is a leafy vegetable of the Convolvulaceae family, and is a tropical plant widely inhabiting southern China and Southeast Asia, and is widely known as Morning Glory in the West. In this study, the anti-inflammatory effects of ethyl acetate extract from Ipomoea aquatic extracts (IAE) were tested against lipopolysaccharide (LPS)-induced activation microglia BV2 cells. The production of nitric oxide (NO) and cell viability were measured using the Griess reagent and MTT assay, respectively. Inflammatory cytokine [interleukin (IL)-6, tumor necrosis factor (TNF)-, and interleukin-1 (IL-1)] were detected qPCR in LPS induced BV-2 cells. Subsequently, nuclear factor (NF)-B, mitogen-activated protein kinases (MAPKs), and nuclear factor erythroid-2-related factor 2 (Nrf2) were analyzed through western blot analyses and immunofluorescence. Ipomoea aquatic down-regulated of inflammatory markers and up-regulated anti-inflammatory and anti-oxidants in BV2 cells.
