        We investigated the anti-inflammatory effects of shiitake mushroom and kelp (SMK) mixture extracts in lipopolysaccharide (LPS)-stimulated murine RAW 264.7 cells. Treatment of RAW 264.7 cells with LPS significantly increased NO (nitric oxide) production, pro-inflammatory cytokines (tumor necrosis factor (TNF)-α, interferon (IFN)-γ, interleukin (IL)-6, and IL-1β), and inflammation-related genes (COX-2 and inducible nitric oxide synthase (iNOS)). In cytotoxicity testing using RAW 264.7 cells, SMK mixture extracts in the range of 1-16 μg/mL did not inhibit cell proliferation. However, SMK mixture extracts significantly inhibited NO production in a dose-dependent manner (p<0.05). SMK treatment significantly decreased TNF-α, IL-6, IFN-γ, and IL-1β levels compared to the LPS group, and similarly, pro-inflammatory cytokine mRNA levels also decreased. SMK mixture extracts reduced the mRNA expression of COX-2 and iNOS in RAW 264.7 cells compared to LPS (p<0.05). The above results show that SMK mixture extracts suppressed the inflammatory response induced by LPS. In particular, the extracts were shown to regulate the inflammatory response by suppressing the expression of inflammatory cytokines and inflammation-related enzymes.

        This study investigated the quality characteristics of the traditional rice cake of Gijeongtteok added with an extract from Cudrania tricuspidate fruit and different amounts of kimchi broth. The pH measurement showed a significant decrease with the increase of the kimchi broth from pH 5.48 (control) to 4.86 (KG180). Hunter color L-values were decreased while redness and yellowness were increased as the amount of kimchi broth increased. Texture profile analysis showed that hardness and adhesiveness significantly increased and decreased as the amount of kimchi broth increased. Gijeongtteok with kimchi broth (KG180) had the lowest specific volume with increased gummy layers in the crumb structure. Sensory evaluation of color, flavor, taste, appearance, and overall acceptance for various levels of kimchi broth showed that the KG140 sample had the highest acceptability. Therefore, this study suggests that Gijeogtteok made with the KG140 mixing ratio has excellent quality and sensory characteristics.

        The purpose of this study was to develop a functional beverage by using Caragana sinica extract and Tremella fuciformis Berk, which have high physiological effects such as anti-diabetic, antioxidant, anticancer, anti-lipidemic and anti-inflammatory. To this end, we used various mixing ratios of Caragana sinica extract, Tremella fuciformis Berk, and isomalto oligosaccharide, and measured the physicochemical properties of those beverages. The analyses showed the following: moisture content of those beverages using Caragana sinica extract and Tremella fuciformis Berk was 50.51~67.64%, pH was 5.19~5.38, and crude fat content was 0.28~0.52%, crude protein content was 1.26~1.80%, ash content was 0.47~0.80 and sugar content was 8.55~26.98°Brix. The lightness (L) of the beverages was 37.70~58.11, the redness (a) was 6.88~16.94, the yellowness (b) was 13.72~23.68, the total polyphenol content was 2.07~3.46 mg/g, and the total flavonoid content was 0.37~0.42 mg/g. Based on the results, it was confirmed that the nutritional components and total polyphenols of functional beverage using Caragana Sinica Extract, Tremella Fuciformis Berk and isomalto oligosaccharide were high at a mixing ratio of 1:1.3:25%. These results will increase the use of Caragana Sinica Extract and Tremella Fuciformis Berk, as functional materials in the future, and provide a framework for the manufacturing of diluted beverages comprising mineral water and carbonated water.

        본 연구의 목적은 효소 가수분해 및 유산균 혼합발효기술을 이용하여 GABA 함량이 강화된 발효 다시마 추출물을 생산함에 있다. TLC 분석방법을 통해 GABA 생성능력이 우수한 균주 L. plantarum KCTC 21004, L. acidophilus KCTC 3164, L. sakei subsp. sakei KCTC 3598를 선정하였 다. 선정된 균주를 이용한 유산균 발효의 특성은 배양 시간이 증가 할수록 적정 산도는 증가하였고 pH 는 감소하는 경향을 나타내었다. GABA 생성능력이 우수한 3종의 균주 중 L. plantarum KCTC 21004 이 GABA 함량이 136.4 mg/100g으로 우수하였다. 이러한 연구 결과는 유산균을 이용한 GABA 생산과 정에 있어서 기초적인 자료로 제공될 것으로 판단된다.

        Cassia tora seed was processed by germination and roasting to develop a germinated Cassia tora tea. The effects of extraction temperature on antioxidative compounds in tea extracts were examined and compared to four different commercial Cassia tora tea products. The contents of antioxidative compounds, such as phenolics, flavonoids, and β-carotene, increased as the extraction temperature increased (5 to 80oC), subsequently increasing in vitro antioxidant activities (DPPH and ABTS radical scavenging activities). Germinated Cassia tora tea had higher antioxidative activity than selected commercial Cassia tora teas, and the difference was significant at low extraction temperature. Therefore, the germinated Cassia tora tea could contribute to a new type of tea product with enhanced bioactive and antioxidative activities by adopting appropriate processing and extraction conditions.

        In this study, Korean Hallabong produced in Jeju Island and coffee were grafted to prepare coffee containing Hallabong extract and the nutritional components were analyzed. As the amount of Hallabong extract increased, the water content and total polyphenol content increased. However, the crude flour, crude protein, and total flavonoid content decreased significantly. The selenium content per 100 g was 91.28 mg in the 1% Hallabong group, and the iron content was 6.84 mg in the 3% Hallabong group. As the content of Hallabong extract in coffee increased, the L-value (brightness) and b-value (yellowness) increased, but the a-value (redness) showed a tendency to decrease. In the case of DPPH(2,2-Diphenyl-1-picrylhydrazyl) radical scavenging activity, the group containing 9% of Hallabong extract showed the highest value at 47.20 μmol/g of TEAC. In particular, the ABTS(2,2’-Azino-bis(3-ethylbenzothiazoline- 6-sulfonate)) and DPPH radical scavenging activity were significantly increased from coffee powder containing 6% or more of Hallabong extract(p<0.05). The caffeine content decreased as the amount of Hallabong extract added to coffee increased. Therefore, when making powder coffee with Hallabong extract added, it is recommended to set the content of Hallabong extract to 6%.

        흰점박이꽃무지 (Protaetia brevitarsis) 유충은 동아시아에서 식용 및 약용으로 널리 사용되고 있다. 흰점박이꽃무지 수요 증가로 인해 유충의 대량 사육 방법이 필요하지만 좁은 공간에서 대량으로 유충을 사육할 경우 곤충 병원성 질병 감염에 취약해진다. Metarhizium anisopliae는 흰점박이꽃무지 유충에 치명적인 곤충 병원성 곰팡이 중 하나이다. 이를 저해하기 위해 항진균 성분이 함유된 옻나무 (Rhus verniciflua Stokes) 추출액을 첨가하였고, 연구 결과 1% 옻나무 추출액을 첨가한 경우 녹강균에 의한 유충의 치사율이 유의미하게 낮았다. 또한 일반톱밥을 급여한 대조구에 비해 옻나무 추출액 처리구에서 유충의 무게가 더 무거웠고 유충 기간이 짧아 지는 것을 확인하였다. 따라서 이 연구를 통해 곤충 병원성 곰팡이에 대한 옻나무 추출액의 항진균 효과를 확인할 수 있었으며 흰점박이꽃무지 유충 사육 시 옻나무 추출액을 첨가 하여 급여한다면 병원성 곰팡이에 의한 유충의 치사율을 낮추고 증체량을 높이는 데 효과가 있을 것이다.

        CKD 추출액은 시멘트공정에서 발생한 폐기물인 CKD를 시멘트 원료로 재사용하기 위해 공정 방해물질로 작용하는 KCl을 추출한 폐수이며, 폐수처리시설 증설 등의 문제로 추출액 무방류 및 이를 재이용하고자 하였다. 이온교환법을 적용하여 KCl을 제거한 결과, 이온교환 후 추출액의 pH는 12.7 에서 pH 2 미만으로 감소하였으며 양이온교환수지의 H+가 이온교환을 거쳐 추출액에 용해되었음을 확인하였다. 이온교환의 선택성에 의해 Ca2+, K+ 순서로 제거되었으며, K+ 이온을 제거하기 위해 접촉시 간의 증가가 필요함을 판단하였다. 이온교환수지와 직접접촉시간이 약 6배 높은 접촉시간을 갖는 회분 식장치에서 연속흐름식장치 대비 4배 높은 K+ 제거 효율을, 7배 높은 Cl- 제거 효율을 확인하였다. 양이온교환수지의 H+가 음이온교환수지의 OH- 대비 1.2배 빠른 교환속도를 가짐을 추출액 pH 변화를 통해 확인하였다.

        This study examined the quality characteristics and anti-oral microbial activity of bamboo leaf jelly prepared with different 5 levels (0, 10, 20, 30, and 40%) of bamboo (Phyllostachys nigra var. henonis) leaf extract. The sugar contents of bamboo leaf jelly were increased significantly by increasing the level of bamboo leaf extract. The luminance and Hunter’s a values of the jelly samples increased with increasing bamboo leaf extract, but the 40% bamboo leaf jelly had the lowest Hunter’s b values. The hardness, adhesiveness, gumminess, and chewiness increased significantly with increasing bamboo leaf extract. Among the mechanical properties, only the flavor of the jelly with 30 and 40% bamboo leaf extract were increased significantly. The extract of bamboo leaves had strong antimicrobial activity against S. mutans, S. sobriuns, P. gingivalis, and P. intermedia at a concentration of 40%. These results suggest that bamboo leaf extract can be useful in the production of high quality jelly.

        모란은 항염증작용, 항알레르기작용, 항균작용, 중추억제, 위액분비억제, 진경작용 등의 약리활성이 있을 뿐 아니라 항산화작용, 미백작용 등이 보고되면서 화장품 원료, 미용식품으로 다양하게 활용 가능성이 탐색되고 있다. 모란의 뿌리, 잎 및 꽃에서 추출용매와 온도, 시간에 따른 추출액 중 성분의 함량 변화를 비교하였다. Paeoniflorin과 paeonol을 지표성분으로 하여 두 성분의 함량을 동시에 정량분석하기 위한 HPLC 분석법을 설정하고 분석 배치마다 검량선과 QC용 검체를 사용하여 시스템적합성을 확인하면서 시료 분석을 실시하였다. Paeonol은 뿌리에서만 검출되었고 잎과 꽃에서는 검출되지 않았으며, paeoniflorin은 잎, 꽃에서의 농도가 뿌리에서보다 높았다. 추출용매와 섞을 때 뿌리의 절도는 구입 당시의 조각을 그대로 사용할 때보다 적당한 크기로 분쇄한 후 사용할 때 두 성분 모두 높은 농도로 추출되었다. 추출용매로는 30% 1,3-butylene glycol을 사용할 때 농도가 가장 높았다. 추출온도와 시간은 고온, 장시간 일수록 농도가 높아지는 경향을 보였으나 75℃, 4 h 이상에서는 농도 상승이 완만했으며, paeonol은 8 h까 지 지속적으로 농도가 상승하는 경향을 보였다. 뿌리, 잎, 꽃의 배합 비율은 2+2+1g/0.5kg로 할 때 두 지표성분의 함량 기준에 도달 가능하다고 판단하였다. 최종적으로 원재료를 2+2+1g/0.5kg로 배합하고 30% 1,3-butylene glycol을 추출용매로 사용하여 75℃에서 4 h 동안 추출하는 방법이 두 지표성분의 활성 농도 기준과 원재료 및 제조공정상의 비용을 함께 고려할 때 적절한 조건이라고 판단하였고, 이 조건에서 10 kg 규모까지 추출액 제조 단위를 스케일업 하였다.

        Coffee (Coffea spp.) is one of the most important agricultural commodities, being widely consumed in the world. Various beneficial health effects of coffee have been extensively investigated, but data on habitual coffee consumption and its bio-physiological effect have not been clearly explained as well as it is not proved the cause and effect between drinking coffee and its bio-physiological reactions. We made the dialyzed coffee extract (DCE), which is absorbable through gastrointestinal tract, in order to elucidate the cellular effect of whole small coffee molecules. RAW 264.7 cells, a murine macrophage lineage, were directly treated with DCE, i.e., DCE-2.5 (equivalent to 2.5 cups of coffee a day), DCE-5, and DCE-10, for 12 hours, and their protein extracts were examined by immunoprecipitation high performance liquid chromatography (IP-HPLC). RAW 264.7 cells differently expressed the inflammation-related proteins depending on the doses of DCE. RAW 264.7 cells treated with DCE showed marked increase of cathepsin C, cathepsin G, CD20, CD28, CD31, CD68, indicating the activation of innate immunity. Particularly, the macrophage biomarkers, cathepsin G, cathepsin C, CD31, and CD68 were markedly increased after DCE-5 and DCE-10 treatments, and the lymphocyte biomarkers, CD20 and CD28 were consistently increased and became marked after DCE-10 treatment. On the other hand, RAW 264.7 cells treated with DCE showed consistent increase of IL-10, an anti-inflammatory factor, but gradual decreases of different pro-inflammatory proteins including TNFα, COX-2, lysozyme, MMP-2, and MMP-3. In particular, the cellular signaling of inflammation was gradually mitigated by the reduction of TNFα, COX-2, IL-12, and M-CSF, and also the matrix inflammatory reaction was reduced by marked deceases of MMP-2, MMP-3, and lysozyme. These anti-inflammatory expressions were consistently found until DCE-10 treatment. Therefore, it is presumed that DCE may have dynamic effects of innate immunity activation and pro-inflammation suppression on RAW264.7 cells simultaneously. These effects were consistently found in the highest dose of coffee, DCE-10 (equivalent to 10 cups of coffee a day in man), that might imply the small coffee molecules were accumulated in RAW 264.7 cells after DCE-10 treatment and produce synergistic cytokine effects for innate immunity activation and anti-inflammatory reaction concurrently.

        Coffee is one of the most familiar beverages to modern human adults, but its bio-physiological effect has not been clearly elucidated. It was known that more than one thousand chemicals were included in the ordinary coffee extract. Among them, the caffein and chlorogenic acid (caffeoylquinic acids) are most abundant and have been investigated by many authors so far. In order to know the real cellular effect of whole coffee extract elements, the dialyzed coffee extract (DCE)1) was made to get coffee elements less than 1000 Da molecular weight, which are freely absorable through gastrointestinal tract. It was directly treated in the culture of RAW 264.7 cells, a murine macrophage lineage. RAW 264.7 cells were treated with DCE equivalent to 2.5 cups of coffee (DCE-2.5), DCE-5, and DCE-10 for 12 hours, and their protein extracts were examined by histological observation and immunoprecipitation high performance liquid chromatography (IP-HPLC). RAW 264.7 cells differently expressed the proliferation-related proteins depending on the dose of DCE. DCE-2.5 and DCE-5 enhanced the cellular growth of RAW 264.7 cells by increasing the expression of β-actin, PCNA, Ki-67, MPM2, MAX, cMyc, E2F-1, and Rb-1, and by decreasing the expression of MAD and p21. These proliferation-related proteins were rarely affected by DCE-10. DCE-2.5 and DCE-5 induced the cellular proliferation of RAW 264.7 cells by the signaling of E2F-1 and cMyc, respectively, but these cellular effects almost disappeared in DCE-10. Therefore, it was presumed that the low dose of coffee, DCE-2.5 and DCE-5 might be effective for the proliferation of murine macrophages, RAW264.7 cells, contrast to the high dose of coffee, DCE-10. It was also suggested that the low dose of DCE-2.5 and DCE-5 be helpful to increase the innate immunity in vivo by increasing the cell number of macrophages in contrast to the high dose of DCE-10.

        홍삼추출물(RGE)의 쓴맛을 개선하기 위해서 증숙시 초산 처리 후 α-, β-, γ-CD을 이용 해서 RGE의 포접화합물을 제조하여, 전자혀 분석을 통해서 RGE-γ-CD에 의한 쓴맛 개선효과가 가장 큰 것으로 확인하였다. 인삼의 열처리 공정에 있어 초산 처리는 Rg3 및 비극성 진세노사이드 성분 함량 을 증가시킴을 알 수 있었다. 전자혀를 이용하여 α, β, γ- CD 첨가량에 따른 쓴맛, 신맛, 짠맛, 우아 미맛 및 단맛을 분석하였다. 그 결과 RGE 대비 10%의 γ-CD를 첨가하여 포접한 REG가 다른 처리구 에 비해 쓴맛이 월등히 낮은 감응도를 나타내었다.

        The objective of this experiment was to evaluate the effect of Chamaecyparis obtus extract to reduce odor emissions released from the swine feeding operations. Finisher pigs [Landrace × (Yorkshire × Duroc)] with an initial body weight averaging 50 kg were housed separately in two rooms with eighty eight pigs in each room at a swine feeding operation site. C. obtus extract was sprayed in the room by ceiling sprayer for one minute at twice per day during two months. Concentrations of odorous compounds from air in the room of the swine feeding operation were analyzed at four times during two months. Levels of butyric acid, valeric acid, i-butyric acid, ivaleric acid, skatole, methylmercaptan, and trimethylamine tended to decrease in C. obtus extract spray treatment group compared to the non-spray treatment group (P > 0.05). Odor activity values of butyric acid, valeric acid, skatole and trimethylamine were higher than other odorous compounds and decreased by 72%, 76%, 54% and 20%, respectively, in C. obtus extract spray treatment group compared to the non-spray group. Taken together, C. obtus extract showed an odor reducing capability in the air of a swine feeding operation suggesting that it possesses anti-bacterial properties as well as having a dust removal and masking effect.
