We performed a study to examine the association between diet quality and nonalcoholic fatty liver disease (NAFLD). Our study included 3,586 women aged 40-64 years who participated in the sixth Korea National Health and Nutrition Examination Survey. The study subjects were classified into the NAFLD group (n=816) and the normal group (n=2,770) using the hepatic steatosis index. The anthropometric indices, blood profiles, and dietary intake data of the subjects were obtained. The waist circumference, body mass index, and the serum levels of triglycerides, fasting blood sugar, HbA1c, and systolic and diastolic blood pressures were higher in the NAFLD compared to the normal groups (p<0.001, respectively). The intakes of protein (g/kg body weight, p<0.001), potassium (p<0.001), and vitamin A (p=0.006) were significantly lower in the NAFLD group. It was observed that the higher the total Korean Healthy Eating Index score, the lower the risk of NAFLD. A reverse relationship was shown between the NAFLD risk and the intakes of total fruits, total vegetables, vegetables excluding Kimchi and pickled vegetables, meat, fish, eggs and beans. Therefore, it is recommended that middleaged women in Korea increase their intakes of fruits, vegetables, and foods high in protein for the proper management of NAFLD.
본 연구는 고온 환경에서 사료내 트립토판과 나이아신의 첨가가 산란계의 생산성, 난품질, 지방간 지표 및 혈액성상에 미치는 상호작용 효과를 조사하고자 수행되었다. 총 384수의 25주령 로만 갈색종 산란계를 4처리 8반복으로 반복당 12수씩 무작위 임의 배치하였다. 기본 사료는 트립토판과 나이아신의 추가적인 첨가는 없으며 모든 영양소 및 에너지는 로만갈색종 산란계의 요구량에 충족하거나 초과하도록 배합되었다. 사료 처리구는 2 × 2 요인 실험 설계법으로 두 가지 수준의 트립토판(0 및 0.16%)과 두 가지 수준의 나이아신(0 및 0.03%)이 포함되었다. 모든 산란계의 사양환경은 일반 농가에서 낮시간에 온도가 올라가는 점을 고려하여 일일 중 8시간은 온도 31.4 ± 1.17℃, 습도 86.0 ± 4.28%으로 설정하였으며, 나머지 16시간은 온도 26.7 ± 1.10℃, 습도 61.7 ± 6.34%로 유지하여 주기적인 고온 환경을 조성하였다. 실험은 10주간 진행되었다. 실험 결과 사료내 트립토판과 나이아신의 주요 효과와 상호작용은 나타나지 않았다. 이와 유사하게, 난각 강도, 난각 두께, 난각색, 난황색 및 호우유닛에 대한 상호작용 도 나타나지 않았다. 주요 효과로 0.03%의 나이아신 첨가는 난황색을 유의적으로 감소시켰으나 호우유닛은 유의적으로 증가시켰다. 하지만, 0.16%의 트립토판 첨가는 난품질에 영향을 끼치지 않았다. 지방간 지표와 혈액성상에서 사료내 트립토판과 나이아신의 주요 효과 및 상호작용은 나타나지 않았다. 결론적으로, 본 연구에서 설정한 농도의 사료내 트립토판과 나이아신의 첨가는 고온 환경에서 사양되는 산란계의 생산성, 난품질, 지방간 지표 및 혈액성상에 긍정적인 영향을 미치지 않았다.
본 연구는 폐경 후 비만 여성의 동물모델을 이용하여 수영운동이 비알코올성 지방간에서 염증에 미치는 영향을 조사하였다. 실험동물은 수영운동을 하지 않은 모의수술군(S/N), 수영운동을 하지 않은 난소절제 수술군(O/N) 및 수영운동을 실시한 난소절제 수술군(O/S)으로 구분되어 8주 동안 고지방식이 사료를 섭취하면서 사육되었다. 간 조직의 지방축적, 간 무게 및 혈청 속 AST와 ALT는 S/N에 비해 O/N 에서 증가하였으나, O/N에 비해 O/S에서는 감소하였다. S/N에 비해 O/N는 간 조직에서의 IκBα의 유전자 발현이 감소하였고 MCP-1, IL-6 및 TNF-α의 유전자 발현은 증가하였다. 그러나 O/N에 비해 O/S는 간 조직에서의 IκBα이 증가하였고 MCP-1, IL-6 및 TNF-α의 유전자 발현은 감소하였다. 결론적으로 본 연구는 난소절제 후 고지방식이의 섭취로 비만이 유도된 비만 쥐에서 수영운동이 비만으로 유도 된 비알코올성 지방간에서 염증을 개선함으로써 건강증진에 효과적임을 제시하였다.
Non-alcoholic fatty liver disease (NAFLD) is emerging as the most common liver disease in industrialized countries. Recently, natural compounds that may be beneficial for improving NAFLD have received increasing attention. Artemisia annua L. is the source of antimalarial phytomolecule, artemisinin, which has been reported to prevent obesity. However, the effect of A. annua extract on hepatic lipid metabolism remains unclear. This study was performed to determine the protective effect of Artemisia annua extract (AAE) on high-fat diet (HFD)-induced hepatic lipid accumulation, and elucidate the molecular mechanisms behind its effects in vivo and in vitro. We found that HFD-fed mice with AAE administration (50 mg/kg/day) for 8 weeks dramatically reduced hepatic lipid accumulation compared to the control mice taken with HFD alone. The body and liver weights of AAE group were significantly lower than those of HFD group, and oral administration of AAE remarkably suppressed the serum levels of triglyceride (TG), total cholesterol (TC), fasting glucose, alanine transaminase (ALT) and aspartate transaminase (AST) in HFD-fed mice. AAE significantly increased the phosphorylation of AMP-activated protein kinase (AMPK) and acetyl-CoA carboxylase (ACC) in the liver of HFD-fed mice and HepG2 hepatocytes. Moreover, AAE downregulated the hepatic expression of sterol regulatory element-binding protein-1c (SREBP-1c) and fatty acid synthase (FAS) in HFD-fed mice and high glucose-treated HepG2 cells. In addition, the inhibitory effects of AAE on the overexpression of SREBP-1c and FAS were attenuated by compound C, which is the specific AMPK inhibitor, in high glucose-treated HepG2 cells. These results indicated that AAE may represent a promising approach for the prevention and treatment of obesity-related NAFLD via the activation of AMPK and the regulation of AMPK-dependent lipogenic genes.
본 연구의 목적은 oleic acid로 지방생성이 유도된 HepG2 세포에서 자색옥수수 색소 1호 포엽 및 속대 추출물이 간 세포 내 지방생성에 미치는 영향을 구명하는 것이다. 자색옥수수 색소 1호 포엽 및 속대 추출물에 의한 HepG2 세포 내 지방 축적의 변화를 확인하기 위하여 배양된 세포에 oleic acid로 지방 축적을 유도하고 추출물에 의한 중성지방생성 억제 효과를 측정하였으며 추출물을 처리하지 않은 대조군과 추출물을 처리한 실험군의 지방합성 및 축적에 관련된 유전자와 단백질 발현량을 RT-PCR과 Western blot을 통하여 측정하였다. Oil Red O와 Nile Red 염색을 통하여 추출물의 처리로 HepG2 세포 내 중성지방 축적이 억제된 것을 확인하였다. RT-PCR에 의하여 mRNA 발현량을 측정한 결과, oleic acid에 의하여 지방 생성이 유도된 대조군에 비하여 모든 추출물 처리군의 SREBP-1c와 SREBP-1a 유전자 발현량이 유의적으로 감소되었다. Western blot을 실시하여 p-AMPK, p-SREBP1, PPARα, FAS 단백질의 발현량을 측정한 결과, 간에서 지질대사에 관여하는 주요 인자인 SREBP1 단백질의 발현은 추출물의 처리 농도에 따라 유의하게 감소하였으며 지방산의 생합성 경로에 관여하는 주요 효소인 FAS의 단백질 발현향은 모든 처리 농도에서 현저하게 감소된 것이 확인되었다. 본 연구 결과는 자색옥수수 색소 1호 포엽 및 속대 추출물이 간세포 내에서 중성지방의 축적을 억제시키고 지질 합성에 관련된 유전자 및 단백질의 발현을 억제시킴으로써 간 세포 내 지질 축적을 완화할 수 있는 기능성 소재로의 활용가치가 높다고 판단된다.
Alcoholic steatosis is a fundamental metabolic disorder and may precede the onset of more severe forms of alcoholic liver disease. In this study, we isolated enzymatichydrolysate from Semisulcospira libertine by alcalase hydrolysis and investigated the protective effect of Semisulcospira libertine hydrolysate on liver injury induced by alcohol in the mouse model of chronic and binge ethanol feeding (NIAAA). In an in vitro study, the hydrolysate protects HepG2 cells from ethanol toxicity. Liver damage was assessed by histopathological examination, as well as by quantitating activities of serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), and alkaline phosphatase (ALP). After the administration of S. libertina hydrolysate, fat accumulation and infiltration of inflammatory cells in liver tissues were significantly decreased in the NIAAA mouse model. The elevated levels of serum AST, ALT, and ALP activities, along with the lipid contents of a damaged liver, were recovered in experimental mice administrated with S. libertina hydrolysate, suggesting its role in blood enzyme activation and lipid content restoration within damaged liver tissues. Moreover, treatment with S. libertine hydrolysate reduced the expression rate of cyclooxygenase (COX-2), interleukin (IL)-1β, and IL-6, which accelerate inflammation and induces tissue damage. All data showed that S. libertine hydrolysate has a preventive role against alcohol-induced liver damages by improving the activities of blood enzymes and modulating the expression of inflammation factor, suggesting S. libertine hydrolysate could be a commercially potential material for the restoration of hepatotoxicity.
목 적 : 간 자기공명영상에서 Screening Dixon, Multi Echo Dixon, High-speed T2-corrected Multi Echo acquisition at 1H MR spectroscopy의 지방간 평가를 초음파 검사와 비교하여 간 자기공명상에서 지방간 평가의 유용성을 알아보고자 하였다.
대상 및 방법 : 2014. 8월~2015. 8월까지 본원을 내원하여 간 자기공명영상 검사를 시행한 환자들 중 검사시행 3개월 전· 후에 초음파 검사를 동시에 시행한 138명(남: 101명, 여: 37명, 평균연령: 60.2세)의 환자를 대상으로 하였다. 연구에 사용 된 MRI 장비는 Magnetom Skyra 3.0T(Siemens Healthcare, Germany)였으며 18ch Body array coil을 이용하여 Screening Dixon, Multi Echo Dixon, High-speed T2-corrected Multi Echo acquisition at 1H MR spectroscopy(이하 HISTO) 기법의 검사를 시행하였다. 초음파 장비는 iu22(Philips healthcare, Netherlands), E9(GE healthcare, America)였으며 C3-5 convex probe를 사용 하였다. 지방간의 분석은 Screening Dixon에서 지방간의 유·무와 Multi Echo Dixon, HISTO 기 법간의 지방간 유무를 각각 비교하였으며 Multi Echo Dixon과 HISTO 기법에서는 지방 함유량에 따라 normal(5% 이하), mild(5~33%), moderate(33~66%), severe(66% 이상)로 나누어 두 기법 간 비교 및 초음파 검사와 비교하여 그 유의성을 SPSS21 통계프로그램을 이용하여 person χ2(카이제곱) 검증하였다.
결 과 : Screening Dixon과 Multi Echo Dixon, Screening Dixon과 HISTO 기법을 비교한 결과, Screening Dixon에서 지 방간을 가진 환자가 가장 많은 것으로 나타났지만 통계적인 차이는 없었으며(p>0.05), Multi Echo Dixon과 HISTO 기법을 normal, mild, moderate, severe 군으로 나누어 비교한 결과 두 기법에서의 유의한 차이는 없는 것으로 나타났다(p>0.05). 반면 Multi Echo Dixon과 초음파, HISTO와 초음파의 각각의 비교에서는 초음파 검사에서 지방간으로 평가되는 환자가 Multi Echo Dixon, HISTO에 비해 많은 것으로 나타났으며(p<0.05), normal 군의 환자가 많이 감소한 반면 moderate, severe군으로 평가되는 환자가 많이 증가하였다.
결 론 : Screening Dixon, Multi Echo Dixon, High-speed T2-corrected Multi Echo acquisition at 1H MR spectroscopy(HISTO)의 지방간 평가를 초음파 검사와 비교한 결과 자기공명영상을 이용한 평가에서는 기법 간에 유의한 차이가 없는 것으로 나타났으나 초음파와 비교에서는 유의한 차이가 있는 것으로 나타났다. 그러나 자기공명영상을 이용한 검사는 간 자기공명영상 검사에서 부가적인 검사로 짧은 시간에 지방간을 정량화 할 수 있다는 장점이 있었다.
This study was performed to investigate the effects of Acer tegmentosum Maxim. extract (ATE) on non-alcoholic fatty liver in Sprague Dawley (SD) rats. During oral administration of ATE, non-alcoholic fatty liver was induced by treatment with DL-ethionine. The lipid, total cholesterol (T-CHO) and malondialdehyde (MDA) in the liver tissue of ATE-fed rats showed lower levels, as compared to ATE-unfed rats. In ATE-fed rats, the serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and γ-glutamyl transferase (GGT) were lower than the case of ATE-unfed rats. Oil red staining of the liver showed that the lipid deposits were decreased by feeding ATE. These results strongly indicated that ATE has positive effects of protection against non-alcoholic fatty liver formation.
The purpose of this report is to discuss the obtained findings gathered from ultrasound screenings of the liver. After running diagnostic tests health, screens were then conducted to analyze and compare the gained results. This data was then charted and used to strengthen our theorized hypothesis. From January 2013 to June 2013 a recorded 2906 people over the age of 20 visited Health Promotion Centers in various areas throughout Daejeon. Of those 2906 participants 1789 underwent screenings for abdominal ultrasonography; these participants as well as their ultrasound results were used as the bases of our study. For the establishment of our baseline and the comparison of our results, we gathered case-controlled studies from various reputable sources. Both the control and the experimental groups were tested to measure the following liver enzymes (AST, ALT, r-GTP, ALP, and etc.). Kidney functions were measured as well enzymes such as BUN, Creatinine, and Uric Acids levels were analyzed and recorded to see if any relationships existed between the levels documented in the liver and those in the kidneys. It was shown that the two primary causes of fat build up within the liver were significantly connected to obesity BMI(OR=4.14) and waist circumference(OR=3.88).
This study was conducted in order to investigate the effects of Acer tegmentosum Maxim. extract (ATE) in prevention of liver injury. After oral administration of ATE to SD rats, liver injuries were induced by treatment with CCl4, galactosamine, or ethionine. Aspartate aminotransferase (AST), alanine aminotransferase (ALT), triglyceride, and cholesterols in blood were used as indicators of liver damage. When acute hepatitis was induced by CCl4 or galactosamine, ATE-fed rats showed a lower level of AST and ALT in plasma than ATE-unfed rats. In the case of ethionine-induced fatty liver, triglyceride levels in plasma were reduced in ATE-fed rats, compared with ATE-unfed rats. These results indicate that Acer tegmentosum Maxim. extract protected against hepatic failure.
This study was carried out to investigate the effect of wormwood meal addition on constituent of blood, and liver fatty acid in broiler chickens. Two-day old 25 male commercial broiler chicks were distributed to 5 treatments (wormwood meal levels ; T1 (1%), T2 (3%), T3 (5%), T4 (10%), and control (C)) for 5 weeks. Free fatty acids of liver were a little different among the treatments and the palmitic, stearic, oleic, and linoleic acids were occupied 92.5% of total free fatty acids. Hemoglobin and hematocrite concentration of blood were significantly decreased (p<0.05) as the levels of wormwood meal increased, and serum albumin concentration of broilers was little different between 0 week and 5 week experiments. Also serum cholesterol, phospholipid, total cholesterol, and triglyceride concentration of liver in broilers were little different during the 5 weeks experiment. Serum triglyceride concentration was significantly increased (p<0.05) as levels of wormwood meal increased. These results indicate that dietary wormwood meal addition has a little positive effect on increasing unsaturated fatty acid and decreasing saturated fatty acid.
Lipid peroxidation is one of the main manifestations of oxidative damage and has been found play an important role in the toxicity and carcinogenesis of many carcinogens. This study was carried out to investigate the effects of aflatoxin B₁ co-administrated with antioxidant vitamins on lipid contents and fatty acids components of liver in mice. For this work, vitamin C and vitamin E, the major antioxidants, were administrated with 10 ㎎/㎏ and 63.8 ㎎/㎏ respectively, through intraperitoneal(i.p) injection to male ICR mice, and 0.4 ㎎/㎏ of the AFB₁ injected by i.p. lhr later. The results were as follows: two fold amounts of free cholesterol, triglyceride, and total cholesterol in serum and liver of mice treated with only AFB₁ were observed, when compared to those of mice co-administrated with antioxidant vitamins. However, the levels of phospholipids in serum and liver of mice treated with only AFB₁ were decreased. Concerning to fatty acids composition of liver from AFB₁-treated mice, P/S ratio was shown more low level in cholesteryl ester, triglyceride, total cholesterol and phospholipid than those of mice co-administrated with antioxidant vitamins. In these data which provide with a reliable evidence on their antioxidantal effects to aflatoxicosis.
The role of cytochrome P-450 2E1 (P450 2E1) in the early phase of alcoholic fatty liver was examined. Female rats were pretreated with either allyl sulfide (200 mg/kg, po), disulfiram (500 mg/kg, po), YH 439 (250 mg/kg, po) or pyrazine (200 mg/kg/day × 2 days, ip). Marked changes in carbon tetrachloride-induced hepatotoxicity and carboxyhemoglobin (COHb) elevation due to dichloromethane administration were observed in rats treated with one of the P450 2E1 modulators. A single dose of ethanol (6 g/kg, po) increased the hepatic triglyceride contents approximately 2 fold, which was inhibited completely by YH 439 pretreatment. However, the other P450 2E1 modulators failed to alter the ethanol-induced hepatic triglyceride accumulation. In vitro hepatic microsomal enzyme activity was determined in 4 week old premature and 12 week old adult rats. Aminopyrine-N demethylation was not different, but p-nitrophenol hydroxylation and p-nitroanisole O-demethylation were significantly higher in premature rats compared to adult indicating that the P450 2E 1 activity decreases with the maturity of rats. However, no difference in the triglyceride accumulation induced by an intraperitoneal dose of ethanol (3 g/kg) was noted between premature and adult rats. The results suggest that the P450 2E1 activity dose not play an important role in the induction of acute alcoholic fatty liver.
To investigate the influence of saturated fats, α-linolenic acid, EPA and DHA on the lipid hydroperoxide concentration and fatty acid composition in liver microsomes and in plasma lipid of rabbits, the animals were fed on the perilla oil rich α-linolenic acid or sardine oil rich EPA and DHA diet for four weeks Were examined. The fatty acid composition of plasma lipid and liver microsomes of rabbits fed on the perilla oil diet was an accumulation of arachidonic acid(AA) 20:4 n-6, eicosapentaenoic acid(EPA) 20:5 n-3, and docosahexaenoic acid(DHA) 22:6 n-3, The fatty acid composition of plasma lipid and liver microsomes of rabbits fed on the sardine oil was an accumulation of α-linolenic acid(LNA) 18:3 n-3, and arachidonic acid(AA) 20:4. The p/s ratio of rabbits fed on the perilla oil diet changed from 7.4 to 2.27 for plasma lipid and 2.47 for liver microsomes. The concentration of lipid hydroperoxide was 3.48 nmol MDA/ml and 4.35 nmol MDA/ml for plasma lipid and liver microsomes, respectively, in perilla oil diet. The lipid hydroperoxide liver was 4.22 nmol MDA/ml and 67 nmol MDA/ml for plasma lipid and liver microsornes in sardine oil diet.
Lipid levels in the tissues of liver and intestines of O.bartrami amounted to 40.0% and 1.5%. The new compounds was found to be ethyl acylates, from a deduction of their detailed 1H-nuclear magnetic resonance(NMR) and 13C-NMR as well as infra red spectra (IR). The fatty acid composition of total lipids were mainly composed of C16:0(19.0%), C18:1(16.2%) and C22:6Ω3(15.7%), followed by C20:1(9.4%), C22:1(6.4%) and C18:0(5.4%). New compound A and B were seemed to derived from the cleavage of glycerol moieties of triglycerides by microbial activities during storage in a frozen state. Compound A contained C16:0(38.2%), C18:1(13:4%), C20:1(13.3%) and C22:1(11.7%) as major components, while compound B predominantly comprised polyunsaturated fatty acid such as C20:5Ω3 (41.2%) and C22:6Ω3(36.1%). In both compounds small amounts of odd numbered fatty acids were also detected (3.8~2.2%).
본 연구는 복부 초음파 검사를 통해 확인된 지방간 환자의 지방간 정도에 따른 혈액학적 고지혈증 지표와의 관련성을 분석하여 관련 연구에 기초자료를 제공하고자 시행되었다. 2017년 1월 1일부터 2017년 12월 31일까지 총 412명의 성인을 대상으로 하였으며, 복부초음파 검사를 진행하여 지방간이 확인된 환자 중 초음파 검사 시행일 기준 3개월 이내의 혈액학적 데이터가 있는 환자를 대상으로 후향적 분석을 실시하였다. 연구결과, 지방간 정도에 따른 혈액학적 수치는 중성지방, AST, ALT에서 통계적으로 유의한 차이가 있었고(p<.05), 지방간은 전체 변수 중 성별, 중성지방, AST, ALT와 관련성이 있었다(p<.05). 결론적으로 지방간 정도는 고지혈증의 혈액학적 수치와 직접적인 영향인자는 아니었지만 그 관련성 확인을 통해 지방간과 고 지혈증 동반 관리의 필요성을 확인했다.
본 연구는 초음파상 지방간을 세 등급으로 구분하여 적용하는 것이 어떠한 임상적 의의가 있는지 알아 보고자 하였다. 2018년 6월부터 2019년 4월까지 부산 P 검진센터에서 복부초음파 검사를 받은 3607명 중 지방간으로 진단된 1047명(남 818명, 여 229명)을 대상으로 하였다. 초음파상 간 실질의 에코정도, 음향감 쇄정도, 간 내 혈관 및 횡격막이 보이는 정도에 따라 GradeⅠ군(경증 지방간), GradeⅡ군(중등도 지방간), GradeⅢ군(중증 지방간)의 세 등급으로 분류하고, 각 군별 비만지표(체질량지수, 허리둘레), 혈액학적 분석 및 대사증후군과의 연관성을 남녀로 구분하여 분석하였다. 결과로 지방간 등급이 올라갈수록 남자는 여자에 비해 중증 지방간의 비률이 높게 나타났다. 성별에서는 남자가 지방간 등급이 올라 갈수록 각 군별 평균연령이 높게 나타났다. 체질량지수, 허리둘레는 남녀 모두에서 유의하게 지방간 등급이 올라갈수록 증가 하는 차이를 나타내었다(p=.000). 혈액학적 분석에서는 남자는 AST, ALT, γ-GTP, TG, 공복혈당, 당화혈색소에서 유의한 차이를 나타내었다(p<.05). 여자는 ALT, γ-GTP, TG에서 지방간의 등급이 올라갈수록 유의한 차이가 나타났다(p<.05). 대사증후군의 유병률은 남녀 모두 지방간의 등급이 올라갈수록 유의하게 증가 하였다(p=.000). 연구결과를 기초로 초음파상 지방간을 등급에 따라 세분하여 적용함에 있어 혈액학적 변수, 대사증후군 등을 고려하여 지방간 등급을 나눈다면 지방간의 치료 및 경과관찰에 도움이 될 것으로 사료된다.
