        This study was conducted to investigate the health effects of the general components of acorns in animals. In Korea, acorns have been used as food material in the form of acorn starch. Acorns contain tannin, gallic acid, digallic acid, and gallotannin. Therefore, the health effects of acorns as functional food are actively being studied. The chemical composition of acorns may vary according to the harvest time, region, and breed. Acorns have excellent antioxidant properties and might be beneficial in controlling hyperlipidemia, obesity, cardiovascular disease, and dementia, as well as strengthening immune competence. Further study is necessary to better understand the benefits of acorns, as it is expected to represent a large part of the food industry.

        Red raspberry (Rubus crataegifolius) is a well-known fruit with excellent antioxidant, anti-inflammatory, anti-bacterial and anti-cancer resources containing bioactive phenolic acids, terpenoids, glucosinolates, and glycosides. The aim of our study is to investigate the effect of red raspberry cookies on quality characteristics and antioxidant activities. Cookies were prepared adding different concentrations of red raspberry powder (0, 5, 10, 20, 30, and 50% to the flour weight). The bulk density, pH, spread ratio, loss rate, leavening rate, color, hardness of dough, and cookies were investigated. As a result, the pH of the dough, leavening and loss rate, and L- and b-value of the cookies are significantly decreased with increasing levels of red raspberry powder (p < 0.05). On the other hand, spread ratio, a-value, and brix of the cookies are significantly increased with increasing levels of red raspberry powder (p < 0.05). Total phenol compounds and FRAP assay are significantly increased with increasing levels of red raspberry powder (p < 0.05). Based on the overall observation, red raspberry cookies with 10-20% Rubus crataegifolius powder is recommended to take advantage of its functional properties with consumer acceptability.

        본 연구는 인체 유방암세포 SK-BR-3, MDA-MB-231과 위암세포 AGS를 대상으로 국화과 식물 중 흰민들레(Taraxacum coreanum, TC), 고들빼기(Youngia sonchifolia, YS), 씀바귀(Ixeris dentate, ID) 추출물에 의한 증식 억제효과를 비교 한 후, 가장 억제효율이 높은 추출물을 선택하여 apoptosis 유도 효과를 조사하였다. 암세포의 증식 억제효과는 TC 추출물에 의한 영향은 약한 반면, YS와 ID 추출물에 의한 영향은 농도 의존적으로 증가하는 것을 확인하였다. 세 가지 추출물 중 가장 효과가 뛰어난 ID 추출물을 이용하여 추가적으로 농도 설정을 진행한 다음 이후 실험을 진행하였다. ID 추출물에 의한 apoptosis 양성세포를 확인하기 위해 DAPI assay를 진행한 결과, ID 추출물을 처치한 군에서 apoptotic body와 세포질 응축을 확인하였다. MTT assay와 DAPI staining의 결과를 바탕으로 ID 추출물이 SK-BR-3, MDA-MB-231, AGS 세포에서 apoptosis와 관련한 단백질 발현 양상에 미치는 영향을 확인하기 위해 western blot을 실시하였다. ID 추출물에 의해 SK-BR-3, MDA-MB-231, AGS 세포에서 pro-apoptosis인 Bax 단백질 발현은 농도 의존적으로 증가하였고, antiapoptosis인 Bcl-2 단백질 발현은 SK-BR-3 세포에서는 발현의 변화가 거의 없었지만, MDA-MB-231과 AGS 세포에서는 감소하였다. 세포사멸의 주요임상지표인 Bax/Bcl-2 ratio는 MDA-MB-231세포에서 가장 유의적으로 증가하였다. 또한, 손상된 DNA를 복구하는 단백질인 PARP의 발현은 감소하면서, cleaved-PARP이 증가하였다. 이러한 결과들을 종합하였을 때, YS와 ID 추출물은 TC 추출물에 비해 암세포의 생존을 효과적으로 억제하였고, 특히 ID 추출물은 낮은 농도에서도 암세포의 생존 억제효과가 우수한 것을 확인하였다. 또한, ID 추출물에 의한 유방암세포 SK-BR-3, MDA-MB-231와 위암세포 AGS의 생존율 억제는 apoptosis 유도를 통해 이루어 지는 것으로 사료되며,그 중 유방암세포 MDA-MB-231에서 apoptosis 유도 효과가 뛰어난 것을 미루어 보아, 유방암의 종류와 그 특징에 따른 차이를 보이지만 ID는 향후 유방암 예방 및 치료제로 개발 가능성을 제시하며, 추후 지속적인 연구를 통하여 in vivo에서의 ID 추출물의 항암효과에 대해 심도 있는 연구가 이루어 져야 할 것으로 사료된다.

        This study aimed to evaluate the health concern, eating habits, dietary behavior, and psychological stress among middleaged adults in Chungnam according to their exercise status. A total of 437 adults with a mean age of 42.6 years participated in this study and completed a questionnaire survey. The subjects were divided into one of three groups according to their exercise status: never doing exercise (NDE; n=144), doing exercise once a week (DEO; n=186), and doing exercise twice a week or more (DET; n=107). Results demonstrated significant differences in concern about health, subjective health status, meal regularity, dietary problems, and stress score among the three groups. The DET and DEO groups were more concerned about their health (p<0.001), and had a perception that their health status was better than that of the NDE group (p<0.01). A larger percentage of the DET group had breakfast everyday (p<0.05) and a smaller percentage of the DET group had dietary problems such as irregular mealtime and skipping meals (p<0.01), compared to the other groups. The average stress score of the DET group was significantly lower than that of the DEO and NDE groups (p<0.05). The results reveal that doing regular exercising at least twice a week is associated with higher health concern, more desirable dietary habits, and lower psychological stress in middle-aged adults. These findings support the beneficial effects of regular exercise for health, dietary habits, and stress control.

        본 연구는 다양한 냉동과 해동처리에 따른 계육 가슴살에 natural microflora, 접종된 L. monocytogenes와 C. jejuni 수와 미세구조 변화 구명을 위하여 연구를 수행하였다. −20℃ 송풍식 냉동처리구의 총 호기성 세균과 C. jejuni 수는 4.06 log CFU/g과 4.09 log CFU/g으로 대조구와 비교하여 각각 약 0.7 log CFU/g과 1.0 log CFU/g의 감소를 보였다. 한편, 계육 가슴살 접종된 L. monocytogenes 수는냉동방법과 냉동온도에 따라 유의적인 차이를 보이지 않았다. 4℃와 25℃ 송풍식 해동처리구의 총 호기성 세균수는 3.70 log CFU/g과 4.02 log CFU/g으로 측정되어 대조구와 비교하여 각각 0.72 log CFU/g과 0.40 log CFU/g 감소한 반면 25℃ 유수식 해동처리구의 총 호기성 세균 수는 해동과정에서 균수가 급격히 증가하여 5.78 log CFU/g으로 관찰되었다. 해동 중 C. jejuni 수 변화는 해동방법보다 해동온도에 영향을 받는 것으로 나타났다. 냉동-해동반복 5회 후 총 호기성 세균과 효모 및 곰팡이 수는 감소하여 각각 4.15 log CFU/g과 2.30 log CFU/g을 보였다. 계육 가슴살에 접종된 L. monocytogenes 수는 냉동-해동 반복처리에 유의적인 영향이 없었지만 C. jejuni 수는 냉동-해동 반복 횟수가 증가함에 따라 감소하는 경향을 보였다. 냉동-해동 반복이 증가함에 따라 근섬유 조직을 재 손상시켰으며 특히 냉동-해동 반복 5회 후에는 계육 가슴살 시료의 조직세포 외부뿐만 아니라 내부에서도 공간이 발생하였으며 불균일하게 찢어진 세포가 관찰되었다.

        To develop an elderly diet food that can easily be chewed and swallowed, we manufactured elderly diet food using chicken breast meat with various amounts (0.9, 1.1, 1.3 and 1.5 g) of gelatin used as a viscosity agent, and evaluated their physico-chemical and sensory properties. As the amount of gelatin decreased, the lightness were increased, but the redness and yellowness were decreased. In the texture profile analysis, hardness, springiness, gumminess and chewiness were significantly increased with increased amounts of gelatin, but adhesiveness gradually decreased. Cohesiveness was no significantly difference. Free amino acid contents in elderly diet food using chicken breast meat did not show trend to increase or decrease, but the tyrosine contents were significantly decreased with increased amounts of gelatin. The sensory evaluations including taste, flavor and color were not significantly different. However, the texture and overall acceptance of elderly diet food using chicken meat containing 1.3 g of gelatin had the highest acceptance.

        Ochratoxin A와 aflatoxin은 곰팡이 독소 생산 균주가 자연 발효되는 식품에 오염되거나 발표식품의 종균을 분별없이 선택했을 때 검출 될 수 있다. 본 연구에서는 다양한 배양 환경이 ochratoxin A와 aflatoxin의 생산에 주는 영향에 대해 연구하였다. Aspergillus usamii KFRI 999와 A. awamori KFRI 983의 배양 온도, 배양 배지, 배양 시간을 달리하여 ochratoxin A생산 정도를 분석하였다. 또한 초기 pH, 온도, 배양 시간, 배양 배지를 다르게 하였을 때 A. flavus KACC 41403와 A. oryzae KACC 46471가 생산하는 aflatoxin의 양을 평가하였다. Ochratoxin A와 aflatoxin의 양은 HPLC로 분석하였다. 연구 결과, 곰팡이 독소는 배양 온도에 큰 영향을 받는 것으로 나타났다. A. oryzae KACC 46471는 aflatoxin을 생산하지 않았다. 곰팡이 독소를 생산하는 균주는 모두 30℃에서 가장 높은 독소 생산량을 보였다. A. awamori KFRI 983은 PDA 배지에서 가장 적은 ochratoxin A 생산량을 보였다. 본 연구 결과는 다양한 식품의 ochratoxin A와 aflatoxin 저감화에 유용하게 활용 될 수 있다.

        Carbonic anhydrase is essential for the cellular transportation of hydrogen and bicarbonate ions and plays a key role in a wide variety of physiological processes. Rainbow trout, Oncorhynchus mykiss is an important freshwater fish in aquaculture industry and is known to be one of the most susceptible species to environmental contamination. In this study, carbonic anhydrase was detected in the kidney and intestine of rainbow trout. Carbonic anhydrase was isolated from cytosolic proteins and identified by using SDS-PAGE, isoelectric focusing, and immunohistochemical methods. A specific protein band with molecular weight of 30 kDa and pI of 7.0 was detected by Western blotting. The immunohistochemical results showed that carbonic anhydrase was located at various cells in the kidney and intestine of rainbow trout.

        Cronobactersakazakii is a newly emerging high hazard pathogen, which causes encephalomeningitis and necrotic colitis. Recently, successful biocontrol of harmful microorganisms in several foods through the use of bacteriophages has been reported. In this study, bacteriophages were isolated from kimchi and sewages. Morphological analysis by TEM indicated that phages belonged to the Myoviridae family. In case of heat stability, KCES2 and ESP 2949-2 phages were susceptible to temperatures above 70oC. KCES2 and ESP 2949-2 phages inhibited the growth of C. sakazakii in culture broth. When KCES2 and ESP 2949-2 phages were applied to biofilm-formed C. sakazakii, C. sakazakii was efficiently reduced. Therefore, newly isolated KCES2 and ESP 2949-2 phage for C. sakazakii might effectively reduce C. sakazakii in various foods.

        본 연구에서는 국내 대표 식육인 소, 돼지, 닭, 오리의4종 식육과 염소, 양, 말, 칠면조의 4종 식육을 동시에 신속하게 감별할 수 있는 2 set의 multiplex PCR법을 개발하고자 미토콘드리아 16S RNA에서 종 특이부위를 선발하고 각 종에 대한 특이도를 높이기 위하여 인위적인 미스매치를 주어 프라이머를 제작한 후 8종 식육의 274개시료를 대상으로 특이도와 민감도를 조사하였다. 그 결과소, 돼지, 닭, 오리 모든 시료에서 각각 279, 94, 192, 477 bp의 증폭산물이, 말, 양, 염소, 칠면조의 모든 시료에서 각각 152 bp, 271 bp, 670 bp, 469 bp에서 뚜렷한 PCR 유전자 산물이 확인되어 모든 축종에서 100%의 특이도를 나타내어 축종별 감별력이 우수한 것으로 나타났다. 8종의 축종별로 DNA를 10 ng/μl으로 정량한 후 혼합물을 10배씩 단계 희석하여 반응여부를 조사한 결과, 소, 돼지, 오리에서는 100 fg까지, 닭에서는 1 pg까지 검출됨을 확인할수 있었다. 소, 돼지, 닭, 오리고기를 99.9%, 99%, 90%,70%, 50%, 30%, 10%, 1%, 0.1%의 비율로 혼합한 식육과 83℃ 20분, 100℃ 30분, 121℃ 10분에서 각각 열처리한 가열 혼합육에 대하여 검출한계를 조사한 결과 마지막단계의 희석 비율인 모든 혼합육의 0.1%에서 검출이 가능하였으며, 열처리 혼합육에서는 닭에서는 1% 농도에서소와 돼지의 혼합육에서 0.1% 농도에서 검출되어 민감도가 높음을 확인할 수 있었다. 본 연구에서 개발된 multiplex PCR법은 특이도 및 민감도에 있어서 국내 대표 식육을 감별하는데 있어서 유용한 것으로 평가된다.

        The purpose of this study was to measure concentrations of K+, Na+, Cl- by ionometer with check salt strip, simple salimeter and Ion-selective electrode (ISE) and compare the results of each mensuration; furthermore, the possibility of inferring the Na+ concentration from Cl- concentration of urine and the impact of K+ on the concentration of each ion was examined. The results showed that ISE determined Na+ and Cl- concentrations in the urine are highly interrelated (R=0.9039); in addition, concentrations of Cl-, measured with strip and ISE from urine are highly interrelated (R=0.9338). The concentration of Na+ in urine, inferred by measuring Cl- concentration with strip, has a high relationship (R=0.8580) with the concentration of Na+ in urine, measured by ISE. The results of our study will increase awareness of Na+ intake and the utility of check salt strip, as well as the possibility of inferred Na+ concentration from measures of Cl- concentration as a screening test for reducing sodium intake.

        A lot of research about applications of natural extracts to both food and medicine is in progress. In this paper, we tested the applicability of Rubus crataegifolius leaf extract in functional materials including its anti-inflammatory effects. We investigated the effects of Rubus crataegifolius leaf extract on the LPS-induced production of inflammatory cytokines and mediators in RAW264.7 macrophage cell line. The protein expression of COX-2, iNOS, IL- 4, TNF-α, and IFN-γ, phosphorylation of MAPKs (ERK, JNK, p38) and the activation of NF-κB with Rubus crataegifolius leaf extract were assayed in LPS-stimulated RAW264.7 cells. Pre-treatment with Rubus crataegifolius leaf extract significantly inhibited the protein expression of iNOS and COX-2 in a dose-dependent item and the secretion of inflammatory cytokines such as IL-4, TNF-α, and IFN-γ. Rubus crataegifolius leaf extract significantly reduced the phosphorylation of MAPKs. The NF-κB was strongly suppressed by extracts. Moreover, the pathological phenotypes of contact dermatitis such as erythema, edema, and scabs were significantly decreased in Rubus crataegifolius leaf extract-treated mice. These results indicated that Rubus crataegifolius leaf extract have an antiinflammatory effect, and that this might be used as a supplement for the functional materials.

        본 연구에서는 V. parahaemolyticus를 신속정확하게 고감도로 검출하기 위해 단클론성 항체를 개발하고 이를 이용하여 일반적인 효소면역분석법(ELISA)과 효소면역분석법의 낮은 민감도를 보완할 수 있는 면역선택여과법(ISF)을 개발하여 민감도 및 효율성을 비교분석 하였다. 먼저heat killed V. parahaemolyticus (HKVP)를 준비하여 7주령 BALB/c mouse에 면역한 후 세포융합 및 클로닝을 통해 HKVP 4H9-9번 및 16번 2종의 hybridoma cell을 확보하였다. Western blot을 통해 보다 특이성이 높은 것으로 확인된 HKVP 4H9-9번 항체를 대량 생산정제하여 분석법을 확립한 결과 검출한계가 ID-ELISA법은 106 cell/mL, ISF법은 5 × 101 cell/mL로 나타나 ISF법의 민감도가 매우 높은 것으로 확인되었다. 분석효율성은 ID-ELISA법의 경우분석을 위해 9단계의 과정을 거쳐 총 16시간의 분석시간이 소요된 반면, ISF법의 경우 3단계의 과정을 거쳐 1시간 이내에 분석을 완료할 수 있는 것으로 확인되었다. 교차반응성의 경우 두 분석법 모두 일부 Vibrio spp. (IDELISA법:V. alginolyticus, ISF법: V. vulnificus)과 S. aureus에 반응성이 있는 것으로 확인되었지만 V. parahaemolyticus에 보다 강한 반응성을 나타내었기 때문에 V. parahaemolyticus에 특이적인 분석법인 것으로 확인되었다. 특히 ISF법은 ELISA법 보다 민감도가 높고 분석에 소요되는 시간도 짧아 V. parahaemolyticus를 신속하게 분석하는데 활용이 가능할 것으로 판단된다.

        Mulberry (oddi) is one of the most popular functional foods with many physiological components. This study investigated and compared the nutritional compositions and physiological activities of four mulberry fruits from Morus albo L. including Cheongilppong, Iksuppong, Suwonppong and Cheongsuppong (a new cultivar from Chungcheongbuk-do). To analyze the nutrient contents, mulberry fruits were freeze-dried. The results showed that the proximate compositions of the four mulberry cultivars ranged from 9.61~14.11% for moisture, 8.28~11.90% for crude protein, 3.70~4.86% for crude ash, 4.28~5.54% for crude lipid and 7.46~10.78% for crude fiber. The above proximate contents of mulberry cultivars were not significantly different. However, Cheongsuppong had the highest content of reducing sugar, 74.7%. Chungsuppong and Iksuppong showed higher contents of total polyphenol and anthocyanin than other mulberry cultivars, while all mulberry cultivars showed high antioxidant activities. Hypoglycemic effect had a slightly higher level in Suwonppong and Cheongsuppong than in the other samples. Taken together, the new cultivar “Cheongsu” oddi can be suggested as a potential source of functional food.

        LC-MS/MS를 이용한 말라카이트 그린 유효성 검증결과 결정계수(R2)가 0.9996이상으로 높은 직선성을 보여주었으며 회수율은 어종에 따라 69.8%에서 93.9%범위를 보였다. 검출한계는 0.013 μg/kg, 정량한계는 0.038 μg/kg으로 CODEX의 가이드라인을 만족하였다. 수거된 어류 200건에서 말라카이트 그린을 분석한 결과, HPLC-DAD로는 1건이 검출되었으나, LC-MS/MS로 분석시 검출률은 18.5%였다. 본연구 결과, LC-MS/MS를 이용한 말라카이트 그린 분석법이 우리나라에서 많이 소비되는 담수어종에서 분석에 적합하며, 1건을 제외한 모든 시료에서 식품공전의 잔류허용기준 이하로 말라카이트 그린이 검출되어서 안전한 것으로 나타났다. 다만 식품공전의 검출한계 이하로 말라카이트 그린이 검출되는 빈도가 18.5%임을 볼 때, 지속적으로 감시하고 모니터링 할 필요가 있음을 시사한다.

        This study was conducted to develop and evaluate an appropriate control device for a purge type controlled atmosphere (CA) storage in Korea. To determine ideal performance, oxygen and carbon dioxide control capability and airtightness were analyzed according to the postharvest management manual of CA storage of Fuji apples. In shortened experiments for CA storage, the condition was delayed CA at 0-0.5oC for three days and stored at 0.1-0.5% carbon dioxide levels for 3 days and then further stored 6 days under 1% carbon dioxide. As a result, the temperature control range of a developed CA container was 0.0-0.5oC, and the relative humidity was more than 90%, except for the defrosting step for the freezer during the storage period. The rate of pressure reduction for the CA container in the negative and positive pressure states was 0.45 and 0.21 mmH2O/min, respectively, and it was twofolds higher than standard airtightness for CA storage. After nitrogen injection, oxygen concentration was achieved at 2%, and carbon dioxide concentration was maintained at 0.1-0.5% for 6 days. Afterwards, carbon dioxide levels were tightly controlled between 0.1-1.0%. These results suggest that a developed purge type CA container could be effective in commercially maintaining the quality of agricultural products.

        This study was performed to develop and evaluate cookies prepared with various amounts (0 g, 12.5 g, 25 g and 37.5 g) of mealworm powder to serve as not only sports snacks but also for the general population as a new protein source. In the chromaticity of mealworm cookies, lightness and yellowness decreased whereas redness increased with more mealworm powder. The hardness of the mealworm cookies increased as more mealworm powder was added. In sensory evaluation, higher preference was shown with the measure of color, taste, and overall preference for mealworm cookies prepared with 50% mealworm powder (MP2). For the general composition of MP2, the moisture, carbohydrate, crude protein, crude fat and crude ash were higher compared with the control cookie. For the mineral contents of MP2, the contents of Ca (33.44 mg/100 g), P (225.13 mg/100 g), Mg (82.21 mg/100 g), Na (246.22 mg/100 g), and K (503.49 mg/100 g) were higher compared with the control cookie. The essential amino acids (valine, isoleucine, leucine, methionine, threonine, phenylalanine, and histidine) in MP2 were also higher compared with the control cookie. It was concluded that MP2 can be used as a new protein source for not only the maintenance of muscle but also for the prevention of muscle loss in old people.

        Twenty four covered barley cultivars of Korea and subsequently produced malts were evaluated for quality characteristics, diastatic power (DP), and enzymatic actvity. Protein, amylase, starch and β-glucan content of covered barley cultivars were 8.2-12.8%, 4.2-19.1%, 47.9-59.7%, and 4.1-7.4%, respectively. The DP of malt during malting showed a maximum value in Day 4, when the bud length was 2.0-2.5 times longer than the length of the grain itself. α-Amylase activity was very low but increased significantly after Day 4, showing greatest activity on Day 6 (185.5-206.1 unit/g). β-Amylase activity increased as the malting progressed and a considerable amount was detected in barley grain on Day 6 (898.6-1591.6 unit/g). The potential diastatic power (PDP) of various cultivar grains ranged from 89.7 °L to 169.3 °L. The PDP of raw barley had strong correlation with β-amylase of raw barley (r=0.977, p<0.001) while the DP of malted barley was also correlated with the β-amylase activity of raw barley (r=0.654, p<0.001). Since the PDP of raw barley and the DP of malted barley are correlated (r=0.644, p<0.001), the PDP and β-amylase activity of raw barley are useful factors in predicting the DP of malted barley.

        The viscosity, overrun, melting-down, moisture, crude fat, total sugar, and color of rice powder and puffed rice powder ice cream, following the addition of α-amylase, were investigated. For identical grain types, the gelatinization degree increased with puffing, and within the same treatment, the short grain was higher than the long grain. Viscosity dropped with increasing α-amylase at the same concentration and grain type, excluding 0.0%, the rice powder was higher than the puffed one, and for the same concentration and treatment, the short grain was higher. The overrun was highest at 0.2%, and for the same concentration and treatment, the short grain exhibited higher overrun. Higher melting-down was observed in puffed and lower viscosity ice cream mix. No significant difference was found in moisture with enzyme concentration. Regardless of puffing, the short tended to have a higher moisture. No difference was noticed in crude fat by concentration, grain type, or puffing. The total sugar was higher with increasing α-amylase; at the same concentration, puffed tended to be higher. The hunter “L” and “b” increased with α-amylase, while the “a” value dropped. At the same concentration and grain type, the “b” values decreased with puffing (p<0.05).

        To enhance the physiological activities of roasted coffee (RC), 30 kinds of green coffee beans (GCB) with different cultivating areas and varieties were fermented with Monascus ruber mycelium (MR) by solid-state culture. After the dried MR-fermented GCB was subjected medium roasting, each RC was extracted with hot-water. Among the hot-water extracts, the highest yield was the hot-water extract of RC from MR-fermented Indonesia Mandheling GCB (15.5%). However, the hot-water extract of RC from MR-fermented Ethiopia Sidamo GCB showed significantly higher polyphenolic contents (3.08 mg GAE/100 mg) and ABTS free radical scavenging activity (25.41 mg AEAC/100 mg). Meanwhile, the hot-water extract of RC from MR-fermented Vietnam Robusta GCB showed not only the effective inhibition of TNF-α level (73.7% inhibition of LPS-stimulated control) from LPS-stimulated RAW 264.7 cells but also significant inhibition of lipogenesis (63.5% inhibition of lipid differentiation control) in 3T3-L1 pre-adipose cells. In conclusion, these results suggest that roasted coffees from Ethiopia Sidamo and Vietnam Robusta green coffee beans fermented with Monascus ruber mycelium using solid-state culture could have industrial applications as functional coffee beverages.
