        Soybean [Glycine max (L.) Merr.] seed is an important dietary source of protein, oil, carbohydrates, isoflavones, and other nutrients for humans and animals. Raffinose and stachyose are the main antinutritional factors in soybean seed. They are carbohydrates belonging to the raffinose family of oligosaccharides, which are not readily digested in humans and cause flatulence or diarrhea. The genetic reduction of the raffinose and stachyose contents in mature soybean seeds will improve the nutritional value of soybean. The objective of this research was to evaluate agronomic traits with 10 F6 strains selected from breeding populations derived from a cross among seven parents. The contents of raffinose and stachyose in mature seeds were detected by high-performance liquid chromatography. Agronomic traits such as flower color, flowering date, harvesting date, lodging, plant height, seed coat color, hilum color, 100 seed weight, and yield were evaluated. Ten intermediate parents showed low raffinose and stachyose contents. The intermediate parent 883-1 had a small seed size, six intermediate parents (15A1, 15D1, RS-5, RS-33, RS-64, and RS-70) had a medium seed size, and two intermediate parents (14G20 and RS-21) had a large seed size. The intermediate parent RS-21 had a black seed coat and a green cotyledon. Four intermediate parents (883-1, 14G20, RS-5, and RS-21) had elite agronomic traits. The new intermediate parents developed through this study will be used to develop improved soybean cultivars with low contents of raffinose and stachyose.

        Soybean [Glycine max (L.) Merr.] is an important source for protein, oil, carbohydrates, isoflavones, and many other nutrients to humans and animals. Lipoxygenase protein is the main antinutritional factor in mature soybean seed. Soybean cultivar, "Choseon" with yellow medium seed size and free of lipoxygenase 2,3 protein was developed. "Choseon" was selected from the population derived from the cross between cultivar, "Jinpum 2" and germplasm, PI408155. "Choseon" has purple flowers, tawny pubescence, a determinate growth habit, and light yellow pods at maturity. The seed of "Choseon" has yellow hilum and yellow seed coat color. Seed protein and oil content on a dry weight basis were 34.2 and 16.9%, respectively. "Choseon" has shown a resistant reaction to soybean necrosis, soybean mosaic virus, Cercospora leaf spot and blight, black root rot, pod and stem blight, and bacterial pustule. "Choseon" matured in 5 October with plant height of 51 cm and a 100-seed weight of 15.1 g. Average yield of "Choseon" was 2.66 (Ton/ha) on the regional yield trials of 2013 at 4 locations. "Choseon" has been registered as a soybean cultivar (registration number: 5985, registration date: April 11, 2016) by Korea Seed & Variety Service, Republic of Korea.

        Soybean cultivar ‘Seonpung’ was developed for soy-paste and tofu. Suwon 224 and YS1325-3S-2 were crossed in 2003 and selected from F3 to F5 by pedigree method. The preliminary yield trial (PYT) and advanced yield trial (AYT) were conducted from 2009 to 2010, and regional yield trial (RYT) in twelve regions was conducted from 2011 to 2013. In RYT, ‘Seonpung’ was stable in variable environments and a high yield cultivar. ‘Seonpung’ is determinate, white flower, yellow spherical seed and yellow hilum. Flowering date and maturity date were Aug. 5 and Oct. 19, respectively. Plant height was similar to ‘Daewonkong (standard cultivar)’. However ‘Seonpung’ has higher node number (16) and seed weight (25.9g/100-seed weight) than ‘Daewonkong’ (14 and 24.2g/100-seed weight). ‘Seonpung’ is resistant to root rot, and it also has high level of resistance to bacterial pustule and soybean mosaic virus. The yield of tofu of ‘Seonpung’ was 241%, and noticeably lighter, and solidity was higher than ‘Daewonkong’. Soybean malt scent, fermented soybean yield and γ-polyglutamic acid (γ-PGA) of ‘Seonpung’ were 4, 181% and 31.7㎎/g. The yield in adaptable regions was 340kg/10a (21% increase compared to ‘Daewonkong’). ‘Seonpung’ is expected to be cultivated and used widely for soy-paste and tofu. (Registration number: 5931)

        The effects of elevated atmospheric CO2 on growth and photosynthesis of soybean (Glycine max Merr.) were investigated to predict its productivity under elevated CO2 levels in the future. Soybean grown for 6 weeks showed significant increase in vegetative growth, based on plant height, leaf characteristics (area, length, and width), and the SPAD-502 chlorophyll meter value (SPAD value) under elevated CO2 conditions (800 μmol/mol) compared to ambient CO2 conditions (400 μmol/mol). Under elevated CO2 conditions, the photosynthetic rate (A) increased although photosystem II (PS II) photochemical activity (Fv/Fm) decreased. The maximum photosynthetic rate (Amax) was higher under elevated CO2 conditions than under ambient CO2 conditions, whereas the maximum electron transport rate (Jmax) was lower under elevated CO2 conditions compared to ambient CO2 conditions. The optimal temperature for photosynthesis shifted significantly by approximately 3°C under the elevated CO2 conditions. With the increase in temperature, the photosynthetic rate increased below the optimal temperature (approximately 30°C) and decreased above the optimal temperature, whereas the dark respiration rate (Rd) increased continuously regardless of the optimal temperature. The difference in photosynthetic rate between ambient and elevated CO2 conditions was greatest near the optimal temperature. These results indicate that future increases in CO2 will increase productivity by increasing the photosynthetic rate, although it may cause damage to the PS II reaction center as suggested by decreases in Fv/Fm, in soybean.

        전통장류로부터 분리 된 효소 활성 및 probiotic효과가 우수한 유산균을 접종하여 제조된 콩 발효물의 품질특성을 분석하였다. 일반세균수는 유산균을 접종한 콩 발효물이 4.10×109-7.75×109 CFU/mL으로 나타났다. 유산균수를 측정 결과 유산균을 접종한 콩 발효물은 2.85×109-4.35×109 CFU/mL으로 높게 나타났다. α-Amylase 활성 측정 결과 I13과 JSA22를 접종한 콩 발효물은 1.05±0.91, 3.13±1.39 unit/mL으로 가장 낮았다. 또한 JSB22는 12.56±1.39 unit/mL으로 유산균 접종 콩 발효물 중 가장 높았다. Protease 활성 측정 결과, control이 333.13±17.68 unit/mL으로 가장 높았으며, 유산균을 접종한 콩 발효물은 JSB22를 접종한 콩 발효물이 166.67±2.37 unit/mL으로 높았다. 환원당 함량을 측정한 결과 유산균 접종 콩 발효물 중 JSB22가 1.23±0.035%로 가장 높았다. 아미노태 질소 함량 또한 JSB22가 94.52±3.86 mg%으로 높게 나타났다. 아미노산함량을 측정한 결과 glutamic acid, aspartic acid, arginine, leusine 순으로 많이 함유하고 있었다. 유기산 함량을 분석한 결과 lactic acid, oxalic acid가 많이 측정되었다. 향기분석 측정 결과 총 39종(ester류 2종, ketose 5종, alcohol 7종, hydrocarbons 14종, heterocyclic compounds 3종, acid 4종, other 5종)이 검출되었다. 세 가지 균주에서 공통적으로 benzene(81.43-98.54 ppm)과 acetic acid, ethyl ester(44.79-56.74 ppm)가 많이 검출되었다. 본 연구는 전통장류 유래 기능성 우수 유산균주를 이용한 콩발효물의 기초적 품질특성에 관한 내용으로 미생물학적 및 이화학적 관련 특성을 분석하여 제시하였다. 유산균 이용 생물전환에 의한 다양한 기능성 물질이 생성 되었을 것으로 생각된다. 본 연구결과가 기능성 콩 발효소재 개발의 좋은 기초자료가 될것으로 생각되며, 장류 품질개선 기능성 스타터 개발에 활용 될 수 있을 것으로 기대된다.

        본 연구에서는 전통 재래식 콩된장의 제조공정을 과학적으로 해석하기 위한 연구의 일환으로 생메주의 건조 및 발효과정에 있어서 수분함량과 단백질 분해에 의한 아미노산 생성과정을 근적외 분광법으로 쉽게 판단 할 수 있는지 그 가능성을 조사하였다. 생메주를 일정한 크기(3×4×3 cm)로 잘라서 수분이 30%가 되도록 25℃ 항온기에 3주간 발효시킨 모델계 발효메주를 만들어 가시광-근적외 스펙트럼을 측정하여 해석하였다. 발효가 진행됨에 따라 수분이 감소되면서 수분과 관련이 있는 1,430 nm대의 흡광도가 감소 및 시프트 되는 것이 관찰되었고, 발효기간이 길어질수록 아미노산과 관련이 있는 2,050 nm대의 흡광도가 증가하는 것을 관찰 할 수 있었다. 전통 재래식 발효메주를 속부분과 겉부분으로 분리하여 1,000-2,250 nm에서의 가시광 스펙트럼과 근적외 스펙트럼 데이터로 판별분석한 결과, 겉부분과 속부분이 잘 판별되었고, 가시광 보다는 근적외 영역에서 잘 구분되었다. 이들 전통메주중의 아미노산 농도를 비파괴 측정하기 위해 PLSR한 결과 R²은 0.9 이상이며, RMSECv 0.23%로 나타났으며, 전통메주중의 수분함량도 R² 0.81, RMSECv 0.83%이어서 근적외 분광법으로 전통메주의 품질을 비파괴 측정 가능할 것으로 판단되었다.

        The purpose of this study was to investigate the effect of a fresh chlorella powder on yield, quality and self-life of organic soybean tofu. After added with a fresh chlorella powder, yields, physical properties, color, shelf-life and sensory properties of the organic soybean tofu added with chlorella powder was compared to the properties of soybean tofu. The yield of soybean tofu increased with the addition of more than 1% chlorella powder. Hardness of the organic soybean tofu with chlorella significantly increased compared to that of the soybean tofu. However, Hunter’s color values of L, a, and b decreased by increases in the chlorella powder concentration. Total microbial population of the soybean tofu was higher than that of the organic soybean tofu with chlorella. However, pH of chlorella tofu decreased during storage period at 4℃. After 7 days storage at 4℃, the total microbial population decreased significantly with the addition of 2% chlorella powder. The L and a value of chlorella tofu was increased. On the other hand, the b value of chlorella tofu was decreased during storage. The results revealed that a fresh chlorella powder was a useful material to improve yield, quality and storage condition of the organic soybean tofu.

        The purpose of this study was to investigate changes in the amino acid content and physicochemical properties of Cheonggukjang prepared by using various soybean cultivars (Daewon, Deapung, Seadanbeak, and Taekwang) and a functional microorganism (Bacillus subtilis HJ18-9). These soybeans were conventional Cheonggukjang (control) and Cheonggukjang fermented with Bacillus subtilis HJ18-9 (treated). The moisture contents of steamed, control, and treated soybean were 62.45~67.12%, 63.28~67.14%, and 64.50~66.87%; amino-type nitrogen contents were 6.53~24.25 mg%, 27.63~122.09 mg%, and 37.29~133.48 mg%; and ammonia-type nitrogen contents were 26.92~47.95 mg%, 45.45~156.36 mg%, and 28.02~121.13 mg%, respectively. The umami taste associated with several amino acids (aspartic acid and glutamic acid) in Cheonggukjang was lower than that for steamed soybeans, while the bitter taste from amino acids (methionine, valine, isoleucine, leucine, and phenylalanine) was higher than that for steamed soybeans. The result of sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis showed that the molecular weight of steamed soybeans was less than 100 kDa, while control and treated groups showed low molecular weights below 34 kDa, confirming their protein hydrolysis to small molecular weight. These results are information for developing functional fermented soybean paste and diversification using soybean cultivars.

        Early maturity of soybean (Glycine max (L.) Merr.) is an important character for double cultivating system with winter crops such as onion or garlic in southern area of Korea. Soybean cultivars released in the past, especially ‘Keunolkong’ was widely cultivated but it was weak at abiotic stress and low yield potential because of too short growing period and short stem height. ‘Hanol’, a new early maturing cultivar showing better abiotic stress tolerant and higher yield potential, was developed from the cross between SS91408 and ‘Hwaeomputkong’, and released in 2009. ‘Hanol’ is, compared to ‘Keunolkong’, Hanol’s growing period is five days longer and its height is 5cm higher. In addition, it showed better excess-water tolerant and 26.5g 100-seed weight. At regional adaptation yield trials (RYT) in six regions from 2007 to 2009, ‘Hanol’ showed 2.04 tons of seed per hectare, 14% higher than ‘Keunolkong’ (1.80ton/ha). ‘Hanol’ is promising for double cropping system with higher yield.

        This study was investigated to develop mass evaluation system for the contents of crude protein, oil and fatty acid in soybean germplasm using NIRS. NIRS equations were created with 345 soybeans, multiple correlation coefficients of crude protein, oil, palmitic, stearic, oleic, linoleic and linolenic acid between data obtained from NIRS and quantitative analysis were 0.983, 0.969, 0.592, 0.514, 0.978, 0.961 and 0.957, respectively. Equation statistics indicated that contents of crude protein, oil and unsaturated fatty acid except palmitic and stearic acid in soybean seed were suitable for determination by NIRS. Those NIRS equations were applied to examine crude protein, oil and unsaturated fatty acid of 854 soybean landraces from Korea. The average contents and ranges of crude protein and oil were 39.2% with a range of 33.7-47.0% and 15.0% with a range of 9.8-20.3%, individually. In addition, those of oleic, linoleic and linolenic acid were 21.4% with a range of 12.1-30.2%, 55.6% with a 47.8-62.3% and 8.1% with a range of 5.9-10.7% respectively. We conducted quantitative analysis to reconfirm with IT154552 (45.1%) and IT023955(46.9%) above 45% of crude protein, the results were similar from NIRS (45.2%, 47.0%). NIRS data for protein from this study made no difference with lab data, which would be useful for mass evaluation. There was negative correlation (-0.203) between crude protein and oil, positive correlation (0.379) between crude oil and oleic acid, and significantly negative correlation (-0.879) between oleic and linoleic acid.

        The present study was conducted to establish the optimal fertilization type and method for no-tillage during the first year of No-tillage (NT) and Conventional-tillage (CT) practices for soybean, using different types of fertilizers. In this experiment, the culm length and stalk diameter showed a greater response to fertilization with surface irrigation than to conventional fertilization. The fastest flowering period (July 28) occurred using chemical fertilization applied via subsurface irrigation. Comparing maturation based on growth characteristics and flowering date revealed that fertilization with subsurface irrigation was more effective for the growth of crops than other methods. Regarding yield, there was no significant difference between livestock and chemical fertilizers in subsurface irrigation, but there were significant differences between these fertilizers when using conventional fertilization methods. Based on the results, livestock fertilizer with subsurface irrigation effectively enhanced crop quantity. Nitrogen absorption of plants using subsurface irrigation was more effective than that using conventional fertilization. Regarding phosphorus absorption of plants, chemical fertilizers showed higher absorption than did livestock fertilizers for both subsurface irrigation and conventional fertilization. Unlike nitrogen, phosphorus was highly absorbed using conventional fertilization. Absorption of phosphorus and potassium were similar but phosphorus was not absorbed using livestock fertilizers applied either using subsurface irrigation or with conventional fertilization.

        A soybean cultivar for soy-paste, ‘Hwangkeumol’, was developed from the cross between SS92414 (crossing line of ‘Pokwangkong’ and ‘Suwon163’) and ‘Hwaeomputkong’ by soybean breeding team in the National Institute of Crop Science (NICS) in 2010. A promising line, SS99409-2B-11-5-4, was selected and designated as the name of ‘Milyang202’. It was prominent and had good result from regional adaptation yield trials for three years from 2008 to 2010 and released as the name of ‘Hwangkeumol’. It has a determinate growth habit, white flower, gray pubescence, yellow seed coat, yellow hilum, spherical seed shape and large seed (28.6 grams per 100 seeds). Maturity date of ‘Hwangkeumol’ was as early as September 15th. Therefore it is suitable for double cropping system. ‘Hwangkeumol’ was resistant to bacterial pustule and resistant to soybean mosaic virus, the major soybean diseases in Korea and tolerant to lodging in fields. The average yield of ‘Hwangkeumol’ was 2.51 ton per hectare in the regional yield trials carried out for three years from 2008 to 2010.

        A soybean cultivar for soy-paste, ‘Uram’, was developed from the cross between ‘Suwon190’ and ‘SS99244’ (Shinpaldal-2 X T243) by soybean breeding team at the National Institute of Crop Science (NICS) in 2010. A promising line, SS00232-B-B-3SSD-9-4-1-1, was selected and designated as the name of ‘Milyang188’. It was prominent and had good result from regional adaptation yield trials (RYT) in southern area of Korea for three years from 2008 to 2010 and released as the name of ‘Uram’. It has a determinate growth habit, white flower, gray pubescence, yellow seed coat, yellow hilum, spherical seed shape and large seed (25.8 grams per 100 seeds). ‘Uram’ was found to be resistant to bacterial pustule and soybean mosaic virus, the major soybean diseases in Korea. The lowest pod height of ‘Uram’ was 19cm and it will be able to reduce seed loss during mechanical harvesting. The average yield of ‘Uram’ is 3.27 ton per hectare in southern double cropping area. Through these results, ‘Uram’ is soybean cultivar that is favorable for mechanization harvesting, resistant to diseases and highly yield.

        Growth of 6-year old ‘Niitaka’ pear (Pyrus pyrifolia Nakai) trees and control of insect and disease occurrences were compared between fermented soybean extracts and rain-shelter system for two years. Foliar application of fermented soybean extracts was applied at 6 times as a pre-experiment in the open-field in 2013, with a rain-shelter system in 2014. Fermented soybean extract treatment increased foliar concentrations of approximately 0.46% T-N, 0.17% K, 0.19% Ca, and 0.06% Mg in 2013 compared to the control, with similar macro-nutrients between the control and soybean extract treatment observed in 2014. Rain-shelter system increased foliar concentrations of T-N, Ca, and Mg compared to the open-field. There were no significantly different between the control and soybean extract treatment for number of leaves per fruit, leaf dry weight, phytotoxicity, and completed shoot growth on August during the two years. Rain-shelter system increased leaf dry weight and did not affect phytotoxicity in the leaves. Fruit quality parameters were mostly similar to control and soybean extract treatment for two years, with higher fruit firmness observed for soybean extract treatment. Rain-shelter system advanced 4 days of harvest dates, and increased approximately 7.0 ton fruit yield per ha, 20 g mean fruit weight, and fruit soluble solid contents compared to open-field in 2014. Soybean extract treatment little suppressed occurrence of disease and insect on the leaves and fruits in both years. Rain-shelter system increased occurrence of Venturia nashicola on the leaves and to 63.8% of Gymnosporangium asiaticum on the fruits in 2014. Strong winds and storms in May elevated relative humidity in the rain-shelter system and caused high infection of the disease occurrence, requiring for an additional green control method. Soybean extract treatment little affected tree growth and would have initiated for a long-term study to evaluate tree physiological characteristics. Rain-shelter system improved fruit productivity and advanced harvest dates, which could have been more effective facility at a Thanks Giving Day between middle and end of September.

        A double-cropping system with soybean (Glycine max) following the cultivation of potato, garlic, and onion is widely adopted in the southern region of Korea. For this system, marginal dates for planting must be determined for profitable soybean yields, because the decision to plant soybean as a second crop is occasionally delayed by harvest of the first crop and weather conditions. In order to investigate the effect of planting date on soybean yield, three cultivars (early and late maturity) were planted on seven different dates from May 1 to July 30 in both paddy and upland fields across 2012 and 2013. Soybean yields were significantly different among the planting dates and the cultivars; however, the interaction between cultivar and planting date was not significant. Based on linear regression, the maximum yield of soybean was reached with a June 10 planting date, with a sharp decline in yield for crops planted after this date. The results of this study were consistent with those of a previous one that recommends early and mid-June as the optimum planting period. Regardless of soybean ecotype, a reduction in yield of greater than 20% occurred when soybean was planted after mid-July. Frost during soybean growth can reduce yields, and the late maturity cultivars planted on July 30 were damaged by frost before completing maturation and harvest; however, early maturity cultivars were safely harvested. For sufficient time to develop and reach profitable yields, the planting of soybean before mid-July is recommended.

        A new soybean cultivar ‘Wonhyun’, was developed by mutation breeding technique using a 250 Gy gamma ray at Korea Atomic Energy Research Institute (KAERI) in 2010. ‘Wonhyun’ has black seed coat and much better agronomic performance than original variety ‘Paldal’. Their total yield (177.1kg/10a) is much higher than that of ‘Paldal’ (126.9 kg/10a). Also, 100 seed weight of Wonhyun was 27g compared to ‘Paldal’ (13.7g). Contents of 4 essential amino acids such as aspartic acid, glutamic acid, lysine, arginine and unsaturated fatty acid including linoleic and linolenic acid have much higher than ‘Paldal’. This cultivar is good for cooking with rice as improved functional ingredient soybean.

        Soyasaponin I, II, III and V contents were investigated in seed, cotyledons and sprouts of soybean (Glycine max (L.) Merill) subjected to germination over five days. High-performance liquid chromatography coupled with tandem mass spectrometric (HPLC-MS/MS) method was used to evaluate the content of soyasaponins. Soyasaponins contents were different according to the varieties. Germination of soybeans dramatically increased soyasaponin contents in soybean sprouts in a time-dependent manner. Cotyledons had a higher contents of soyasaponins compared to dried seed (p<0.05). After five days of germination, Soyasaponin I and II increased 10 times higher after germination. Soyasaponin I and II are major metabolites in cotyledons and hypocotyls. Soyasaponin III and V were also detected in seed and increased depended on the germination stage. Soyasaponin V was at its highest levels in the hypocotyl, almost 7 times higher than the initial content in soybean seeds. Therefore, the germination of soybean sprouts significantly increased soyasaponin content.

        A new soybean variety, ‘Joongmo 3009’ (Milyang 222) was developed at the National Institute of Crop Science (NICS) in 2012. ‘Joongmo 3009’ was released by pedigree selection from the cross between ‘Cheongja 2(Milyang 121)’ and ‘Daemangkong’. It has determinate growth habit, white flower, brown pubescence, brown pod color, green seed coat, green cotyledon, spherical seed shape, oval leaf shape and large seed size (29.3 grams per 100 seeds). It was late 16 days in maturing date than the check cultivar ‘Cheongjakong’. The average yield of ‘Joongmo 3009’ was 2.91 ton per hectare, which was higher 36 percentage than the check variety, in the regional yield trials carried out in three adaptable locations of Korea from 2010 to 2012.         Plant regeneration via organogenesis and somatic embryogenesis was investigated in Korean soybean cultivars including Cheongja 3, Jinpumkong 2, Taekwangkong and Uram. Cotyledon, cotyledon+hypocotyl and hypocotyl segments of 7-day-old seedlings were cultured on MS medium containing various concentration (0, 1, 2 and 4 ㎎/L) of BA and TDZ. The results showed that MS medium supplemented with BA 2.0 ㎎/L yielded the highest shoot formation ratio of 83.3%. In 4 cultivars, Taekwangkong showed the highest ratio of shoot formation. When various sizes of immature cotyledons (S: 1∼ 2 ㎜, M: 3∼5 ㎜, L: 6∼8 ㎜) were tested on MS medium containing 2,4-D 40 ㎎/L for somatic embryogenesis, the optimum size for embryogenic callus induction was 3∼5 ㎜ in length of immature cotyledons. In 4 cultivars, Taekwangkong showed the highest percentage of embryogenic callus induction. The results indicate that Taekwangkong is the best soybean cultivar for plant regeneration via organogenesis and embryogenic callus induction among the 4 cultivars.

        In order to investigate the utilization of the Omija (Schizandra chinensis Baillon) extract as a natural coagulant for manufacturing soybean curd, the quality characteristics of white (Baktae) and black (Seoritae) soybean curds, coagulated by the Omija extract or MgCl2, were evaluated. Crude protein (6.14±0.30 and 6.25±0.18%, respectively) and crude lipid (10.86±1.74 and 11.29±1.69%, respectively) contents of white and black soybean curds coagulated using the Omija extract were higher than those coagulated using MgCl2. Black soybean curds coagulated using the Omija extract showed higher L, a, and b values than those using MgCl2. The most abundant amino acid in white and black soybean curds coagulated using the Omija extract was arginine (3.74 and 3.71 mg/100 g, dry basis, respectively). The amounts of Ca, K, Mg, and Na were the highest in both soybean curds prepared with the Omija extract. The sensory evaluation (color, flavor, taste, texture, and overall preference) showed that white and black soybean curds coagulated using the Omija extract were more preferred than those produced using MgCl2. The results suggested that using the Omija extract as a natural coagulant agent could improve the quality and sensory characteristics of soybean curds.
