Background: Brassica oleracea var. italica (broccoli), a rich source of antioxidants,
can prevent various diseases and improve human health. In this study, we investigated
the antioxidative effects of broccoli sprout extract on oxidative stress induced by
lipopolysaccharide and cisplatin in cell and organ tissue models.
Methods: Antioxidative effect of BSE was evaluated using DPPH and ABTS in RAW
364.7 cells, and effects of BSE on testes were investigated using Cisplatin-induced
testicular damage model with an in vitro organ culture system.
Results: The DPPH assay showed that the antioxidant activity of the alcoholic
broccoli sprout extract was higher than that of the water extract. Additionally, the
expression levels of antioxidation-related genes, Nrf2 , Gsr , HO-1, and catalase , were
significantly increased in broccoli sprout extract-treated RAW 264.7 cells, and the
extract suppressed lipopolysaccharide-induced mitochondrial dysfunction. Based on
the results in the RAW 264.7 cell culture, the antioxidative effects of the extracts were
investigated in a mouse testis fragment culture. The expression of Nrf2 , HO-1 , and
Ddx4 was clearly decreased in cisplatin-treated mouse testis fragments and not in
both broccoli sprout extract- and cisplatin-treated mouse testis fragments. In addition,
the oxidative marker O-HdG was strongly detected in cisplatin-treated mouse testis
fragments, and these signals were reduced by broccoli sprout extract treatment.
Conclusions: The results of this study show that broccoli sprout extracts could serve
as potential nutraceutical agents as they possess antioxidant effects in the testes.
Kale (Brassica oleracea var. acephala) is one of the most frequently consumed leafy vegetables globally, as it contains numerous nutrients; essential amino acids, phenolics, vitamins, and minerals, and is particularly rich in glucosinolates. However, the differences in the biosynthesis of glucosinolates and related gene expression among kale cultivars has been poorly reported. In this study, we investigated glucosinolates profile and content in three different kale cultivars, including green (‘Man-Choo’ and ‘Mat-Jjang’) and red kale (‘Red-Curled’) cultivars grown in a vertical farm, using transcriptomic and metabolomic analyses. The growth and development of the green kale cultivars were higher than those of the red kale cultivar at 6 weeks after cultivation. High-performance liquid chromatography (HPLC) analysis revealed five glucosinolates in the ‘Man-Choo’ cultivar, and four glucosinolates in the ‘Mat-Jjang’ and ‘Red-Curled’ cultivars. Glucobrassicin was the most predominant glucosinolate followed by gluconastrutiin in all the cultivars. In contrast, other glucosinolates were highly dependent to the genotypes. The highest total glucosinolates was found in the ‘Red-Curled’ cultivar, which followed by ‘Man-Choo’ and ‘Mat-Jjang’. Based on transcriptome analysis, eight genes were involved in glucosinolate biosynthesis. The overall results suggest that the glucosinolate content and accumulation patterns differ according to the kale cultivar and differential expression of glucosinolate biosynthetic genes.
This study evaluates the gastroprotective effect of cabbage extract with sulforaphane content of 5.19 mg/L and Smethylmethionine content of 469.28 μg/L. In vitro, the lipopolysaccharide (LPS)-treated group had an increased NO activity compared to the normal group, and the concentration of NO was reduced when the cabbage extract was treated in the dose manner. The level of IL-6 induced by LPS was dose-dependently reduced when the extract was treated. The cabbage extract concentration was orally administered in rats at 5.75 mg/kg, 11.5 mg/kg, and 23 mg/ kg, and the inhibitory effect on gastric damage by HCl-ethanol was observed. Histological analysis exhibited mucosal erosion in the gastritis model compared to the normal group, while the ameliorating effect of the generated erosion was observed in the cabbage-treated group. The histamine concentration was significantly increased in the gastritis-induced animal model, and the histamine concentration was decreased in the 23 mg/L-treated group of cabbage extract. In conclusion, the results of this study suggest that cabbage extract not only down-regulates cytokines in vitro, but is also directly involved in histamine secretion in an animal model of gastritis; therefore, cabbage extract can help inhibit gastrointestinal disorders by improving the protective barrier.
본 연구는 케일(Brassica oleracea L. var. acephala)의 노지 재배 시, 생육 초기에 노출될 수 있는 건조 스트레스 조건에서 Glutamate (Glu)의 스트레스 경감 및 생육 회복 효과를 구명하기 위해 실시하였다. 건조 조건(D)에 노출된 케일에 5㎛ Glu 용액을 경엽처리하였다(D+Glu). Glu 처리 전 및 처리 후 4, 8일 차(0, 4, 8DAT)의 생육과 엽록소 함량을 측정하고 식물 체내의 ABA와 Glu, Pro의 함량을 분석하여 정상 환경 및 D, D+Glu 처리구 간의 비교를 실시하였다. 엽록소 함량의 경우, 8DAT에서 D+Glu 처리구는 일정한 수준을 유지한 반면에, D처리구는 14% 감소하였다. 엽면적으로 대표되는 생장 또한 D+Glu 처리구가 D 처리구에 비해 높게 나타났다. 또한 4, 8일차의 ABA 함량이 D 처리구에서 D+Glu 처리구에 비해 44.13, 49.18% 높게 나타났으며, 체내 아미노산 대사 및 건조저항성 지표인 Glu, Pro 함량은 D 처리구에 비해 D+Glu 처리구에서 보다 높은 수준을 유지하였다. 이러한 결과를 통해, Glu 처리에 의해 건조 스트레스가 경감되고 생육이 회복될 수 있다고 판단된다.
식물공장에 재배 가능한 작물은 매우 다양할 것으로 본다. 식물공장에서 재식밀도에 대한 중요성이 인식되고 있다. 본 연구는 완전제어형 식물공장에서 콜라비의 재배에 적합한 재식밀도를 구명하기 위하여 수행되었다. 식물공장 형태는 형광등을 이용한 완전제어형태로, 박막수경재배를 이용하여 재배하였다. 재식밀도는 22plants/m2(15×30cm), 27 plants/m2(15×25cm), 그리고 33 plants/m2(15×20cm)로 처리 하였다. 식물체당 지상부 생체중과 건물중 또는 식물체당 벌브의 지상부 생체중와 건물중에는 재식밀도간 유의적인 차이를 보이지 않았다. 단위면적당 지상부 생체중과 건물중 또는 단위면적당 벌브의 지상부 생체중과 건물중에는 재식밀도가 높은 처리구(33plants/m2)에서 높게 나타났다. 재식밀도와 초장, 엽면적, 광합성, 경도 및 엽록 소간에는 유의적인 차이를 보이지 않았다. 재식밀도와 벌브 높이와 지름 및 당도간에는 유의적인 차이를 보였다. 재식밀도가 낮은 22plants/m2 처리구에서 가장 높은 벌브 높이와 지름을 보였으며, 당도 또한 높았다. 이상의 결과를 바탕으로 결론을 내리면, 경제성을 고려한 생육적인 측면에서는 단위면적당 생산량이 많은 재식밀도 33plants/m2 (15×20cm)가 적정하였으나, 당도와 같은 품질적인 측면에서는 재식밀도 22plants/m2 (15×30cm)가 적정하였다.
In order to examine the functionality of broccoli sprout (Brassica oleracea, BS), solvent extracts were prepared and their anti-oxidative and immunomodulating activities were compared with those of broccoli (B). EtOH extracts (E) were potently higher than hot-water extracts (HW) in the antioxidant contents and radical scavenging activity. In particular, the total polyphenolic contents in addition to ABTS and DPPH radical scavenging activity were significantly higher in EtOH extract of broccoli sprout (BS-E; 9.15 mg GAE/g, 4.52 mg AEAC/g, and 1.14 mg AEAC/g) compared with that of broccoli (B-E; 7.83 mg GAE/g, 3.63 mg AEAC/g, and 0.97 mg/AEAC/g). Whereas, total flavonoid content was significantly higher in B-E (1.60 mg QE/g) than BS-E (1.43 mg QE/g). Anti-inflammatory activity was investigated using LPS-induced cell line model at a concentration of 10~100 μg/mL, in which all solvent extracts of both broccoli sprouts and broccoli were not toxic to RAW 264.7 cell lines. In anti-inflammatory activity of broccoli sprouts, EtOH extracts also showed significantly more potent activity than hot-water extracts in all sample concentrations tested. In addition, BS-E (100 μg/mL) inhibited nitric oxide (NO) and IL-6 production to 60.9% and 68.9% compared with the LPS inflammation group (without extracts), whereas B-E inhibited 49.6% and 54.9%. On the other hand, in immunostimulating activity by splenocytes and macrophages, hot-water extract showed significantly higher activity than EtOH extract. Especially, BS-HW stimulated the splenocyte proliferation (1.2-fold against saline group) and IFN-γ production (264.39 pg/mL) at 100 μg/mL, and the production of IL-6 (1.33-fold), IL-12 (1.09-fold) and TNF-α (1.49-fold) from macrophages was also significantly enhanced over broccoli. In conclusion, broccoli sprouts showed more potent anti-oxidative, anti-inflammatory and immunostimulating activity than broccoli, suggesting the possibility of using broccoli sprouts as functional food materials.
This study was performed in order to examine the functional and quality characteristics of raw noodles with added kale powder. The raw noodles were prepared at a concentration of 4, 8, and 12% (w/w) of the kale, based on flour weight. The antioxidant activity of kale powder, specifically DPPH free radical scavenging, reducing activity, and total phenolic content were found to be 75.16%, 1.24 (O.D.) and 44.47 mg GAE/g, respectively. Using Hunter Lab color values for raw noodle with added kale powder, L (lightness) values decreased with an increased concentration of kale powder, whereas a (redness) values decreased (p<0.001). In a texture analysis, chewiness (p<0.01), gumminess (p<0.001), and cohesiveness (p<0.001) increased significantly as the amount of added kale powder increased. According to sensory evaluations: color, flavor, taste, texture, and overall palatability had the highest score in noodle with 8% kale powder (p<0.001). During periods of storage, moisture contents were decreased when kale powder concentrations increased, but pH did not differ in any of the groups and bacteria cell counts were between 0% and 8%. Kale noodles were observed at 8% then 0%, decreased by 102 log CFU/g. The antioxidant activities of raw noodles with added kale powder were increased by 8%, a 30~50% increase over the control group. These results suggest that kale powder is useful as a functional food resource with antioxidant activities.
본연구는 콜라비 추출물의 항산화 및 항염 효능을 조사하기 위하여 수행하였다. 콜라비는 70% 에탄올을 이용하여 조추출한 후 헥산, 에틸아세테이트, 부탄올을 이용하여 용매 극성에 따라 순 차적으로 분획하였다. 항산화 활성은 총폴리페놀 함량 측정과 ABTS 라디칼 소거활성을 측정하여 평 가하였다. 에틸아세테이트 분획물이 총폴리페놀 함량(27.33±0.26 mg GAE/g)과 ABTS 라디칼 소거활 성(IC50 172.9±1.6 ㎍/mL)이 가장 높게 측정되었다. 항염 활성은 RAW 264.7 세포를 이용하였으며, 에 틸아세테이트 분획물이 가장 높은 항염 활성을 보였다. 에틸아세테이트 분획물의 LPS로 유도된 RAW 264.7 세포에서 전염증성 매개인자들에 대한 저해효과를 농도별로 측정하여 확인하였다. 콜라비 에틸 아세테이트 분획물은 NO, PGE2 생성과 iNOS와 COX-2 및 TNF-α, IL-6, IL-1β와 같은 전염증 성 사이토카인들의 단백질 발현을 농도의존적으로 저해하였다. 이러한 결과들은 콜라비가 항산화 및 항염 효능을 가지는 소재로서의 개발 가능성이 있음을 시사한다.
Broccoli, one of Brassica vegetables, has been known to possess various health beneficial activities including anti-inflammation, anti-oxidation and anti-cancer etc. Various metabolites were indicated as active compounds in broccoli. Glucosinolates such as glucoraphanin, glucobrassicin, glucoerucin and isothiocyanates, i.e. sulforaphane, which is produced through the enzymatic action of myrosinase are getting focus as their bioactivities. In this study, we treated broccoli with pulsed electric field (PEF) processing and the metabolite profiles were investigated based on the metabolomics analysis. PEF process was applied to stem and floret of broccolis with three different levels; 500, 1000, 1500 pulses with 2 kV/cm, then metabolites were extracted with 70% methanol. Metabolomic analysis was carried out with mass spectrometry through multivariate statistical analysis based on the OPLS-DA model. Significant changes of metabolite profiles were observed by PEF treatment and specific metabolites were affected as dose dependent manner. Content of major glucosinolates compounds such as glucoerucin, glucoiberin, glucoraphanin, glucobrassicin, 4-hydroxy glucobrassicin, and 4-methoxy glucobrassicin were significantly reduced, while sulforaphane was distinctively increased in PEF treated broccolis. The results implied that myrosinase released from vacuole by PEF processing transformed glucosinolate into isothiocyanate, which would be useful findings for enhancing bioactivity of broccoli by simple PEF treatment.
본 연구는 새만금 간척지에서 비닐하우스 작물 재배 가능성 검토를 위해 녹색꽃양배추를 대상으로 일일 관수량을 달리하여 관수량에 따른 토양 염농도 및 생육특성 등을 조사하여 관수량에 의한 재염화 억제효과를 알아보고자 수행하였다. 수확기의 표토의 평균 토양 EC 는 1.5 및 3.0mm·day-1 처리구에서 각각 10.9 및 11.5dS·m-1 였으며 6.0mm·day-1 처리구에서 5.1dS·m-1로 1.5 및 3.0mm·day-1 처리구보다 52~56% 낮게 나타나 점적관수 량에 따른 제염효과를 확인할 수 있었다. 화뢰의 무게는 6.0mm·day-1 처리구에서 주당 371.3g으로 1.5 및 3.0mm·day-1 처리구의 60.9g 및 129.1g보다 높은 값을 나타냈다. 50%의 수량감소를 보이는 토양 EC는 7.6dS·m-1였으며 점적관수에 의한 토양 제염효과는 6.0mm·day-1 처리에서 30~40cm 깊이까지 나타났다. 따라서 새만금간척지에서 녹색꽃양배추 재배시 점적관수에 의한 토양 재염화 억제를 위해서는 6.0mm·day-1 수준의 관수량으로 총 422mm의 물량이 필요할 것으로 예측된다. 그러나 염류의 이동은 토양 이화학적 특성 및 계절적 요인 등 여러 가지 환경요인에 영향을 받으므로 간척지 비닐하우스에서 점적관수에 따른 염류의 이동특성에 관한 추가적인 연구가 필요할 것으로 판단된다.
잎브로콜리에 심각한 피해를 주는 배추좀나방의 적절한 방제시기 설정을 위한 기초자료로 이용하기 위하여 유충접종밀도와 잎브로콜리 피 해량의 관계를 분석하여 경제적피해허용수준과 요방제 수준을 설정한 결과는 다음과 같다. 배추좀나방 3~4령 유충을 주당 0, 0.5, 1.0, 1.5, 2.0 마리를 5월 24일에 접종하고, 6월 1일, 6월 9일, 6월 18일에 발생밀도와 엽수량을 조사한 결과, 초기 접종밀도가 증가할수록 배추좀나방의 발생 밀도는 증가하였고 시간이 경과할수록 엽수량은 감소하였다. 이를 토대로 배추좀나방의 접종밀도와 수량감소율을 회귀식을 이용하여 분석한 결 과 y=1636-394x (R 2 =0.79***)로 추정되었다. 잎브로콜리에서 5% 피해율을 경제적 피해한도로 볼 때 배추좀나방의 경제적피해허용수준은 10 주당 2~3마리이고, 요방제 수준은 10주당 1-2마리 수준으로 추정된다.
해충저항성 유전자변형 작물은 대상 해충에 대한 방제 효과를 증진시킬 수 있는 효과적인 수단이 될 수 있지만, 생태계 내의 다른 비표적 생물에 부정적인 영향을 줄 가능성 또한 있기 때문에 상업적인 재배에 앞서 이에 대한 충분한 연구가 이루어져야 한다. 본 연구에서는 배추좀나방 내성 유전자변형 Bt (Cry1Ac1)양배추가 비표적 곤충인 복숭아혹진딧물에 미치는 영향을 알아보기 위한 기초적인 실험으로서 기주선택실험 및 성장실험을 실시하였다. 기주선택실험에서, 두 개체씩의 Bt 양배추와 모본 양배추에 각각 10마리의 유시 성충(alate virginoparae)을 접종하고, 사흘간 생식(reproduction) 기회를 제공한 후 생산한 약충의 마리수를 측정한 결과, Bt 양배추와 모본 양배추에 낳은 약충의 수가 각각 21.9±1.8와 22.5±2.2로 비슷하였다. 성장실험에서, 갓 태어난 무시 약충(apterous virginoparae)을 Bt 양배추 및 모본 양배추를 기주로 하여 키웠을 때, 성충이 될 때까지 걸린 시간은 Bt 양배추와 모본 양배추에서 각각 5.8±0.2, 5.9±0.1 일(day)로 통계적으로 유의한 차이가 없었고, 개체군 생장률(rm) 또한 0.7±0.1, 0.8±0.1로 유사하였다. 이상의 결과들로 볼때 Bt 양배추가 복숭아혹진딧물에 미치는 영향이 없거나 미미함을 알 수 있으나, 다양한 관점에서의 보완 연구가 더 필요하다.
This study was conducted to develop economic injury level (EIL) and economic threshold (ET) of Cabbage armyworm, Mamestra brassicae L. on cabbage (Brassica oleracea L. var). The changes of cabbage biomass and M. brassicae density were investigated after introduction of larval M. brassicae (2nd instar) at different densities: 0, 1, 2, 4, 8, and 16 larvae per plant at 40 d after planting for an open field experiment, and 0, 2, 5, 8 and 12 larvae per plant at 25 d after planting for a glass house experiment. In the field experiment, the yield loss of cabbage was not significantly different among treated-plots at 30 d after the larval introduction, showing an over-compensatory response of cabbage plants to M. brassicae attack. In the glasshouse experiment, however, the biomass of cabbage at 15 d after the larval introduction significantly decreased with increasing the initial introduced number of M. brassicae, resulting in 38.3, 36.7, 21.7, 23.3 and 16.7g in above treated-plots, respectively. The relationship between cumulative insect days (CID) and yield loss (%) of cabbage was well described by a nonlinear logistic equation. Using the estimated equation, EIL of M. brassicae on cabbage was estimated at 44 CID per plant based on the yield loss 14%, which take into account of an empirical gain threshold 5% and marketable rate 91% of cabbage. Also, ET was calculated at 80% of the EIL: 35 CID per plant. Until a more elaborate EIL-model is developed, the present result may be useful for M. brassicae management at early growth stage of cabbage.
This study was conducted to develop the economic threshold for the diamondback moth (Plutella xylostella (L.) on Leaf Broccoli (leaf vegetable) in 2007. To investigate the relationship between initial density of diamondback moth larvae and broccoli leaf yield, experimental plots with five treatments (0, 0.5, 1.0, 2.0, 4.0 larvae per plant) as initial density were established. We inoculated larvae in chesses cloth covering to survey larvae density change. When grown to eat, leaves of broccoli were harvested by periods. 60-70% of larvae were removed whenever we picked the leaves. High levels of larvae were associated with significant reductions in leaf yield. There were 85%, 64%, 58%, and 56% yield reductions from the diamond back moth larvae density in 0, 0.5, 1,0, 1.5, 2.0 per plant, respectively 25days after larvae inoculation. The regression equation used to predict leaf yield based on the number of initial larvae density per a plant was y=1635-393x(R2=0.79***). The economic injury level of diamondback moth on leaf broccoli was 2-3 larvae per 10 plants for a damage level of 5%. The economic thresholds was 1-2 larvae per 10 plants. Thus, the diamond back moth management should be initiated 1-2 larvae occurrence per plant.
Alcoholic fatty liver disorder has become a frequent health concern worldwide. To investigate the effects of Brassica oleracea (B. oleracea) sprout extract (BOE), the present study was designed with alcoholic fatty liver in the rat. Initially, the effects of BOE on liver parameters were examined. Male rats were divided into five groups. The normal control group was fed the normal diet, and the BOE group was fed the high fat diet and ethanol with/without BOE for 4 weeks. After 4 weeks feeding period, rats were sacrificed and their livers and blood were used for fatty liver-related biomarkers analyses. As a result, BOE ameliorated fatty liver-related enzymes profiles in liver tissues and also reduced blood alcohol concentration in rat model. We demonstrated that BOE protected the high fat diet and alcohol-induced fatty liver in rat model. Furthermore, BOE increased detoxificative abilities against alcohol.
Analyzing the 13 sort contents of broccoli leaves by using GC/MS, sulforaphane was found in 11 sort of broccoli leaves for the result. After being grinded by the blinder, amount of sulforaphane in broccoli leaves was rapidly raised after thirty minutes and maintained the amount till sixty minutes have passed. Among the parts of broccoli, the root had the most sulforaphane. In freezing temperature, biosythesized sulforaphane maintained longer than in room temperature. However, even in frozen condition, the amount of sulforaphane was reduced to half or less after 3 weeks.
Cabbage head splitting can greatly affect both the quality and commercial value of cabbage (Brassica oleracea). To detect the genetic basis of head-splitting resistance, a genetic map was constructed using an F2 population derived by crossing “748” (head-splitting-resistant inbred line) and “747” (head-splitting-susceptible inbred line). The map spans 830.9cM and comprises 270 markers distributed in nine linkage groups, which correspond to the nine chromosomes of B. oleracea. The average distance between adjacent markers was 3.6cM. A total of six quantitative trait loci (QTLs) conferring resistance to head splittingwere detected in chromosome 2, 4, and 6. Two QTLs, SPL-2-1 and SPL-4-1, on chromosomes 2 and 4, respectively, were detected in the experiments over 2 years, suggesting that these two potential loci were important for governing the head-splitting resistance trait. Markers BRPGM0676 and BRMS137, which were tightly linked with head-splitting resistance, were detected in the conserved QTL SPL-2-1 region using bulked segregant analysis. Synteny analysis showed that SPL-2-1 was anchored to a 3.18Mb genomic region of the B. oleracea genome, homologous to crucifer ancestral karyotype E block in chromosome 1 of Arabidopsis thaliana. Moreover, using a field emission scanning electron microscope, significant differences were observed between the two parental lines in terms of cell structures. Line “747” had thinner cell wall, lower cell density, larger cell size, and anomalous cell wall structure compared with the resistant line “748”. The different cell structures can provide a cytological base for assessing cabbage head splitting.
