World Health Organization(WHO) revised the classification of neoplasms and other tumours related to odontogenic apparatus in 1992. The aim of this study was to classify the odontogenic tumors of Korean according to the WHO Histologic classification. A total of 271 cases were reviewed for the study which were diagnosed as odontogenic tumors at the department of Oral Pathology, Yonsei University College of Dentistry for the period from Jan. 1997 to March 2003. Clinical and pathology reports were reviewed & radiographic feature were examined. The following results were obtained : 1. Among 271 cases, 269 cases(99.3%) were diagnosed as benign odontogenic tumors, and the remaining 2 cases(0.7%) were malignant tumors, which were diagnosed as odontogenic ghost cell carcinoma and squamous cell carcinoma ex odontogenic cyst. 2. Four cases were not able to classify into the WHO classification. All of them were belonged to mixed odontogenic tumors; two cases of adenomatoid odontogenic tumor with calcifying epithelial odontogenic tumor, one case of adenomatoid odontogenic tumor with odontoma, odotogenic cyst and one case of ameloblastoma with immature odontoma. 3. The most frequent odontogenic tumour was odontoma(45.2%), followed by ameloblastoma (29.2%), odontogenic fibroma(9.2%) 4. One case of atypical amelobalstoma and one case of calcifying odontogenic cyst with ameloblastic fibroma were not able to subclassify histologically. 5. Male to female ratio of odontogenic tumors was 1.2:!. Odontogenic tumors mainly occured in the first and second decade, occurred twice as much as in the mandible than in the maxilla 6. The odontogenic tumors was discovered by routine oral x-ray examination, whereas the chief complaint of ameloblastoma were swelling, pain. 7. Ameloblastoma, adenomatoid odontogenic tumor, calcifying odontogenic cyst and odontoma were related to the impacted teeth and tooth displacement. The root resorption was frequently observed in ameloblastoma and calcifying odontogenic cyst.

Osteoarthrosis in TMJ is known to accompany characteristic features of the craniofacial malformation and malocclusion. In order to explain the mechanism that brings about the typical mandibular changes relating to the frequent clinical symptom, i.e., the limited mouth opening in TMJ patients. The present study investigated the mandibular morphology of the rabbit, also intentional TMJ osteoarthrosis was performed on the TMJ of the rabbits and their morphological changes were observed. A total of 12 New Zealand house rabbits weighing an average of 3 kgwere used and all left side joints were surgically meniscectomized and right side joints were used as controls. The rabbits were sacrificed in one week, two weeks, one month, three months and 5 months after the menisectomy, and the removed TMJs were examined by histological and radiographical methods and analyzed statistically. It was found that the mandibular morphology caused by osteoarthritis in the TMJ was characterized by the decrease in size of the ascending ramus, posterior ramus bowing, deep antegonial notch and thickening of the cortex in inferior and posterior borders of the mandible, that the similar features were also observed in the rabbit mandible. When the menisectomy was performed in the unilateral side of rabbit TMJ, the mandibular changes including the increased depth of antegonial notch and posterior ramus bowing occurred only on the affected side. These data suggested that the limitation of the mandibular movement and altered masticatory pattern by meniscectomy affects the structural remodeling of the mandible and TMJ likely the TMJ osteoarthrosis

The temporomandibular joint (TMJ) showed characteristic anatomy and functions contrast to the other joints. In particular, children or young adolescent with the progressive temporomandibular disorder (TMD) showed remarkable changes in their mandibular growth and occlusion. The purpose of this study was to elucidate the functional and biomechanical causes for the maxillofacial deformities of TMD. A typocranial experiment was performed using a plastic skull model by simulating the mandibular movement after condylectomy. The directional forces of masticatory and suprahyoid muscles were reproduced on the typocranial skull model by elastics connecting between mandible and adjacent bony structures. Resultantly, the mandible of unilaterally or bilaterally condylectomized mandible was rotated downward and backward by the activation of masticatory and suprahyoid muscles. When unilaterally or bilaterally condylectomized mandible was activated by masticatory, suprahyoid and perioral muscles, the mandible was less rotated downward and backward than the cases activated by masticatory and suprahyoid muscles only. In the typocranial experiment after unilateral condylectomy the mandible was rapidly deviated toward the affected side, but this deviation was much reduced when the mandible was intensely supported by perioral muscles. By the compensatory response against the overload of TMJ due to the mandible body resection the typocranial experiment also produced the deepening of antegonial notch by the increased force of mylohyoid and perioral muscles. These findings suggest that after condylectomy the posterior fibers of mylohyoid and perioral muscles become hypertrophic and induce the characteristic mandibular deformities, i.e., deepening of antegonial notch and bowing of posterior ramus, which are similar to the mandibular changes in the progressive TMD.

Epithelial-mesenchymal interaction is well known to have an importance during the organ development as well as cell growth and differentiation. However, in vitro experimental model is not well developed to reproduce in vivo cellular micro-environment which provide a epithelial-mesenchymal interaction. The aims of this study were to develop and evaluate the in vitro experimental model that maintains epithelial-mesenchymal interaction by organotypic raft culture, and to characterize biologic properties of three-dimensionally reconstituted human normal oral kertinocyte(NHOK) and immortalized human oral keratinocytes(IHOK) by histological and immunohistochemical analysis. The results were as follows; 1. Best condition of three dimensionally reconstituted IHOK & HaCaT cells are 14 days air-exposure cultivation, 3 days of submerged state, and dermal equivalent consisting type I collagen and IGF cells. 2. In comparison to IHOK, there was better preservation of the overall epidermal strucutures in oral cracioma cells (HN30) & HaCaT cells by organotypic cultures. But orgnaotypic co-culture of the normal keratinocyte showed the thinnest epithelial layer formation. 3. PCNA was detected primarily in the basal layer of normal mucosa and NHOK, whereas was shown throught the epithellium except surface layer of IHOK cells, and it's expression was similar to that of CIS of biopsied patient's tissue 4. Involucrin is expressed in the upper layer of oral mucosa and NHOK raft, but staining for involucrin was induced in the IHOK rafts indicating differentiation is incomplete, and the staining pattern in the IHOK raft was not uniform. 5. Normal oral keratinocyte raft showed weak immunostaining for p53, and p53 expression of IHOK raft increased rahter than in NHOK. In organotypic cultures of normal cells and IHOK, p53 expression was restricted to the proliferative part of epithelium. This is consistent with expression pattern in biopsy specimens of the normal and CIS tissue. 6. In artifically reconstructed NHOK, the pattern of keratin staining showed both similarity and differences from that of intact normal mucosa. An obvious difference was increased expression of CK10 & CK19, and decreased expression of CK6 in a reconstructed NHOK rather than in normal mucosa, and similar expression was in CK4 and CK16. 7. CK19 & CK16 were strongly positive in HPV immortlaized keratinocyte rafts rahter than in NHOK, an indicator of premalignant or malignant changes, while CK10 & CK6 were decreased, and organotypic cultures of IHOK express was similar keratin expression as epithelial dysplasia or CIS tissue. These results suggest that three-dimensional organotypic co-culture of normal oral & immortalized keratinocytes with dermal equivalent consisting type I collagen and fibroblasts results in similar morphologic and immunohistochemical characteristics to in vivo patient specimens. Thus this organotypic system can be used for studing mechanism of epitehlial-mesenchymal interaction in particular regulating epidermal diffenentiation and morphogenes

Insulin-like growth factor 2 (Igf-2) and H19 genes are closely linked imprinted genes which have a pivotal role in embryogenesis and fetal development. Igf-2 and H19 are coexpressed in tissues of mesodermal, endodermal and neuroectodermal origin. Rat Igf-2 has a complex structure with three promoters and a complicate imprinting mechanism having an exception of biallelic expression in the choroid plexus, leptomeninges, and fetal tissues of neuroectodermal origin. To detect the expression of maternal and paternal alleles of Igf-2 and H19 during orofacial development, fetal and neonatal hybrid rats, obtained from Wistar and Fisher interstrain rat crosses were used. We also detected the promoter-specificity of Igf-2 transcripts by primers selected from P1, P2, and P3 of Igf-2 gene. RT-PCR analysis of Igf-2 and H19 showed the monoallelic expression of Igf-2 from the paternal allele and H19 from the maternal allele in E15.5 to E19.5 orofaciall structures including the maxilla, tongue, and salivary gland. P3 promoters were active in all tested samples, whereas transcripts derived from P2 promoter arised with approximately half of the tested cases and showed variable alternation. P1 promoter was not transcribed in all tested samples. These results suggest that Igf-2 and H19 may be involved in orofacial development and exhibit parent-of-origin monoallelic expression. On the other hand, in orofaciall development, P2 and P3 promoters except for P1 promoter are transcribed with variable alternative transcripts

The purpose of this study was to research the osseointegration in the interface of dental gold alloy. Some investigator presented that gold was biologically safe but occurred fibrous integration on implant interface. We used 4 different gold composition implant to investigate the osseointegration on implant interface. Osseointegration occurred on the all implant interface. We thought that osseointegration depended not only on gold composition but also on precious metal composition such as platinum or silver. If we use precious metal implant, it might reduce the Galvanic current and chemical corrosion which were occurred between titanium implant and percious metal suprastructure. We recommend that precious metal implant was superior to non-precious metal implant without financial problem.
