In odontogenic osteolytic lesions, the mechanism involved in inflammation 때d osteolysis is still under debate. We investigated the role 。OL-l ~ -1 ß, -4, -6, -8, TNF- a in the expansion of the odontogenic cysts, by using 50 cases of excised odontogenic cysts. The degrees of inflammation were graded into 2 groups and immunohistochernical stainings were performed, The cytoplasrnic reaction in squamous epithelial cells, stromal cells and infIitrating inflammatory cells was examined. The relationship between the expressions of IL-1 q -1 ß, -4, -6, -8, π-JF- aand the degree of inflammation and the correlation among them were analyzed. 까1e 비Stilαγtic expressions of IL-l ~ IL-l ß and TNF-awere 66.6%, 66.6% and 4O.00Al respeαively and the epithelial exp~않sions of IL-4, -6, -8 were 32.00Al, 38.00Al and 24.00Al respeαively. IL-4, -6 and 용 were positively stained in plasma cells in 38.00Al, endothelial cells in 40.00/0 and neutrophils in 24. 00Al respectively. The expresssion rate of IL-4 in plasma cells and IL-8 in epithelium and neutrophils were inα얹sed in ca않s with marked inflammation. 까1e exp!1않sion rate of IL-1 ßin 피해ocytes and IL-4 in plasma cells were positively correlated with the expæssion of TNF-1 ain histiocytes. π1e expression rate of anti-inflammatory cytokine IL-4 in the epithelium were correlated with the expression of IL-6 in the epithelium and endothelial cells. A1though statistically insi.맑표ìcant, the expression rate of IL-6 were decreased in cases with marked inflammation and nega디vely correlated with IL-8, the proinflammatory cytokine, and the expressions of IL-4 and IL-6 in the epithelium can be considered as inhibiting the inflammatory reaction. These results suggest that the expressions of IL-4 and IL-6 suppress and IL-8 stimulates the inflammatory reaction in Odontogenic Cysts.
Amino acid transporters play an important role in supplying organic nutrient to cells. The expression of L-type arnino acid transporter 1 (LATl) and its subunit 4F2 heavy chain (4F2hc) was evaluated to deterrnine the alterations to these transporters in oral norrnal mucosa (ONM) , oral precancerous lesion (OPL) and oral squamous cell carcinoma (OSCC). Sections from formalin-ftxed, paraffm-embedded S따nples of ONM, OPL or OSCC were exarnined using immunohistochernical staining to detect LATl and 4F2hc proteins. 까le LATl and 4F강lC expression increased progressively from ONM to hypeφ,Iastic and to dysplastic lesions and OSCC. In partiαlar, LATl rnay be a more S야dftc indicator of tumor prog~않sion than 4F2hc. 까le gradually increasing LA Tl and 4F2hc expression detected during the multistep progressive change shows that the protein rnay have an important role in the early stages of multistep oral carcinogenesis. In addition, the specific inhibition of LA Tl and 4F2hc rnight be a new rationale to suppress oral cancer progression.
Periodontalligament (PDL) fibroblasts have an ectomesenchymal origin and are known to participate not only in formation of PDL but also in the repair and regeneration of the a이acent alveolar bone and cementum. However, little is known about the molecular mechanism which is related to the development and differentiation of PDL cells. Recendy, we reported the PDLs (a periodontalligament-specific) 22 as a PDL fibroblast-specific mRNA which is not expressed in gingival fibroblasts. In this study, to examine the expression and functional characterization of PDμ22 mRNA and prαein in development and differentiation of periodontal 따sue , we carried out northem analysis, insitu hybridization, immunofluorescence and immunohistochemistry. The expression of PDLs22 mRNA was increased with PDL cell differentiation from the confluent to multilayer stage but decreased slighdy with mineralized nodule formation in vitro. πle PDLs22 protein was localized on the nuclear membrane and expressed throughout the differentiation of PDL fibroblasts in vitro. The PDLs22 mRNA and protein were expressed in the differentiating cementoblasts, PDL fibroblasts and osteoblasts along the r∞t surface and alveolar bone of the developing rat teeth. These results indicate that the PDLs22 plays an irnportant role in the differentiation of cementoblasts and osteoblasts and thus homeostasis of cementum, PDL and alveolar bone.
To deterrnine the effects of insulin-like growth factor system in the aging process of the rat submandibular gland, expression pattems of 19f-1, 19f-2 and 19f-1 receptor were investigated by immunohistochem엽따. 1n addition, age-related changes of 19f-1 and 19f-2 concentrations in serum were deterrnined by radioimmunoassay 19f-1, 19f-2 and 19f-1 receptor were principally expressed 띠 the duct cells. 19f-1 receptors were strongly expre잃ed in 2-week-old group but were gradually decreased with advancing age. 19f-1 expressions were the strongest in the excretory and striated ducts of 7-month-old and 17-month-old groups. 19f-1 expressions, however, were weakiy expressed in the cells of granular convoluted tubules of 7-month잉ld , 17-month-old and 27-month-old groups. 19f-2 expressions were found in some cells around duαs of 2-week-old group but were persisted relatively weak. The concentrations of 19f-1 and 1향2 in serum were the highest in 2-week-old group but were decreased and maintained at low level with advancing age. These results show that all components of 19f system were expressed in the rat submandibular giand in an age-related manner. Serum concentrations of all components of 19f system were not dosely related with the intra-밍and비ar expresslon patterns 까lerefore , 1양 systerns may play important roles in the aging process of the rat submandibular gland in an autocrine/paracrine manner
Oral cavity offers a good environment for the bacterial growth. 까le species diversity of oral bacteria has been studied for many years based on the classification of liable bacteria. In order to acquire more comprehensive insight into the baα.erial community of human oral cavity, we used molecular ecological methods to clone 16S rDNAs from human saliva. DNA was direαly extracted from saliva collection of human according to age. By 27F, 1492R primer for 16s rDNA’s amplification were enforced. Cloning was achieved from the amplified DNA using pGEM-T Easy Veαor and competent cellJM 109. 159 different base sequences were obtained from saliva samples of four different persons, chosen according to the age group. In all samples, Streptococcus sp. were the most abundant rnicrobes, followed by Prevotella sp., except in the saliva of an old person where Rothia sp. presented the second dominant group 까lis saliva sample showed another and more irnportant characteristics that there were increased species diversity, from pathogenic bacteria like Haemophilus sp. and lautropia sp. to the higher pr'φ。rtion of unculturable bacteria. The same tendency, but less clearly, was found for the saliva of adult smoker in the forties. These results suggest that environmental factors in the oral caviψ of smoker and older person makes favourable condition for the infectious bacteria.
Florid cemento-osseous dyspl잃ia with concommitant traumatic bone cyst is uncommon condition. We report a case of florid cemento 。,sseous dysplasia involving four quadrants of the jaws and associated with large cystic lesion of the mandible misdiagnosed as a multiple odontogenic keratocysts in a 37-year-old Korean woman. Histological examination revealed unencapsulated fibroblastic proliferation with formation of bone and cementum, showing different developmental stages, and cys디c lesions resembling a traumatic bone cyst.
